    Notes: This paper reports an automatic instrument developed for characterization of optical waveguides. A specially designed waveguide excitation unit offers a flexible platform for measurement of optical waveguide characteristics like number of propagating modes, mode field intensity distribution, the refractive index profiles, waveguide geometry, attenuation, and dispersion. Further, the availability of attachments like filters, polarizers, chopper, intensity profiler, and CCD camera make the instrument flexible enough for a variety of other experiments also. The instrument is PC controlled and assisted by user friendly menu-driven software. A large number of waveguides have been analyzed using the setup. These results are also presented in the paper. © 1995 American Institute of Physics.

    Notes: This paper presents a Digital Signal Processor (DSP) based implementation of Hybrid of Fuzzy Logic Controller (FLC) and Proportional-Integral (PI) speed controller for Direct Torque Controlled (DTC) Permanent Magnet Synchronous Motor (PMSM) drive. The fuzzy membership function is used for hybrid control of these two FLC and PI speed controllers in such a way that at the time of dynamic conditions such as starting, the degree of belonging for FLC speed controller is higher than the PI speed controller and near set point the degree of belonging of PI controller is having higher weightage. The simulation model of the drive system is developed in MATLAB environment with simulink, PSB and FLC toolboxes to analyze the performance of the PMSM drive system. This hybrid speed controller is found suitable for DTC based PMSM drive to maintain the high level of performance while maintaining the excellent response at the time of starting, speed reversal, load perturbation and steady-state operating condition of the drive.

    Notes: A linear stability analysis of the viscous fingering of miscible non-Newtonian flow displacements in a rectilinear Hele-Shaw cell is presented. The shear-thinning character of the non-Newtonian fluid is described using the Carreau model which involves two rheological parameters De and n. Flows where either the displacing or displaced phase has a shear-thinning behavior are examined and compared with those of Newtonian flows. It is found that the shear-thinning character of the non-Newtonian fluid has an important effect on the flow instability. In particular, a flow where the driving fluid is shear-thinning is always more unstable than its Newtonian counterpart. For this flow, the maximum growth rate and the spectrum of unstable wave numbers are larger than in the Newtonian case which suggests that more ramified structures will develop as the finger instability grows. On the other hand, when the displaced fluid is non-Newtonian, a stronger shear-thinning rheological behavior leads in general to a less unstable flow. The mechanisms responsible for the changes in the flow instability are explained in terms of the different sources contributing to the generation of the vorticity disturbance. © 2002 American Institute of Physics.

    Notes: A. virus causing mosaic and leaf deformation of Physalis minima has been identified as an isolate of cucumber mosaic virus (CMV) on the basis of its transmission by aphids in a non-persistent manner, polyhedral particles of 29 nm diameter, molecular weight of coat protein subunits us 24-5 kDa. serological relationship with a CMV isolate and a tripartite single-stranded RNA genome with a subgenomic RNA4- Furthermore. cDNA representing coat protein gene was synthesized and cloned. Complete nucleotide sequences (890 nt) were obtained which showed a coat protein gene open reading frame of 657 residues. THE nucleotide sequences provided the 218 amino ACID sequences of the coat protein. Nucleotide as well as amino acid sequences revealed more than 90% identity with the CMV subgroup I strains.

    Notes: The transformations of phyllosilicates and the changes in potassium (K) balances caused by long-term cropping have been examined in soil from the 153-year-old Broadbalk experiment. Samples taken between 1856 and 1987 and kept in archive were fractionated for particle size and analysed for changes in mineralogy by X-ray diffraction and changes in total K of the coarser and fine fractions. These were compared with K balances made from records of cropping and fertilizer application. No change in mica due to cropping and K-depletion could be detected. Deep ploughing after 1925 mixed chlorite from the subsoil into the surface soil, but this had weathered by 1987. Potassium balances did not agree with measured changes in total K presumably because of unquantified uptake of K from the subsoil and poorly quantified K leaching.

    Notes: The numerical evaluation of certain integral transforms is required for the interpretation of some geophysical exploration data. Digital linear filter operators are widely used for carrying out such numerical integration.It is known that the method of Wiener–Hopf minimization of the error can be used to design very efficient, short digital linear filter operators for this purpose. We have found that, with appropriate modifications, this method can also be used to design longer filters. Two filters for the Hankel J0 transform (61-point and 120-point operators), and two for the Hankel J1 transform (47-point and 140-point operators) have been designed. For these transforms, the new filters give much lower errors compared to all other known filters of comparable, or somewhat longer, size. The new filter operators and some results of comparative performance tests with known integral transforms are presented.These filters would find widespread application in many numerical evaluation problems in geophysics.

    Notes: Background Allergen extracts are unstable, heat labile or susceptible to proteases. Stability of allergen extracts is important for proper diagnosis and therapy of allergic disorders.Objective The present study was undertaken to determine the preservation and stabilization conditions of Imperata cylindrica (Ic) grass pollen extract.Methods The Ic extract was kept with 0.1 mε-aminocaproic acid (EACA), 0.75 m sucrose, 5% glycerol, 0.03% human serum albumin (HSA) or 0.4% phenol for different time periods. The extracts were stored for 3, 6 and 12 months each at 4 °C, 4 °C with daily exposure to room temperature (RT) for 1 h, and RT. The quality of extracts was analysed by SDS-PAGE, Western blot, ELISA, ELISA inhibition and skin test.Results Extracts kept with EACA and sucrose retained most of the protein bands followed by glycerol as determined by SDS-PAGE and Western blot during all storage periods and conditions in comparison with standard extracts. The extracts kept with HSA, phenol and without preservative (WP) showed protein degradation below 33 kDa after 3 months storage at all conditions. However, a 67-kDa allergen was stable in these extracts. EACA extract required 75 to 120 ng of protein for 50% inhibition in IgE binding under different conditions, whereas standard extract required 70 ng for the same. ELISA also demonstrated high allergenic reactivity of EACA extract. ID test on allergy patients with EACA extract demonstrated same allergenic potency as that of standard extract.Conclusion EACA is the best preservative/stabilizing agent of Ic pollen extract, followed by sucrose and glycerol. Ic extract kept with phenol, HSA and without preservative showed degradation within 3 months. EACA preserved extract is equally potent as that of standard extract up to 1 year's storage.

    Notes: Background Fusarium solani (FS) is an important allergen source afflicting 4% of the nasobronchial allergy patients. Fus s I3596*, a 65 kDa major glycoprotein allergen of FS reacts with 95% fungus sensitive patients.Objectives To purify and characterize a potent peptide from Fus s I3596* which may be useful for therapeutic purposes.Methods The 65 kDa protein was sequentially cleaved with trypsin and cyanogen bromide (CNBr). The cleaved products were purified on reverse phase high performance liquid chromatography (rpHPLC) column and functionally characterized by in vitro and in vivo methods for its IgE binding and histamine release.Results The protein on cleavage showed 11 peaks (I to XI). Of these, peaks I, III, IV and V were highly allergenic as determined by IgE ELISA. These peaks were further purified and peptide IV-1 was most potent in comparison to other peptides by ELISA-inhibition. This peptide showed IgE binding but could not evoke intradermal response in Fusarium-sensitive patients. Heparinized blood challenged with peptide IV-1 does not release histamine. Preincubation of heparinized blood with peptide IV-1 and challenging with crude extract blocked histamine release in a dose dependent manner.Conclusion Peptide IV-1 binds to IgE but does not release histamine, demonstrating its potential use in therapy of Fusarium-allergic patients.
