    Notes: Application of drug substances to the skin for systemic absorption or action in a particular layer of the skin is a rather old approach. However, over the last years it has received much more attention, as a consequence of the development of new membrane-moderated and matrix reservoir devices. As new reservoir systems, solid lipid nanoparticles (SLN™) and nanostructured lipid carriers (NLC™) have been successfully tested for dermal application of different physicochemical substances. The knowledge obtained from rheological investigations of these systems may be highly useful for the characterization of the newly developed topical formulation. In the present study, an oscillation frequency sweep test was used for the evaluation of storage modulus (G_), loss modulus (G_), and complex viscosity (_*) of 12 different SLN and NLC formulations, over a frequency range from 0 to 10 Hz. The lipidic aqueous dispersions were prepared using three different solid lipids (Softisan_138, Compritol_888 and stearyl alcohol) as matrix material. Miglyol_812, tocopherol, sunflower oil and long-chain triacylglycerols were the chosen liquid lipids for NLC preparation. The objective of the present work was to investigate the effect of these different liquid lipids on the rheological properties of aqueous dispersions of NLC as model systems. It was found that the liquid oil component of the formulation has a strong influence on the viscoelastic parameters, which are dependent on the particle size, zeta potential and crystallinity of the lipid particles, as well as on the solid lipid used.

    Notes: Normal term human decidua-chorion, amnion and placenta were examined for possible synthesis of prolactin (PRL) by organ culture of tissue fragments in medium M199. Before incubation, higher PRL concentrations were found in decidua-chorion (27.7±5.7 ng/mg protein; mean±SE) and amnion (19.7±6.9 ng/mg protein) than in placenta (3.1±0.5 ng/mg protein; p 〈0.005). Incubated deciduachorion released significantly more PRL into culture medium than did amnion or placenta. After 72 hours culture, the PRL concentration of decidua-chorion remained high (17.9±6.4 ng/mg protein); that in amnion (2.3±1.9 ng/mg protein) fell to approach that of placenta (1.4±1.0 ng/mg protein). In further studies, fragments of decidua-chorion were incubated in medium RPMI 1640 without or with puromycin (0.04 mM) or cycloheximide (0.1 mM). A decrease in medium PRL concentration was observed within four hours of adding these inhibitors of protein synthesis. At four hours, the PRL content of decidua-chorion cultured in puromycin (12.4±3 8 ng/mg protein) or cycloheximide (11.9±2.9 ng/mg protein) was lower than tissue incubated in medium alone (25.5±4.1 ng/mg protein; p 〈0.05). Comparable differences in tissue content and medium concentration were also seen at 24 and 72 hours. We conclude that human decidua-chorion synthesizes a protein immunologically simjlar to PRL and that this tissue may represent the principal source of amniotic fluid PRL.

    Notes: We report a woman who had a moderate elevation of serum triglycerides with a type III pattern of hyperlipoproteinaemia when taking a normal diet. She developed eruptive xanthomata with a grossly raised serum triglyceride concentration and chylomicronaemia when pregnant and also when taking a combined oral contraceptive pill containing 50 μg of oestrogen. The xanthomata cleared and the triglyceride level fell when the combined oral contraceptive pill was changed to one with a lower oestrogen content, clofibrate was prescribed and the diet was restricted in carbohydrate and fat. Persistent chylomicronaemia is a serious complication of pregnancy because of the risk of pancreatitis and the potential risk of fetal malnutrition. Treatment with diet and clofibrate is indicated. High oestrogen-containing contraceptive pills appear to be contraindicated in patients with type III or other hypertriglyceridaemic states.

    Notes: Abstract— The hypothesis that the ATPase and phosphatidyhnositol (PI) kinase activities of chromaffin vesicle membranes are catalysed by same enzyme was investigated. The two activities exhibited entirely different responses to variations in Mg2+ or Mn2+ concentrations. In the presence of 1 mM ATP, maximal ATPase activity occurred with 1 mM Mg2+ while maximal PI kinase activity required 100 mM Mg2+ Similar differences were observed with Mn2+ with the exception that maximal ATPase activity occurred with 0.5 mM Mn2+ and maximal PI kinase activity occurred with 5 mM Mn2+ Mn2+ was more effective than Mg2+ in stimulating PI kinase activity at low concentrations, but at optimal concentrations of each, the maximal activity obtained with Mg2+ was 5-fold greater than the maximal activity obtained with Mn2+ The heat stabilities of the two enzymes are vastly different. At 50°C the ATPase activity of the intact membranes was stable for up to 20 min while the tl/2 of PI kinase was less than 2 min. After solubilization in Lubrol PX or at higher temperatures both enzymes were less heat stable, but PI kinase was still inactivated at a much greater rate than the ATPase. The evidence suggests that the ATPase and the PI kinase are different proteins.The major phosphorylated product was diphosphatidylinositol and once formed, it was stable. Phosphorylation of membrane protein accounted for less than 10% of the total 32P-incorporated into chromaffin vesicles. SDS gel electrophoresis of the solubilized membranes showed the presence of at least 2 major phosphorylated high molecular weight components.

    Notes: A number of states are following the federal government's lead and requiring environmental impact reviews of new development proposals. This paper examines ambitious EIR programs in Florida and California with the aim of assessing the costs and benefits of their programs, and, by extension, of state-mandated EIR programs in general. The focus is on new housing production. The findings are that EIR shows some promise of enabling communities to better protect themselves from adverse side effects of new development. At the same time, EIR adds what appears to be a modest increment to the overall costs of new housing. However, because EIR has expanded local government planning options, one of the side effects of EIR is likely to be the enhanced ability of localities to exclude moderate income families from new housing.
