    Notes: Background:  Prominent infiltration of eosinophils into the airway mucosa and release of inflammatory mediators upon their adhesion onto airway epithelial cells are the immunopathogical mechanisms of allergic asthma.Objective:  We investigated the effect of normal and paraformaldyhyde-fixed human eosinophils on BEAS-2B cells, a human bronchial epithelial cell line, for the release of inflammatory cytokine interleukin (IL)-6.Methods:  Interleukin-6 in cell culture supernatant, protein amount of p38 mitogen-activated protein kinase (MAPK), and nuclear factor-κB (NF-κB) activity in BEAS-2B cells were analyzed by Western blot and enzyme-linked immunosorbent assay (ELISA). IL-6 gene expression was assessed by reverse transcriptase-polymerase chain reaction (RT-PCR), and p38 MAPK activity and inhibitor (I)κB-α induction were evaluated by Western blot.Results:  Co-culture of BEAS-2B cells and eosinophils induced a significant elevation of IL-6 expression in BEAS-2B cells. Interaction of eosinophils and BEAS-2B cells led to a marked induction in phospho-p38 MAPK, phospho-IκB-α and activity of NF-κB in BEAS-2B cells. NF-κB inhibitor BAY 11-7082 and p38 MAPK inhibitor SB 203580 significantly decreased IL-6 release in a co-culture of BEAS-2B cells and eosinophils. Fixed human eosinophils were able to maintain their ability to induce IL-6 release in co-culture, activate p38 MAPK and NK-κB, and up-regulate IL-6 gene expression in BEAS-2B cells.Conclusions:  These data indicate that the interaction of eosinophils and bronchial epithelial cells plays an important role in airway inflammation, at least partly, via IL-6 induction.

    Notes: ‘Ketan Nangka’, the donor of the wide compatibility gene (WCG) showed typical hybrid sterility when crossed to a landrace, ‘Bai Mi Fen’, of Yunnan province in China. A genome-wide analysis was performed for a backcrossed population of ‘Ketan Nangka’/‘Bai Mi Fen’//‘Ketan Nangka’ using a total of 143 simple sequence repeat markers and an expressed sequence tagged marker to cover the entire rice linkage map. As a result, two independent loci were found to cause hybrid sterility via female gamete abortion. The locus on chromosome 4 may correspond to S9, but the other, on chromosome 2, was different from all the previously reported hybrid sterility loci and was designated as S29(t) following the hybrid sterility nomenclature. On the basis of allelic interaction which causes female gamete abortion, two alleles were found: S29kn(t) in ‘Ketan Nangka’ and S29bi(t) in ‘Bai Mi Fen’. In the heterozygote, S29kn(t)/S29bi(t), which was semi-sterile, female gametes carrying S29bi(t) were aborted. An Aus variety from the Indian subcontinent, ‘Dular’, was found to have a neutral allele, S29n(t). Two molecular markers, RM185 and RM425, linked to S9 and S29(t), respectively, will be useful for marker-aided transfer of WCGs in hybrid rice breeding.

    Notes: The brown planthopper (BPH) is one of the most destructive insect pests of rice. Resistant varieties have proved to be one of the most economic and effective measures for BPH management. In this study, an indica rice ‘DV85’ showed resistance to biotype 2 of BPH by bulked seedling test, and a recombinant inbred line (RIL) population derived from a cross between a susceptible rice ‘Kinmaze’ and ‘DV85’ was phenotyped to map genetic factors conferring BPH resistance in ‘DV85′. Composite interval mapping revealed that one quantitative trait locus (QTL) with a LOD score of 10.1 was detected between XNpb202 and C1172 on chromosome 11. This QTL was designated as Qbph11. Qbph11 explained 68.4% of the phenotypic variance of BPH resistance in this population. The allele from the resistant parent ‘DV85’ at Qbph11 reduced the damage caused by BPH feeding and would be very useful in breeding resistant rice varieties via marker-assisted selection.

    Notes: We conducted a double-blind, randomised, placebo-controlled study evaluating the efficacy of prophylactic metaraminol for preventing propofol-induced hypotension. Thirty patients aged 55–75 years undergoing general anaesthesia were randomly allocated to receive either metaraminol 0.5 mg or saline before administration of fentanyl 1 µg.kg−1 and propofol 2 mg.kg−1. Induction of anaesthesia was associated with a decrease in mean and systolic arterial pressure in both groups (p = 0.0001). However, there was no significant difference between the two groups. These results show that prophylactic use of metaraminol 0.5 mg does not prevent the decrease in blood pressure following fentanyl and propofol induction in older patients.

