    Notes: Significant progress has been made in understanding the structure of high molecular weight (HMW) glutenin subunits and their role in determining the end use quality of wheat grains. However, few reports have dealt with the development and characterization of knock out mutants for HMW glutenin subunit genes. Here, the molecular analysis of MB14, a mutant derived from an elite Chinese wheat variety Xiaoyan 54 through chemical mutagenesis is described. SDS-PAGE and Western blot experiments revealed that, in the seeds of homozygous MB14 plants, the expression of the 1Bx14 subunit was specifically blocked whereas the remaining four subunits (1Ax1, 1By15, 1Dx2, 1Dy12) accumulated to levels comparable to those in the wild type plants. The 5′-flanking region and the open reading frame (ORF) of the mutant 1Bx14 allele were amplified and compared to the corresponding regions of wild type 1Bx14. The nucleotide sequences of the 5′-flanking regions from the mutant and wild type 1Bx14 alleles were identical. However, the ORF of the mutant allele differed from that of the wild type 1Bx14 by three point substitutions, one of which resulted in a premature stop codon in the mutant ORF. Interestingly, the mutant 1Bx14 allele was still transcribed in the developing seeds, but no truncated translation product could be detected by Western blot analysis. Potential application of the 1Bx14 knock out mutant in studying the biological function of 1Bx14 and its contribution to the end use quality control in hexaploid wheat is discussed.

    Notes: Three lines conferring resistance to powdery mildew, Pm97033, Pm97034 and Pm97035, were developed from the cross of Triticum durum-Haynaldia villosa amphidiploid TH3 and wheat cv.‘Wan7107’ via backcrosses, immature embryo and anther culture. Genomic in situ hybridization analysis showed that these lines were disomic translocation lines. Cytogenetic analysis indicated that the F1 plants of crosses between the three translocation lines and ‘Wan7107’ and crosses between the three translocation lines and substitution line 6V(6D) formed 21 bivalents at meiotic metaphase I. Aneuploid analysis with ‘Chinese Spring’ double ditelocentric stocks indicated that the translocated chromosomes were related to chromosome 6D. Biochemical and restriction fragment-length polymorphism (RFLP) analyses showed that the translocation lines lacked a specific band of 6VL of H. villosa compared with the substitution and addition lines but possessed specific markers on the short arm of the 6V chromosome of H. villosa. The three translocation lines lacked specific biochemical loci and RFLP markers located on chromosome 6DS. The results confirmed that Pm97033, Pm97034 and Pm97035 were T6DL.6VS translocation lines.

    Notes: Xeroderma pigmentosum (XP) is a recessively inherited genodermatosis associated with extreme sun sensitivity, defective repair of several types of sunlight induced adducts in cellular DNA, and numerous, early-onset skin cancers. The dry, rough skin corresponds to progressive cytologic atypia and loss of polarity in the underlying epidermis. Associated with these changes are immune deficiencies against ultraviolet radiation-induced skin cancer. 5-Fluorouracil (5-FU) is a DNA synthesis antimetabolite used against several types of cancers. Applied topically in normal subjects it is associated with moderate to severe inflammation in areas where actinic keratoses have arisen followed by ablation of the actinic keratoses which is dependent on the inflammation. We applied 5-FU to the sun-exposed skin of two patients with XP, a 14 year-old light complected black male and a 14 year-old Caucasian female. No inflammation was observed, but marked improvement in the clinical presentation of the skin was seen, as well as an absence of new malignancies. This change was confirmed histopathologically and correlated with normalization of polarity and cytologic changes in the epidermal cells. These histologic findings were quantitated using computerized image analysis. These results may be due to activation of alternative DNA repair pathways in these nucleotide excision repair deficient cells.

    Notes: Ischemic brain injury is acute local inflammation, leading to accumulation of pro-inflammatory cytokines. Cytokines influence the recruitment of leucocytes and play a key role in the inflammatory injury processes. Recently, a number of studies have demonstrated a close relationship between brain ischemia and cytokines. Interleukin-17 (IL-17) is a newly identified T-cell-specific cytokine. In this study, we evaluated the source and the action of IL-17 over the course of cerebral ischemia in rats (Sprague-Dawley) and humans. The levels of IL-17 in the ischemic hemisphere of the human brain, which was removed at necropsy, were assayed immunohistochemically. In rats, permanent middle cerebral artery occlusion (pMCAO) was obtained by inserting nylon monofilament into the right external carotid artery, occluding the right middle cerebral artery. The expression of IL-17 mRNA in rat was assayed using oligoprobe in situ hybridization. IL-17 production by neuroglial cells was assayed by double-staining using antibody glial fibrillary acidic protein (GFAP) and antibody IL-17. Levels of IL-17 were elevated in the ischemic hemispheres of human brain compared with the opposite normal hemispheres and peaked at days 3–5 after brain ischemia. The IL-17-positive cells were found in the ischemic lesion region. IL-17 mRNA was also elevated in ischemic hemispheres of pMCAO-operated rats, which were slightly elevated after 1 h and peaked at 6 days. IL-17 and GFAP double-stained were extensive in rat ischemic hemisphere. The ischemia-induced IL-17 expression in human brain reported here for the first time was very similar to that in rat model except that the peak was slightly earlier. We found for the first time that IL-17 was involved in an intense inflammatory reaction of brain ischemic injury in human. In pMCAO-operated rats, our findings suggest that IL-17 is produced by the neuroglial cells in the brain region undergoing ischemic insult. We suggest that in additional to T cells the neuroglial cell may be another cellular origin of IL-17 in later progression of brain ischemia.

