    Notes: We show that covariant field theory for sections of π : E→M lifts in a natural way to the bundle of vertically adapted linear frames LπE. Our analysis is based on the fact that LπE is a principal fiber bundle over the bundle of 1-jets J1π. On LπE the canonical soldering 1-forms play the role of the contact structure of J1π. A lifted Lagrangian L: LπE→R is used to construct modified soldering 1-forms, which we refer to as the Cartan–Hamilton–Poincaré 1-forms. These 1-forms on LπE pass to the quotient to define the standard Cartan–Hamilton–Poincaré m-form on J1π. We derive generalized Hamilton–Jacobi and Hamilton equations on LπE, and show that the Hamilton–Jacobi and canonical equations of Carathéodory–Rund and de Donder–Weyl are obtained as special cases. © 2000 American Institute of Physics.

    Notes: Classical Mendelian experiments were conducted to determine the genetics and inheritance of quinclorac and acetolactate synthase (ALS)-inhibitor resistance in a biotype of Galium spurium. Plants were screened with the formulated product of either quinclorac or the ALS-inhibitor, thifensulfuron, at the field dose of 125 or 6 g active ingredient (a.i.) ha−1 respectively. Segregation in the F2 generation indicated that quinclorac resistance was a single, recessive nuclear trait, based on a 1 : 3 segregation ratio [resistant : susceptible (R : S)]. Resistance to ALS inhibitors was due to a single, dominant nuclear trait, segregating in the F2 generation in a 3 : 1 ratio (R : S). The genetic models were confirmed by herbicide screens of F1 and backcrosses between the F1 and the S parent. F2 plants that survived quinclorac treatment set seed and the resulting F3 progeny were screened with either herbicide. Quinclorac-treated F3 plants segregated in a 1 : 0 ratio (R : S), hence F2 progenitors were homozygous for quinclorac resistance. In contrast, F3 progeny segregated into three ratios: 1 : 0, 3 : 1 and 0 : 1 (R : S) in response to ALS-inhibitor treatment. This segregation pattern indicates that their F2 parents were either homozygous or heterozygous for ALS-inhibitor resistance. Therefore, there were clearly two distinct resistance mechanisms encoded by two genes that were not tightly linked as demonstrated by segregation patterns of the F3.

    Notes: Abstract  The effects of temporal and spatial fluctuations in moisture on the microfungal community were studied in birch litter plus pine humus microcosms over 6 months. Two treatments were used: a uniform treatment in which moisture was maintained at the original moisture content throughout the profile over the course of the experiment; and a fluctuating moisture treatment in which the moisture content of the whole microcosm fluctuated weekly around the original moisture content. At 6 months, fungal species richness was higher and diversity was lower under moisture fluctuations than under uniform moisture. The number of fungal isolates and proportional diversity was significantly higher in the bottom layer in the fluctuating moisture treatment. The fluctuating moisture treatment and the bottom layer correlated significantly with the first PCA axis, which accounted for 66% of the variation in the fungal species data. The experiment suggests that even without drying, fluctuations in moisture can affect fungal community structure.

    Notes: Abstract Immature maize (Zea mays L.) embryos were treated with aflatoxin B1 concentrations, ranging from 0.1 μg ml−1 to 25 μg ml−1. Below 5 μg ml−1 aflatoxin B1, root and shoot elongation was not significantly inhibited. Ultrastructurally, root tip cells showed little deterioration, except a possible diffused clearing in mitochondria and plastids. As the toxin concentration was increased above 5 μgml−1, shoot, and particularly root elongation, was progressively inhibited. Associated with this, there was an apparent decrease in the ribosome population. Furthermore, membranes, particularly the vacuolar membrane, became abnormal and vacuolar distension occurred. At 20 and 25 μg ml−1, these effects were exacerbated, and mitochondria and plastid structure was disrupted. At these concentrations, there was evidence of a disruption in lipid metabolism. The results are discussed in the context of known aflatoxin effects on cellular control mechanisms and ultrastructure in animal systems.

