    Notes: Weather damage reduces the value of commercial mungbean. but hard-seededness can reduce the level of damage. However. attempts lo breed large- and hard-seeded mungbean varieties have been unsuccessful. To understand the relationship between seed weight and hard-seededness. these trails were investigated using a quantitative trail loci (QTL) mapping approach with a recombinant inbred population derived from a cross between a completely soft-seeded variety and a completely hard-seeded genotype. The two parental genotypes also had a sixfold difference in seed weight. QTL analyses revealed four loci for hard-seededness and 11 loci for seed weight. Two of the hard-seeded ness loci co-localized with seed weight QTL. When seed weight was used as a covariate in the analysis of hard-seededness from the field data, two of the four hard-seeded QTL remained significant with the effect al one of these remaining unchanged. These results explain why retaining hard-seededness in large seeded mungbean lines has been unsuccessful. The existence of a persistent locus, however. indicated that breeding large and persistently hard-seeded varieties of mungbean may be possible.

    Notes: Background:  A 15-fold increased risk of gastrointestinal bleeding has been reported with concurrent use of selective serotonin reuptake inhibitors and non-steroidal anti-inflammatory drugs. Recent guidance cautions against concurrent prescription, particularly in older people.Aim:  To quantify the risk of gastrointestinal bleeding associated with current exposure to non-steroidal anti-inflammatory drugs, selective serotonin reuptake inhibitors, and both drugs concurrently.Methods:  We conducted a case–control analysis of 11 261 cases with upper gastrointestinal bleeding and 53 156 controls matched by gender, age and general practice from computerized primary care data. We coupled this with self-controlled case series analysis.Results:  Both drugs were associated with a twofold increased risk of gastrointestinal bleeding (odds ratio =2.38, 95% confidence interval 2.08–2.72 for selective serotonin reuptake inhibitors and odds ratio = 2.15, 95% confidence interval 2.02–2.28 for non-steroidal anti-inflammatory drugs). This increased risk was marginally higher for concurrent prescription (odds ratio = 2.93, 95%confidence interval 2.25–3.82). The self-controlled analysis showed a greater incidence rate ratio for gastrointestinal bleeding with non-steroidal anti-inflammatory drugs (2.71, 95% confidence interval 2.51–2.91) and lower incidence rate ratio with selective serotonin reuptake inhibitors (1.71, 95% confidence interval 1.48–1.98). The incidence rate ratio when both drugs were combined was 3.25, 95% confidence interval 1.95–5.42. Estimates were similar after restricting to people over 80 years of age. Increased risk of gastrointestinal bleeding was not specifically related to class of non-steroidal anti-inflammatory drugs and was similar when we looked at tricyclic anti-depressants.Conclusions:  Our study suggests that the risk of gastrointestinal bleeding is not substantially increased when non-steroidal anti-inflammatory drugs and selective serotonin reuptake inhibitors are prescribed together, compared with their use alone.

    Notes: Cambrian orogenesis (550–490 Ma) in the Lambert Province of the southern Prince Charles Mountains resulted in three successive stages of deformation. The earliest of these deformations resulted in the development of a layer-parallel foliation (S1) that was folded into macro-scale recumbent folds (F2). Subsequent deformation buckled the rocks into long-wavelength (c. 20 km), SW- to NW-trending antiformal closures (F3) mostly separated from each other by west to SW trending, steeply dipping, high-strain zones. Metapelitic rocks from the region are divisible into two compositional types: a high-Al, -Fe and -K type and a high-Mg, -Ca and -Na type. In rocks of both composition, relic staurolite preceded the formation of upper amphibolite facies garnet + biotite + sillimanite ± muscovite mineral assemblages that record peak pressures and temperatures of c. 650–700 °C and 6–7 kbar. Subsequent decompression of c. 3 kbar is implied from texturally late plagioclase and a reduction in the modal abundance of garnet in the high-Al, -Fe and -K metapelites, and from texturally late cordierite in the more magnesium rocks. This clockwise P–T–t path, with prograde heating followed by rapid decompression, is: (i) equivalent to that recorded in the same-aged rocks at Prydz Bay located 600 km to the north, and (ii) similar to the modelled response of the crust to thickening following continent–continent collision. These results indicate that large areas of East Antarctica were thickened and rapidly exhumed, probably in response to collisional orogenesis during the Early Cambrian. This supports the inference that Early Cambrian orogenesis in the Prydz Bay–Prince Charles Mountains region of East Antarctica marks one of the fundamental lithospheric boundaries within Gondwana.

