Notizen: Abstract Eight male adult schizophrenic patients undergoing chronic treatment with chlorpromazine were investigated. Clinical assessments were made weekly by a psychiatrist who was unaware of the biochemical findings. Blood samples were drawn 3 h after the first daily dose and the plasma concentrations of chlorpromazine and 11 of its metabolites were assessed by a direct scan technique employing quantitative thin layer chromatography. Patients that responded well to treatment had high plasma levels of chlorpromazine and 7-hydroxychlorpromazine; whereas non-responders had high plasma levels of chlorpromazine sulphoxide. These preliminary findings support the hypothesis that chlorpromazine metabolites play an important role in determining clinical response to treatment, which arose from the failure of previous investigators to show a statistically significant relationship between plasma levels of the parent compound and clinical response.

Notizen: Abstract Common bean,Phaseolus vulgaris L., is known to be ‘inefficient’ in nodulation and N2 fixation although it responds to applied nitrogen. An experiment was conducted to identify and to characterize bean cultivars nodulating in the presence of a high level of nitrogen. Sixteen cultivars and a check for inefficient nodulation, OAC Seaforth, were inoculated and grown for 40 days in replicated pots supplied with zero, 3.5 and 10.5 mM combined nitrogen as NO 3 − and NH 4 + . Seven traits relating to nodulation and N2 fixation were all significantly affected by N level (N), cultivar (Cv) and N × Cv interactions (except for root dry weight), indicating that cultivars responded differently to the N treatments. Total dry weight (W) and shoot to root ratio (S/R) increased with the increased N levels. Nodule dry weight (Wn), visual nodulation score (Nv) and nodulation index (Nx) decreased as the N increased. Percent N and N content per plant increased with the increased N level. Plant weight (W) was positively correlated with Wn, Nv and N content and negatively correlated with %N. Nodulation score was positively associated with Wn and plant N content. Genotypes superior in nodulation and N2 fixation in the presence of N were identified. Cultivars Italian Barlotti, California Light Red Kidney, Kentucky Wonder A and Pueblo 152 were selected for further testing and use in improving the nitrate tolerant nodulating characteristic of bean.

Notizen: A carrier-supported mycelial growth of Penicillium chrysogenum was applied to penicillin fermentation system using celite as a support material. Hyphal growth through the pore matrices of the material showed strong anchorages and provided highly stable biofilm growth. With bioparticles developed in such a manner, both cell growth and penicillin production were observed to increase significantly compared to the conventional dispersed filamentous cultures. Maximum values of specific penicillin production rate were found to be constant regardless of the growth form. A three-phase fluidized-bed fermentor was designed and tested for penicillin production using the bioparticles. Two modes of operation, semicontinuous and repeated fed batch, of the fermentor were tried. It was noted that the overgrowth of free mycelia and the development of fluffy loose bioparticles caused poor mixing and made the fermentor operation quite difficult. Control of the bioparticle size and the extension of production phase were therefore considered important to maintain the reactor productivity at a desired level. From the results of repeated fed-batch operation it was found that the control of bioparticle size could be successfully achieved by phosphate-limiting culture condition. Penicillin production under this condition was also observed to be maintained at a high level (about 80% of the maximum) for at least 1 month.