    Notes: Abstract The effects of aflatoxin B1, (0.5–25 µg ml−1) on in vitro root and shoot development in young tobacco explants were investigated. Despite an initial apparent stimulatory effect on most measured parameters at 0.5 µg ml−1 AFB1, the number of leaves, root and leaf mass per plantlet were progressively inhibited with increasing AFB1 concentration. The number of explants developing roots was reduced to 34% at the highest (25 µg ml−1) AFB1 concentration, following 3 weeks exposure to the toxin. Leaf chlorophyll content at this toxin concentration was significantly lower than that measured for control plantlets. Thin layer chromatography confirmed the absorption of AFB1 by the plantlets. Using immunocytochemical techniques, AFB1 was immunolocated predominantly in the vacuoles, the nucleus and the cytoplasm (possibly intravesicularly). The results are discussed in terms of this immunolocation within the cell.

    Notes: Abstract  A precise organization of contractile proteins is essential for contraction of heart muscle. Without a necessary stoichiometry of proteins, beating is not possible. Disruption of this organization can be seen in diseases such as familial hypertrophic cardiomyopathy and also in acquired diseases. In addition, isoform diversity may affect contractile properties in such functional adaptations as cardiac hypertrophy. The Mexican axolotl provides an uncommon model in which to examine specific proteins involved with myofibril formation in the heart. Cardiac mutant embryos lack organized myofibrils and have altered expression of contractile proteins. In order to replicate the disruption of myofibril formation seen in mutant hearts, we have developed procedures for the introduction of contractile protein antibodies into normal hearts. Oligonucleotides specific to axolotl tropomyosin isoforms (ATmC-1 and ATmC-3), were also successfully introduced into the normal hearts. The antisense ATmC-3 oligonucleotide disrupted myofibril formation and beating, while the sense strands did not. A fluorescein-tagged sense oligonucleotide clearly showed that the oligonucleotide is introduced within the cells of the intact hearts. In contrast, ATmC-1 anti-sense oligonucleotide did not cause a disruption of the myofibrillar organization. Specifically, tropomyosin expression can be disrupted in normal hearts with a lack of organized myofibrils. In a broader approach, these procedures for whole hearts are important for studying myofibril formation in normal hearts at the DNA, RNA, and/or protein levels and can complement the studies of the cardiac mutant phenotype. All of these tools taken together present a powerful approach to the elucidation of myofibrillogenesis and show that embryonic heart cells can incorporate a wide variety of molecules with cationic liposomes.

    Notes: Abstract Calli ofNicotiana tabacum (tobacco) were treated with two dose ranges of aflatoxin B1 (0.1–2.0 µg ml−1 - low does; 5–25 µg ml−1 aflatoxin B1). The ability of calli to recover following 3 weeks of toxin exposure was also investigated. The I50 (50% inhibition) value for fresh mass accumulation was approximately 2 µg ml−1 AFB1. Fresh mass accumulation was significantly lower than the control value from 0.5 µg ml−1 AFB1. Following 3 weeks growth without a toxin source, the growth of calli up to and including 10 µg ml−1 AFB1, was significantly greater than control calli, indicating reversibility of the toxic effects. With increasing toxin concentration, chlorophyll content of callus was inhibited from 0.5 µg ml−1. Transfer to a toxin-free medium resulted in a degree of recovery (up to 0.5 µg ml−1). In the dose range 5–25 µg ml−1, the levels of chlorophyll were drastically reduced, with no recovery following AFB1 removal. Electron microscopy revealed a disruption of chloroplast structure as an early deteriorative event in AFB1 exposure of callus cells. Protein levels were less sensitive, with inhibition manifested only in the high dose range. Shoot development occurred at all concentrations, but was significantly inhibited from 5 µg ml−1 AFB1. Recovery following toxin removal was minimal at these higher AFB1 concentrations. The number of necrotic calli increased progressively from 5 µg ml−1 as toxin levels increased.