    Notes: The prograde pressure–temperature (P–T) path for the complexly polydeformed Proterozoic Broken Hill Block (Australia) has been reconstructed through detailed structural analysis in conjunction with calculation of compositionally specific P–T pseudosections of pelitic rock units within a high-temperature shear zone that formed early in the tectonic evolution of the terrane. Whilst the overall P–T path for the Broken Hill Block has been interpreted to be anticlockwise, the prograde portion of this path has been unresolved. Our results have constrained part of this prograde path, showing an early heating event (M1) at P–T conditions of at least c. 600 °C and c. 2.8–4.2 kbar, associated with an elevated geothermal gradient (c. 41–61 °C km−1). This event is interpreted to be the result of rifting at c. 1.69–1.67 Ga, or at c. 1.64–1.61 in the Broken Hill Block. Early rifting was followed by an episode of lithospheric thermal relaxation and burial, during which time sag-phase sediments of the upper Broken Hill stratigraphy (Paragon Group) were deposited. Following sedimentation, a second tectonothermal event (M2/D2) occurred. This event is associated with peak low-pressure granulite facies metamorphism (c. 1.6 Ga) and attained conditions of at least 740 °C at c. 5 kbar. A regionally pervasive, high-temperature fabric (S2) developed during the M2/D2 event, and deformation was accommodated along lithology-parallel, high-temperature shear zones. The larger-scale deformation regime (extensional or shortening) of this event remains unresolved. The M2/D2 event was terminated by intense crustal shortening during the Olarian Orogeny, during which time the first mappable folds within the Broken Hill Block developed.

    Notes: 1 To investigate anti-inflammatory activity of organic germanium, we measured the effect of germanium-concentrated yeast on arachidonic acid release, prostaglandin E2 (PGE2) production, histamine release, and intracellular H2O2 or hydroperoxide generation in RBL 2H3 cells, and carrageenan-induced paw oedema in rats. 2 Germanium-concentrated yeast dose-dependently inhibited carrageenan-induced paw oedema, suggesting that germanium-concentrated yeast has anti-inflammatory activity in acute inflammation. 3 Germanium-concentrated yeast significantly inhibited melittin-induced arachidonic acid release and PGE2 production in RBL 2H3 cells. 4 Germanium-concentrated yeast did not affect melittin-induced histamine release and silica-induced intracellular H2O2 or hydroperoxide generation in RBL 2H3 cells. 5 These results suggest that anti-inflammatory activity of germanium-concentrated yeast appears partly to be related to the inhibition of arachidonic acid release and PGE2 production in RBL 2H3 cells.

    Notes: We present a case of inflammatory epidermolysis bullosa acquisita (EBA) with IgA antibodies to plectin. Analysis of lesional skin biopsies by electron microscopy revealed the split level to be in the sublamina densa zone, corresponding to the diagnosis of EBA. Direct immunofluorescence of perilesional skin demonstrated u-serrated depositions of IgG and IgA that under immunoelectron microcopy were shown to be located in the sublamina densa. In contrast, indirect immunofluorescence on salt-split skin revealed circulating IgA antibodies that stained the roof rather than the floor of the blister. Immunoblotting showed these serum antibodies to be directed to the cytoplasmic hemidesmosomal antigen plectin. The antiplectin specificity of these antibodies was confirmed by ‘knockout’ immunofluorescence analysis; the serum IgA did not bind to skin sections of a patient with plectin-deficient epidermolysis bullosa. To our knowledge, this case demonstrates for the first time the existence of IgA antibodies against plectin.

