    Notes: In the present study, magnetic nanowires with various cross-sectional dimensions have been fabricated by evaporating a magnetic material on the side edges of the patterned nonmagnetic thin film. The method allows the lengths of the two short axes to be defined precisely by controlling the thickness of the oxide (Wx) and magnetic material (Wy). The MFM images of the produced Co wires (2 μm length) with dimensions of Wx=Wy=200 nm and Wx=Wy=50 nm present the single domain configuration. The MFM and magnetoresistance (MR) measurement demonstrate that the longitudinal magnetization switching field increases with the decrease of Wx and Wy, which becomes 1.3 kOe for wires with Wx=Wy=50 nm. The profile of the MR hysteresis suggests that considerable coherent rotation takes place before the irreversible longitudinal switching. Transverse saturation fields larger than 6 kOe were measured in the orthogonal two short axis directions, which confirms the successful fabrication of three-dimensional wires with comparable cross-sectional dimensions. © 2000 American Institute of Physics.

    Notes: The rice dwarf1 (d1) mutant, which is deficient in an α subunit (Gα) of heterotrimeric G protein, was used to obtain specific evidence on the functions of Gα protein in defence signalling in rice. Using proteome analysis, a probenazole-inducible protein (PBZ1) was detected in the cytosolic fraction of leaf blade of the wild type, but not the d1 mutant. After treatment with probenazol, PBZ1 reached maximal levels at 72 h in the wild type but 96 h in the d1 mutant. The induction of PBZ1 by probenazole treatment was inhibited by protein kinase inhibitors. A 48-kDa putative mitogen-activated protein kinase (MAPK) and a 55-kDa putative Ca2+-dependent protein kinase (CDPK) showed lower activities in the cytosolic fraction of the d1 mutant than that of the wild type. The activities of these protein kinases were enhanced at 24 h in the wild type and 48 h in the d1 mutant after probenazole treatment. Although the d1 mutant responded to the rice blast fungus similarly to the wild type, the d1 mutant developed rice blight symptoms earlier than the wild type when infected with Xoo. In addition, the blight symptoms were more severe on the mutant than on the wild type, and wilting was frequently observed in the d1 mutant. Furthermore, induction by the bacterial infection of the 48-kDa putative MAPK and PBZ1 was delayed by 2 and 4 d, respectively, in the d1 mutant compared with the wild type. These results indicate that the Gα protein plays a role in the induction of PBZ1 and protein kinases by probenazole and Xoo, and suggest that the 48-kDa putative MAPK may be involved in a signalling pathway for resistance to bacterial infection.

    Notes: Protein kinases in plants have not been examined in detail, but protein phosphorylation has been shown to be essential for regulating plant growth via the signal transduction system. A Ca2+- and phospholipid-dependent protein kinase, possibly involved in the intracellular signal transduction system from rice leaves, was partially purified by sequential chromatography on DE52, Phenyl Superose and Superose 12. This protein kinase phosphorylated the substrate, histone III-S, in the presence of Ca2+ and phosphatidylserine. The apparent molecular mass of the Ca2+- and phosphatidylserine-dependent protein kinase (Ca2+/PS PK), determined by phosphorylation in SDS-polyacrylamide gel containing histone III-S, was 50 kDa. The protein kinase differed from Ca2+-dependent protein kinase (CDPK) in rice leaves in that Ca2+/PS PK showed phospholipid dependency and the molecular mass of Ca2+/PS PK exceeded that of CDPK. Investigations were carried out on changes in Ca2+/PS PK and CDPK activity in the cytosolic and membrane fractions during germination. The maximum activity of Ca2+/PS PK in the cytosolic fraction was observed before imbibition and that of CDPK in the membrane fraction was noted at 6 days following imbibition. Protein kinases are likely to regulate plant growth through protein phosphorylation.

    Notes: Abstract The response of plants to cold stress is not well understood at the biochemical level, although it has been studied extensively at the ecological level. To investigate whether protein phosphorylation may play an important role in cold stress, we exposed rice seedlings to low temperatures, prepared protein extracts from the leaves and incubated these in the presence of [γ-32P]ATP. The proteins were then separated by two-dimensional polyacrylamide gel electrophoresis. While several proteins were found to be phosphorylated upon cold stress one protein, pp35, which has an isoelectric point of 8.0, was more phosphorylated than the others. The pp35 protein was found to be phosphorylated when rice seedlings were incubated for 6 h at 5°C before the leaf protein extract was prepared and radioactive labeling was performed. The pp35 was, however, significantly more phosphorylated in cold-tolerant rice varieties. Antibodies were raised against purified pp35 in adult rabbits. Using this pp35 antibody, which can recognize the RuBisCO large-chain subunit (LSU), and from amino acid sequencing of pp35, we were able to identify and confirm the pp35 protein as the fragment of RuBisCO LSU (EC 4.1.1.39). Phosphorylation of the RuBisCO LSU may be important in cold tolerance.

    Notes: Abstract  Ca2+-dependent protein kinases (CDPKs) play an important role in plant signal transduction. Protein kinase(s) activities induced by 5°C cold stress in rice (Oryza sativa L.) seedlings were investigated in both leaf and stem tissues in an early (up to 45 min) and late (up to 12 h) response study. The leaf had 37-, 47- and 55-kDa protein kinase activities, and the stem had 37-, 47- and 55-kDa protein kinase activities. A 16-kDa protein showed constitutive kinase activity in the rice seedling leaf and stem. It was further identified that the 47-kDa protein kinase activity induced by cold in both the cytosolic and membrane fractions of the stem was strictly Ca2+-dependent. This CDPK activitiy increased in the presence of the Ca2+ ionophore A23187 in stem segments, whereas it was decreased by the Ca2+ channel blocker, LaCl3, and the Ca2+ chelator, EGTA. The general protein kinase inhibitor, staurosporine, completely inhibited this CDPK activity in vitro, and both W7, a calmodulin antagonist, and H7, a protein kinase C inhibitor, could only partially decrease this activity. The protein phosphatase inhibitor, okadaic acid, increased CDPK activity. This CDPK activity was also induced by salt, drought stress and the phytohormone abscicic acid. Among the 18 rice varieties tested, this cold-induced 47-kDa CDPK activity was stronger in the cold-tolerant varieties than in the sensitive ones.

    Notes: Abstract Ultra-fine diamond powders were prepared from C2H2 and H2 by quenching a d.c arc plasma jet on a metal disc cooled with water. The powders obtained were agglomerated with very small particles about 20 nm in diameter, with an average size of about 80 nm. Raman spectra, X-ray diffraction, and transmission electron micrographs of them showed that each particle consisted of very small diamond crystals and a small amount of amorphous carbon, and that a single crystal particle had a twin plane. The formation mechanism of the powders has been briefly discussed from the viewpoint of the supersaturation ratio based on the classical theory of homogeneous nucleation.
