    Notes: T-cell sensitization to indirectly presented alloantigens (indirect pathway of allorecognition) plays a critical role in chronic rejection. The usual very efficient priming of such self-restricted, T helper type 1 (Th1)-deviated CD4+ T cells obviously conflicts with the fact that allogeneic MHC molecules are poorly immunogenic per se. The aim of the present study is to elucidate whether direct allosensitization induces production of inflammatory mediators that may affect recruitment and activation of immature bystander (host) dendritic cells (DC). These potential mechanisms were studied in vitro by conducting primary allogeneic mixed leucocyte reactions (MLR), mimicking the priming phase in secondary lymphoid organs, and secondary MLR, mimicking the effector phase within the graft. Primary, and particularly secondary, MLR supernatants were found to contain high levels of monocyte/immature DC-recruiting CC chemokines and pro-inflammatory cytokines. Exposure of immature DC to primary or secondary MLR supernatants was found to upregulate CD40 expression and further enhanced lipopolysaccharide-induced interleukin-12 (IL-12) p70 production. Secondary MLR supernatants additionally induced upregulation of CD86 and deviated allogeneic T-cell responses towards Th1 (enhanced interferon-γ production without concomitant induction of detectable IL-4 or IL-10 production). These findings indicate that direct allorecognition may act as a Th1-deviating adjuvant for indirect allosensitization.

    Notes: Eosinophil granular proteins are a useful eosinophilic activation marker in asthmatic patients. In this study, the eosinophil peroxidase (EPO) levels were assessed in different stages of bronchial asthma, in 123 patients suffering from asthma, classified as mild (n=49), moderate (n=49), and severe (n=25), according to the International Consensus Report on Diagnosis and Treatment of Asthma, as well as in 27 healthy controls, with the aim of evaluating the importance of this protein as a severity marker in bronchial asthma, and its possible correlation with parameters such as anamnesis, respiratory function tests, and peripheral blood eosinophil count, and also with some allergologic diagnostic tests, both in vivo and in vitro. The geometric mean serum level of EPO was 9.3±11.3 ng/ml (median±SD) in controls, and 28±37.8 ng/ml in the asthmatic patients. Depending on the asthma severity, the EPO levels were 25±30.5; 29±37.1, and 41 ±47.3 ng/ml in mild, moderate, and severe asthmatics, respectively, being the significant differences between the group of patients with mild and severe asthma (P〈0.001). The number of eosinophils (eos) in peripheral blood was 157±20 eos/mm3 in the controls, 334+35 eos/mm3 in mild asthmatics, 510 ±87 eos/mm3 in moderate asthmatics, and 658±72 eos/mm3 in severe asthmatics, with significant differences between all the groups (from P〈0.05 to P〈0.001). Both the serum levels of EPO and the number of eosinophils were greater in patients with active asthma than in patients with inactive asthma (P〈0.001). Significant negative correlations (P 〈 0.001) were found between serum levels of EPO and FEY, (rs= 0.30), MEF25-75 (rs= -0.33), and MEF50 (rs= -0.34), and a good positive correlation (rs= 0.80, P〈0.001) was found between EPO levels and the number of eosinophils in peripheral blood. We also found a significant positive correlation between eosinophil number and clinical score (rs= 0.54, P〈0.001) and between EPO levels and the mentioned score (rs= 0.46, P〈0.001).

    Notes: Abstract Chronic exposure to mild unpredictable stressors (CMS) has previously been found to reduce the consumption of palatable, sweet solutions in rats. In the present study, the utility of this procedure was assessed in mice. Male AP mice subjected to CMS showed reduced consumption of a 2% or 4% sucrose solution. This effect was reversed by chronic (3 weeks) treatment with the tricyclic antidepressant imipramine (20 mg/kg per day). These results extend previous reports of a generalized decrease in sensitivity to reward (anhedonia) in rats caused by CMS and the efficacy of antidepressant treatment in this paradigm. Chronic unpredictable mild stress in mice appears to provide a realistic animal model of depression.

    Notes: Summary.  Recombinant vaccinia viruses expressing foot-and-mouth disease virus (FMDV) P1 and VP1 genes have been used to study the immune response induced by these viral polypeptides in guinea pigs. Anti-FMDV antibodies, but not neutralizing activity, were detected in the sera from immunized animals. The results indicate that both CD4+ and CD8+ FMDV-specific T cells were induced by the vaccinia recombinants. Consistently with the activation of CD4+ T cells, lymphocytes from immunized animals specifically proliferated in vitro in response to whole virus. The induction of virus-specific CD8+ T cells was determined by CTL assay of immune splenocytes restimulated in vitro with FMDV infected cells. Altogether, the results obtained indicate that both B and T cell immune responses to FMDV are elicited upon immunization of guinea pigs with vaccinia recombinants expressing FMDV structural polypeptides.

    Notes: Abstract This report summarises and builds on the results of the “Directions and Methodologies for Empirical Software Engineering Research” group discussion. In particular, we considered the strengths, weaknesses, opportunities and threats to empirical software engineering research in light of the discussions and presentations during the workshop. The following sections describe each of these aspects of our discussion in turn. In addition, to finalise our discussion we agreed on a three-point plan for future work.

    Notes: Abstract Selected chemicals in sediments and the abundances and biomasses of macrobenthic species were determined at seven stations from two littoral ecosystems in the Gulf of Cádiz. The marine macrobenthic communities were described in both ecosystems that are subjected to different sources and levels of contamination. A qualitative relationship between source of contamination and biological effects for each station is proposed and the results of the univariate and multivariate analysis used are discussed. Univariate analyses using conventional community descriptive parameters (number of species, abundance and richness, Shanon–Weaver and evenness indices) and the numerial contribution of major taxonomic groups (i.e., Polychaeta, Oligochaeta, Mollusca and Crustacea) could not be used as a means of classifying the sites as clean or polluted with respect to the contamination measured. The results showed that multivariate methods are much more sensitive than univariate techniques. Abundance/biomass comparison (ABC) plots classified the macrobenthic communities into different classes mainly related to organic contamination. However, this analysis did not reflect the alteration due to inorganic sources of contamination. This kind of analysis is valuable for the evaluation of alteration of effects but it cannot discriminate between two different phenomena: pollution (adverse effect associated with chemical concentrations) and alteration (adverse effect associated with chemical concentrations or with natural variations).