    Notes: White spot syndrome virus (WSSV) is a large double-stranded DNA virus, causing considerable mortality in penaeid shrimp and other crustaceans. WSSV produces five major structural proteins, including two major envelope proteins, VP28 and VP19. To produce VP28 and VP19 as a single protein for antibody production, DNA sequences encoding both open reading frames were fused together and cloned into pET-22b(+) expression vector. The fusion protein, VP(19+28), was expressed in Escherichia coli, purified using Ni2+His affinity chromatography and injected into a rabbit. Antiserum collected from the immunized rabbit was tested in vivo for ability to protect crayfish, Cambarus clarkii, from disease caused by WSSV. Fifteen days after challenge with WSSV, treatment with VP(19+28) antiserum gave 100% protection against disease in the ambient temperature range of 15–22 °C and 65% protection at a constant temperature of 26 °C. These results demonstrated VP(19+28) antiserum is effective in protection of crayfish from WSSV and confirmed that VP19 and VP28 play an important role in WSSV host infection. Targeting both VP19 and VP28 may be effective for the design of both immunotherapeutic medicines and reagents to detect WSSV.

    Notes: Structural changes in C15 Laves phase DyCo2 on heating using a pressure differential scanning calorimeter (PDSC) in a hydrogen atmosphere between 0.1 and 5.0 MPa were investigated by a powder X-ray diffractometer (XRD), a differential scanning calorimeter under an argon flow atmosphere (Ar-DSC), atransmission electron microscope (TEM) and a hydrogen analyzer. As the temperature of DyCo2 increases, the reactions such as hydrogen absorption in a crystalline state, HIA (hydrogen-induced amorphization), precipitation of DyH3 and decomposition of the remaining amorphous phase into β-Co + DyH3 occurredexothermically for every hydrogen pressure. The mechanism of HIA in DyCo2 is discussed on the basis of the experimental results

    Notes: Ba8Ga16Ge30 clathrate material has being intensely investigated as a candidate of promisingthermoelectric materials. In this work, Ba8Ga16+xSbxGe30-2x (x=0,1) single crystals have been synthesizedby the Ga flux method, using high purity elemental Ba, Sb, Ga, and Ge as starting materials. PowderX-ray diffraction, Scanning electron microscopy equipped with energy-dispersive X-ray spectroscopyand Raman spectroscopy were used to characterize the single crystals. Seebeck coefficient of the crystalswas measured from 300 to 800 K. The result indicates that the crystals are p-type semiconductor, which isdifferent from the crystals synthesized by the Czochralski method. The Seebeck coefficient almost doesnot change after doping Sb in the whole temperature measured and it is in the range of 200 to 300μV/K

    Notes: Infrared-based lie detection experiment methodology can open the human life window(0.6µm-16µm) through which the body spontaneous or active infrared responding images be gatheredand analyzed by spectrum scoring in features of geography and computation. These raw imagesmainly load the indirect physiological, biochemical or direct brain evidences of cognitive lie andtruth. Aiming at the mimic play card test, three tests of lie detection based on the medium or nearinfrared spectrum (NIRS) in four kinds of spectrum analysis are discussed. The instruments includethe lock-in phase amplifier of Stanford University, TTM (Thermal Texture Maps) in US patent, andself-made auricular point NIRS (Chinese Jiangsu appraisal certificate No. 1017). The subjects’signals sampling positions are on glabella, brows, abdomen or auricular point Antitragus No. 2. Thespectrum analysis methods utilize the self-built feature extraction algorithms, the wavelet microscopy,and the bispetrum in validities of 92%, 100%, 80% and 80%. The results support the lie-truth law of“truth be the baseline of lie detection in view of brain power” (the statistical finding from review of3D-MRI lie-detection reports included our work, subjects n = 4) for making foundation to the futurelie detection 3D-SoC design

    Notes: AgPb18SbTe20 nano-powders have been synthesized by a hydrothermal synthesis method at 180°C for 20 h, using AgNO3, Pb(NO3)2, Sb(NO3)3, Na2TeO3 as starting materials and KBH4 as a reductant.AgPb18SbTe20 bulk thermoelectric materials were obtained by pressureless sintering at 450 °C or 520 °Cunder argon atmosphere, after uniaxial pressing the as-synthesized powders into pellets. The phasecomposition and thermal effects of as-prepared powders were characterized by X-ray diffraction (XRD)and DSC-TG analyses, respectively. Electrical properties of the as-prepared bulk materials weremeasured from room temperature to about 700 K, and the maximum power factor of 85 μW/mK2 wasachieved at 600 K for the sample sintered at 723 K for 3 h
