    Notes: There is evidence that an inflammatory microglial reaction participates in the pathophysiology of dopaminergic neuronal death in Parkinson's disease and in animal models of the disease. However, this phenomenon remains incompletely characterized. Using an in vitro model of neuronal/glial mesencephalic cultures, we show that the dopaminergic neurotoxin 1-methyl-4-phenylpyridinium (MPP+) stimulates the proliferation of microglial cells at concentrations that selectively reduce the survival of DA neurones. The mitogenic action of MPP+ was not the mere consequence of neuronal cell demise as the toxin produced the same effect in a model system of neuronal/glial cortical cultures, where target DA neurones are absent. Consistent with this observation, the proliferative effect of MPP+ was also detectable in neurone-free microglial/astroglial cultures. It disappeared, however, when MPP+ was added to pure microglial cell cultures suggesting that astrocytes played a key role in the mitogenic mechanism. Accordingly, the proliferation of microglial cells in response to MPP+ treatment was mimicked by granulocyte macrophage colony-stimulating factor (GM-CSF), a proinflammatory cytokine produced by astrocytes and was blocked by a neutralizing antibody to GM-CSF. Thus, we conclude that the microglial reaction observed following MPP+ exposure depends on astrocytic factors, e.g. GM-CSF, a finding that may have therapeutic implications.

    Notes: Background:  Lupus erythematosus (LE) is a chronic autoimmune disease with a broad clinical spectrum reaching from primarily cutaneous manifestations [cutaneous LE (CLE)] up to systemic disease [systemic LE (SLE)]. In patients with SLE, the expression of activation markers on circulating T cells reflects disease activity. Here, we investigated whether this also holds true for patients with CLE.Patients and methods:  The expression of the activation markers human leukocyte antigen (HLA)-DR and CD25 on circulating T lymphocytes was measured by flow cytometry in 24 patients suffering from different types of active CLE. Simultaneously, the disease activity was assessed clinically using a CLE activity index. Eighteen healthy donors were analyzed for control purposes.Results:  HLA-DR was expressed on a significantly elevated percentage of both CD4+ and CD8+ circulating T cells in active CLE patients when compared with healthy controls. The percentage of HLA-DR-expressing T lymphocytes closely correlated with the disease activity. Interestingly, in disseminated scarring chronic discoid LE, a significantly increased percentage of CD25+ cells was observed only in the subset of skin-homing cutaneous lymphocyte antigen (CLA)+CD4+ and CD8+ T cells.Conclusion:  Our results provide evidence that activation markers on peripheral blood T cells might help to objectively assess the disease activity in CLE. Furthermore, a significant population of CD25+CLA+CD8+ T cells can only be detected in a subgroup of patients with disseminated scarring CLE and might reflect the systemic expansion of activated cytotoxic T lymphocytes involved in destruction of epidermal tissue.

    Notes: Hydrogenosomes are anaerobic forms of mitochondria in at least four independent groups of anaerobic eukaryotes, the parabasalids, the heterolobosea, the ciliates and the anaerobic chytridiomycetes. Their hydrogenosomes generate ATP from the fermentative decomposition of pyruvate but lack the hallmark pathways of mitochondrial metabolism, the TCA cycle and membrane-associated electron transport. They also, with one exception, do not contain their own genome. We are analysing the proteome of the hydrogenosomes from the chytridiomycete Neocallimastix frontalis through 2D electrophoresis and tandem mass spectroscopy to assess the functional capacities of these organelles and to further characterize their relationship with mitochondria.

    Notes: Trichomonas vaginalis is an anaerobic protozoan parasite of the human urogenital tract. It lacks functional mitochondria but contains hydrogenosomes, anaerobic forms of mitochondria that generate ATP from the fermentative degradation of pyruvate. To determine the similarities and differences between hydrogenosomes and mitochondria, we are characterizing the proteome of purified hydrogenosomes from T. vaginalis by 2D electrophoresis and ESI-Q-TOF MS/MS. In the process, we have identified two proteins of bacterial origin that may function as peroxidases and this may be part of the answer to the unsolved question of how hydrogensomes in T. vaginalis are protected against the detrimental effects of oxygen: rubrerythrin is a non-heme iron protein found predominantly in anaerobic bacteria that can reduce H2O2 and was shown to complement catalase-deficient E. coli mutants. Among eukaryotes we have found only one further rubrerythrin homologue in the anaerobic, amitochondriate amoeba Entamoeba histolytica. Peroxiredoxins are ubiquitous peroxidases using thiols to reduce alkyl hydroperoxides. T. vaginalis hydrogenosomal thiol peroxidase (Tpx) shows 65% homology to bacterial 20-kDa Tpx. These peroxidases contain three highly conserved Cys residues, Cys62, Cys82, and Cys95, and belong to a peroxiredoxin subfamily that previously contained eubacterial sequences only. The finding of two putative bacterial-type peroxidases in T. vaginalis hydrogenosomes suggests that proteins involved in peroxide detoxification in these anaerobic organelles may have been acquired laterally from eubacteria.
