Notes: We have isolated a hybrid gene, composed of the first 455 nucleotides of hisP and nucieotides 275–1107 of malK, the genes coding for the nucleotide-binding components of the high-affinity transport systems for histidine and maltose in Salmonella typhimurium, respectively. The fusion had occurred by recombination within 11 homologous base pairs located between the two DNA fragments. In the chimeric protein peptidic motifs A and B, proposed to be part of the nucleotide-binding fold, originate from HisP and MalK, respectively. Plasmid pES42-39, harbouring the hybrid gene, was shown to complement only a malK mutation but failed to complement a hisP deletion mutation. The chimeric protein was identified by immunoblotting as a protein with an apparent molecular mass of 49kDa. Removal of the C-terminal 77 amino acid residues from the chimeric protein resulted in the loss of function in transport. In contrast, 51 amino acid residues could be removed from the C-terminus of wild-type MalK without any effect. Upon overproduction the chimeric protein, as wild-type MalK, inhibited expression of the malB regulon. However, both truncated proteins, when overproduced, did not exhibit this activity. Based on these results, a tentative model of the functional domains of MalK is presented.

Notes: In the framework of the European project aimed at the sequencing of the Bacillus subtilis genome the DNA region located between gerB (314°) and sacXV (333°) was assigned to the Institut Pasteur. In this paper we describe the cloning and sequencing of a segment of 97 kb of contiguous DNA. Ninety-two open reading frames were predicted to encode putative proteins among which only forty-two were found to display significant similarities to known proteins present in databanks, e.g. amino acid permeases, proteins involved in cell wall or antibiotic biosynthesis, various regulatory proteins, proteins of several dehydrogenase families and enzymes II of the phosphotransferase system involved in sugar transport. Additional experiments led to the identification of the products of new B. subtilis genes, e.g. galactokinase and an operon involved in thiamine biosynthesis.

Description / Table of Contents: Summary Three measurements were taken on each of 25 lower jaws of hare to estimate the possible interdependence in growth in length of the jaw, the tooth, and in width of the oral cavity. The date were not sufficient to establish clearly an age dependence, but a high probability points in this direction. This work will hopefully instigate further collection of data to enable age-estimation with the aid of age-dependent growth factors.

Notes: Abstract One of the primary obstacles to be overcome for the development of economical fuel cells is the high cost of the membrane electrolyte. The currently favoured polymer electrolytes consist of poly(tetrafluoroethylene) backbone structures and poly(perfluorosulphonic acid) side chains. In an effort to find lower cost membranes, some radiation-grafted copolymer membranes were investigated. All the membranes contained poly(styrenesulphonic acid) side chains. Three different backbone polymer structures were studied: low-density poly(styrene), poly(tetrafluoroethylene)/poly(perfluoropropylene), and poly(tetrafluoroethylene). The results indicate that the membrane consisting of a poly(tetrafluoroethylene)/poly(styrenesulphonic acid) copolymer is a promising candidate as a fuel-cell electrolyte.

Notes: Summary Comparative labelling of the mouse brain with tritiatedd, l-proline andl-proline reveals a dissimilarity of incorporation between the optical isomers of this amino acid. The differential regional distribution pattern ofd-proline can be correlated to the cerebral distribution of the enzyme activity ofd-aminoacid-oxidase in the mouse brain.

Notes: Summary The influence of transforming growth factor-β (TGF-β) on hematopoiesis has been evaluated by adding blocking antibodies against TGF-β to colony forming assays (CFU-c). When optimum concentrations of recombinant growth factors, granulocyte-macrophage colony stimulating factor (GM-CSF), and interleukin-3 (IL-3) were added to stem cells from the peripheral blood of healthy individuals and certain patients with tumors or HIV infection, the anti-TGF-β capable of blocking 5 ng/ml of active TGF-β had no significant influence on erythroid or myeloid colony formation. However, in certain immunosuppressed individuals, anti-TGF-β resulted in a significant decrease of erythroid colony formation and slight suppression of myeloid colony formation. The significant inhibition of hematopoiesis by plasma of HIV patients could be due to the presence of active forms of TGF-β. The results of the blocking experiments are consistent with the concept that TGF-β in low concentrations is essential for erythropoiesis and myelopoiesis but that higher levels of TGF-β primarily inhibit erythropoiesis in vitro. TGF-β serves as a coordinating factor when efficient recruitment of granulocytes and monocytes is more essential than erythropoiesis and stem cell growth.

Notes: Summary In 26 patients (aged 43–74 years) polygraphic night sleep recordings, prior to and following long term administration ofl-DOPA, were performed. These results were correlated with the neurological, psychiatric and psychological test findings registered in each patient. There is a correlation between sleep disturbances and clinical symptoms: marked signs of Parkinson's disease are associated with a significant delay in the onset of sleep, prolonged waking periods and a reduction of light synchronous sleep. There are also correlations between pathological results in reaction time and tests of mood and intellectual functions, prolonged time before onset of sleep, longer waking periods and a reduction of REM sleep. Long term administration ofl-DOPA — alone or combined with a decarboxylase inhibitor — causes an increase in REM sleep as well as NREM sleep, corresponding with a marked improvement in the clinical symptoms of Parkinson's disease. This effect ofl-DOPA on sleep is attributed to regained mobility. There is also a specific influence ofl-DOPA on REM sleep, viz., prolongation of REM latency with an REM rebound in the second half of the night. The differentiation of a specific inhibitory effect ofl-DOPA on REM sleep and a nonspecific one on both stages of sleep in Parkinsonian patients may explain the diverse results of many authors. Thus, in patients without motor disability, general effects on the course of sleep with an increase of NREM sleep should not be expected. In such cases the inhibitory effect ofl-DOPA on REM sleep is to be seen, especially when administered just before the onset of sleep. Likewise there was no effect on NREM sleep after a single dose ofl-DOPA in patients in whom there was not already, definite improvement in the clinical signs of parkinsonism, but rather, an inhibitory influence on REM sleep occurred.

Notes: Summary 20 patients with endometrial carcinoma were given an injection of an average dose of 1000 mg 17-hydroxynorprogesteroncapronat (Depostat) within fourteen days after a first abrasion. The number of mitosis was determined by microscopical examination of 100 visual fields of the material obtained by abrasion immediately before and after this treatment. In 50% of the cases the number of mitosis showed a decrease by more than 50%, in 5 cases by more than 80%. The average decrease of the number of mitosis was 50% and therefore identical with earler results obtained by using nortestosterone preparations. The differentiated carcinomas showed a larger decrease than the less differentiated carcinomas.—In the control group of 25 endometrial carcinomas a small increase of the number of mitosis by 8.3% was found in the second abrasion.—A gestagen-application in cases of endometrial carcinoma appears reasonable particularly in the progressive stadium but also in combination with the primary treatment pre- and post-operatively.