    Notes: In Study A, the incidence of arterial oxygen desaturation was studied using pulse oximetry (Sao2) in 100 sedated and 100 nonsedated patients breathing room air who underwent diagnostic upper gastrointestinal endoscopy. Hypoxia (Sao2 92% or less of at least 15 s duration) occurred in 17% and 6% of sedated patients and nonsedated patients, respectively (p 〈 0.03). Mild desaturation (Sao2 94% or less and less than 15 s duration) occurred in 47% of sedated patients compared with 12% of nonsedated patients (p 〈 0.001). In Study B, the effects of supplementary oxygen therapy and the effects of different pre-oxygenation times on arterial oxygen saturation (Sao2) in sedated patients were studied using pulse oximetry. One hundred and twenty patients who underwent diagnostic upper gastrointestinal endoscopy with intravenous sedation were studied. Patients were randomly allocated to one of four groups: Group A (n = 30) received no supplementary oxygen while Groups B–D received supplementary oxygen at 4 l.min−1 via nasal cannulae. The pre-oxygenation time in Group B (n = 30) was zero minutes, Group C (n = 30) was 2 min and Group D (n = 30) was 5 min before sedation and introduction of the endoscope. Hypoxia occurred in seven of the 30 patients in Group A and none in groups B, C and D (p 〈 0.001). We conclude that desaturation and hypoxia is common in patients undergoing upper gastrointestinal endoscopy with and without sedation. Sedation significantly increases the incidence of desaturation and hypoxia. Supplementary nasal oxygen at 4 l.min−1 in sedated patients abolishes desaturation and hypoxia. Pre-oxygenation confers no additional benefit.

    Notes: Rheological properties of cooked noodles made on a small-scale laboratory sheeting machine developed at the Cereal Research Centre were strongly correlated with the same properties of noodles made on a pilot noodle sheeting machine. Values obtained from both machines were evaluated against 22 laboratory tests used in our breeding program. For example, cooked noodle viscoelasticity (r=0.89, P 〈 0.01) and cooked noodle cutting force (r = 0.79, P 〈 0.01) were similar. Cooked noodle viscoelasticity of fresh noodles made on the laboratory machine correlated significantly (P 〈 0.05) with 17 of the tests and cooked noodle cutting force correlated significantly with 18 tests. Cooked noodle viscoelasticity of dried noodles made with the pilot plant noodle machine had significant correlations (P 〈 0.05) with only 14 of the tests and cooked noodle cutting force had significant correlations with 15 tests. The pilot plant machine requires a minimum of 500 g for one sample, while the small-scale sheeting machine requires only 5-10 g of flour.

    Notes: The dispersion of the first molecular hyperpolarizability β of strongly charge-transfer chromophores is investigated. The investigation extends the well-known Oudar–Chemla nonresonant two-level model into the resonant regime. An equation for β that includes the effect of dephasing and vibrational structure of electronic states is derived. It is shown that if only the dephasing mechanism is included in the two-level model, the intrinsic hyperpolarizability β0 calculated using the Oudar–Chemla equation from the hyper-Rayleigh scattering (HRS) experimental data will decrease with decreasing the excitation wavelength. The trend is reversed when the vibrational structure is also incorporated. It is concluded that in order to obtain a consistent β0 in the HRS experiment using an excitation wavelength in the resonance regime, it is inadequate to include only the dephasing mechanism. The vibronic structure also plays an important role and needs to be considered in the data analysis in order to obtain a β0 to agree with the off-resonance experimental result. © 2000 American Institute of Physics.

    Notes: In this article we present a simple, nonoptical shear force detection scheme to control probe–sample distance for a scanning near-field optical microscope (SNOM). Shear force detection is realized by attaching a tapered optical fiber probe to a piezoelectric bimorph cantilever in which one piezo layer generates a maximum piezo voltage when the cantilever is excited at resonance by the other piezo layer. The amplitude of the piezo voltage will decrease as the probe approaches a sample's surface due to probe–sample interacting shear force. Keeping the piezo voltage constant provides a very sensitive method by which to control probe–sample distance. Based on the shear force detection scheme, a shear force SNOM system has been built, operating in transmission collection mode. Shear force topographic and optical images have been taken using uncoated optical fiber probes fabricated by a chemical etching technique. The results suggest that the system is very reliable, repeatable, and easy to use. © 2001 American Institute of Physics.
