    Notes: Abstract The role of histamine H3-receptors in the control of acetylcholine release from peripheral cholinergic neurons was evaluated in the isolated guinea pig ileum, previously loaded with3H-choline. When tested in the presence of H1- and H2-blockade, histamine (0.1–100 μmol/l) and (R)α-methylhistamine (0.01–1 μmol/l) dose-dependently reduced the electrically-evoked choline outflow, with (R)α-methylhistamine being a partial agonist. Selective H3-receptor blocking drugs, thioperamide (0.1 μmol/l) and impromidine (0.1 μmol/l) reversed the histamine-induced inhibitory, effect. These data suggest that intestinal cholinergic nerves are endowed with histamine H3-receptors whose activation produces an inhibitory effect upon acetylcholine release. The practical implications of these findings are obvious.

    Notes: Abstract CD44 is a transmembrane glycoprotein, which can exist in a multitude of isoforms due to alternative splicing of the pre-mRNA. We have generated monoclonal antibodies to several of these variant regions, which are encoded by 10 additional exons in the extracellular part of the molecule. CD44 variant isoforms have been reported to be involved in the malignant progression of rat and human tumours. The precise localization of CD44 variant isoforms in normal developmental and morphogenetic processes is essential for diagnostic studies of human tumorigenesis. Therefore, we have analysed a large number of different human tissues by immunohistochemistry for the expression of CD44 isoforms containing either exons 4v, 6v or 9v. Expression of exon 9v-isoforms was detected in almost all epithelia analysed, with a few exceptions. Exon 6v isoforms are expressed only in squamous and glandular epithelia, e.g. skin epidermis, sweat and sebaceous glands, oesophagus, ducts of the mammary gland, salivary and prostate glands. Detection of exon 4v-encoded isoforms was restricted to the epidermis and the oesophagus. Similar tissue distributions of CD44 variant isoforms were observed in 10-week-old fetal tissues. Since one of the ligands of CD44 is hyaluronic acid (HA), we also analysed the tissue distribution of HA synthetase. HA synthetase was detected in all tissues analysed, showing good correlation with the expression of the standard form of CD44, CD44s.

    Notes: Abstract Alpha 6 integrin is an adhesion molecule that connects cells with extracellular matrix molecules of the laminin family. The laminin interaction seems to be essential for cell differentiation during embryogenesis and for the subsequent maintenance of tissue integrity in the adult. Alpha 6 integrin can also interact with laminin-independent cellular ligands and in this way plays a role in homing of leucocytes. Furthermore, in cancer biology α6 integrin has an important role in metastasis and as a possible new prognostic factor; exact knowledge of α6 integrin distribution in normal human tissues is therefore a crucial element. By immuno-histochemical methods we have screened α6 integrin expression of representative human tissues from the adult and the embryonic organism. All tested epithelia were α6 integrin positive, except for the endocrine cells of the pancreas and the adrenal glands. Heterogeneous staining was found on non-epithelial tissues. Strong staining was evident in peripheral nerves (Schwann cells), germ and Sertoli cells, endothelia, and smooth muscle cells of the myometrium. Weak staining was found in nerve cells of the stratum granulosum, the microglia, Kupffer's cells and stromal cells of the ovary. All fibroblasts, striated muscle cells and astrocytes were negative. The tissue distribution of α6 integrin and the semi-quantitative estimation of their expression level should provide a better understanding of α6 integrin function under normal and phathological conditions, in particular in tumour progression.

Motivated by recent experimental findings, we investigate the possible occurrence and characteristics of quasicrystalline order in two-dimensional mixtures of point dipoles with two sorts of dipole moments. Despite the fact that the dipolar interaction potential does not exhibit an intrinsic length scale and cannot be tuned a priori to support the formation of quasicrystalline order, we find that configurations with long-range quasicrystallinity yield minima in the potential energy surface of the many particle system. These configurations emanate from an ideal or perturbed ideal decoration of a binary tiling by steepest descent relaxation. Ground state energy calculations of alternative ordered states and parallel tempering Monte-Carlo simulations reveal that the quasicrystalline configurations do not correspond to a thermodynamically stable state. On the other hand, steepest descent relaxations and conventional Monte-Carlo simulations suggest that they are rather robust against fluctuations. Local quasicrystalline order in the disordered equilibrium states can be strong.

    Description / Table of Contents: Zusammenfassung Es wird gezeigt, daß in der Wiederherstellungschirurgie bei Benutzung spindelförmiger Exzisionen mit Steigerung des Verhältnisses Länge L/DurchmesserW die Wahrscheinlichkeit eines zufriedenstellenden Verschlusses erhöht wird. Die Ergebnisse zeigen, daß es einen Minimalwert für das VerhältnisL/W bei einem vorgegebenen Hautbereich gibt, bei dessen Unterschreitung an den Enden der Exzision bei Verschluß kleine „stehende Kegel“ auftreten. Das läßt den Schluß zu, daß es ein Maximum für den Winkel gibt, der am Ende der Exzision durch deren beide Seiten gebildet wird, welcher noch zufriedenstellend verschlossen werden kann. Bei anderen chirurgischen Verfahren können sich nicht nur stehende, sondern auch liegende Kegel bilden, wenn bei dem Verfahren ein Winkel geöffnet statt geschlossen werden muß. Die Erscheinung dieser stehenden und liegenden Kegel in menschlicher Haut, die der Wiederherstellungschirurgie ausgesetzt ist, ist, wie die Untersuchungen zeigen, abhängig von den Eigenschaften des Gewebes bei einachsiger Kompression. Für diese Kompression wird ein kritischer Wert abgeleitet, ehe sich die Haut in vivo zu einem Grat oder Kegel verzieht. Es wird ein halbempirisches Verfahren entwickelt, mit dessen Hilfe der Chirurg den maximalen Exzisionswinkel vorherzusagen vermag, der ohne die Bildung von stehenden oder liegenden Kegeln geschlossen werden kann.
    Notes: Summary It can be demonstrated in reconstructive surgery when using a fusiform excision that as the ratio of its lengthL to widthW is increased, the probability of a satisfactory closure is also increased. It is found that there is a minimum value ofL/W for a given area of skin beyond which small “standing cones” appear at the ends of the excision on closure. This indicates that there is a maximum value of the angle, included at the ends of the excision by its two sides, that can be closed satisfactorily. In other surgical procedures not only standing cones may form but also lying cones when the prodecure involves opening an angle instead of closing it. The phenomena of these standing and lying cones in human skin subject to reconstructive surgery are shown to be dependent on the characteristics of the tissue in uniaxial compression. It can be demonstrated that there is a critical value of this compression before the skin in vivo “buckles” into a ridge or cone. A semi empirical procedure is outlined that allows the surgeon to predict the maximum excisional angles which may be closed or opened without causing the formation of standing or lying cones.