    Notes: Dysfunction of the ubiquitin-dependent protein degradation system, either at the level of the proteasome itself, or at the level of ubiquitination, may play a role in the pathogenesis of Parkinson's disease (PD) and other related neurodegenerative disorders. We have employed a cellular model of this dysfunction in which lactacystin or epoxomicin, selective pharmacological inhibitors of the proteasome, are applied to primary cultures of embryonic rat ventral midbrain. Proteasomal inhibition with either agent led to apoptotic death specifically within phenotypically defined tyrosine hydroxylase (TH)-positive dopaminergic neurons, with little or no apoptotic death induced in GABAergic neurons. Inhibition of the proteasome also led to the formation of ubiquitin and α-synuclein-positive cytoplasmic inclusions in TH-positive and TH-negative neurons. Inclusions were observed in viable as well as apoptotic neurons, and required new or ongoing transcription. Tyrosine hydroxylase immunolabeling was often present within the inclusions. Such mislocalization may lead to dysfunction of dopamine biosynthesis. Interestingly, dopaminergic neurons, unlike other neurons within these cultures or cultured cortical neurons, failed to induce the chaperone Hsp70 in response to proteasomal inhibition. This failure may explain in part the increased sensitivity of these neurons to proteasomal inhibitors.

    Notes: Methyllycaconitine (MLA) is reported to be a selective antagonist for the nicotinic acetylcholine receptor α7 subtype and has been found in animal behavioral studies to reduce nicotine self-administration and attenuate nicotine withdrawal symptoms. While MLA crosses the blood–brain barrier (BBB), no studies have assessed brain uptake in animals subjected to chronic nicotine exposure. Given that chronic nicotine administration has been reported to alter BBB parameters that may affect the kinetic BBB passage of MLA, we evaluated MLA brain uptake in naive and S-(–)nicotine-exposed rats (4.5 mg/kg/day for 28 days; osmotic minipumps) using in situ rat brain perfusions. Our results demonstrate that in situ3H-MLA brain uptake rates in naive animals approximate to intravenous kinetic data (Kin, 3.24 ± 0.71 × 10−4 mL/s/g). However, 28-day nicotine exposure diminished 3H-MLA brain uptake by ∼60% (Kin, 1.29 ± 0.4 × 10−4 mL/s/g). This reduction was not related to nicotine-induced 3H-MLA brain efflux or BBB transport alterations. Similar experiments also demonstrated that the passive permeation of 14C-thiourea was diminished ∼24% after chronic nicotine exposure. Therefore, it appears that chronic nicotine exposure diminishes the blood–brain passive diffusion of compounds with very low extraction rates (i.e. permeability-limited compounds). These findings imply that the pharmacokinetics of neuropharmaceutical agents that are permeability limited may need to be re-evaluated in individuals exposed to nicotine.

    Notes: The aim of this work was to investigate the interrelated effects of glucose, nitric oxide (NO) and erythropoietin on neuronal survival in retinal cultures, thereby exploring the mechanism of neuronal death in the diabetic retina. Rat retinal cells were cultured in low (5 mm) or high (15 mm) glucose concentrations. After 9 days, cell viability was assessed by (3,4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay and NO production was determined by the Griess reaction. Immunohistochemistry was used to quantify GABA-labelled neurones and cells staining for DNA breakdown. High or low glucose concentrations had no effect on basal NO production or the survival of neurones in culture, but treatment with N-nitro-l-arginine methyl ester reduced extracellular levels of NO and increased neuronal survival at both concentrations of glucose. Erythropoietin decreased cell death and NO levels, but only in cultures grown in low concentrations of glucose. It is concluded that erythropoietin's neurotrophic function in the retina is attenuated at glucose concentrations similar to those which occur in diabetes.
