Zusammenfassung Albinoratten mit chronischen Elektroden über dem visuellen und dem sensomotorischen Cortex, dem Bulbus olfactorius sowie im dorsalen Hippocampus wurde Ethosuximid intraperitoneal appliziert und sein Einfluß auf die bioelektrische Hirnaktivität und auf Verhaltensparameter untersucht. Photische Nachentladungen und photische Rekrutierung, ausgelöst durch 0,5/sbzw. 7/s-Lichtblitzserien, sind nach Ethosuximid reduziert oder nicht mehr auslösbar; stattdessen beobachtet man gesteigerte Primärkomponenten anstelle der rekrutierten Potentiale. Das EEG ist desynchronisiert. Muskeltonus und durch Klickreize ausgelöste Startle-Reflexe sind dosisabhängig vermindert. Es besteht eine mäßige bis deutliche Verringerung der Herz- und Atemfrequenz. Durch Isatinverabfolgung induzierte Spindelaktivität wird durch nachfolgende Ethosuximid-Applikation unterdrückt, die zuvor geförderte Zahl photischer Nachentladungen vermindert. Mögliche Mechanismen der Ethosuximid-Wirkung im Zentralnervensystem werden erörtert.

Summary The influence of intraperitoneally administered ethosuximide on bioelectrical brain activity and behavioural parameters was investigated in freely moving albino rats with chronic electrodes over the visual and sensory motor cortex, olfactory bulb and in the dorsal hippocampus. Photically evoked afterdischarges and recruiting (0.5/sec or 7/sec light flash series) are reduced or abolished after ethosuximide, while the primary components of evoked potentials are increased. The EEG is desynchronised. Muscle tone and acoustically evoked startle response decrease, depending on the size of the dose administered. Heart beat and respiration rate are slightly slowed down. Isatine—induced spindle activity is abolished after the administration of ethosuximide and the increase of the photically evoked afterdischarge after isatine, is reduced by ethosuximide. The possible mechanisms behind the action of ethosuximide in the central nervous system are discussed.
