    Notes: Abstract Although triacylglycerol stores play the critical role in an organism's ability to withstand fuel deprivation and are strongly associated with such disorders as diabetes, obesity, and atherosclerotic heart disease, information concerning the enzymes of triacylglycerol synthesis, their regulation by hormones, nutrients, and physiological conditions, their mechanisms of action, and the roles of specific isoforms has been limited by a lack of cloned cDNAs and purified proteins. Fortunately, molecular tools for several key enzymes in the synthetic pathway are becoming available. This review summarizes recent studies of these enzymes, their regulation under varying physiological conditions, their purported roles in synthesis of triacylglycerol and related glycerolipids, the possible functions of different isoenzymes, and the evidence for specialized cellular pools of triacylglycerol and glycerolipid intermediates.

    Notes: The response of the antioxidative systems of leaf cell mitochondria and peroxisomes of the cultivated tomato Lycopersicon esculentum (Lem) and its wild salt-tolerant related species Lycopersicon pennellii (Lpa) to NaCl 100 mM stress was investigated. Salt-dependent oxidative stress was evident in Lem mitochondria as indicated by their raised levels of lipid peroxidation and H2O2 content whereas their reduced ascorbate and reduced glutathione contents decreased. Concomitantly, SOD activity decreased whereas APX and GPX activities remained at control level. In contrast, the mitochondria of salt-treated Lpa did not exhibit salt-induced oxidative stress. In their case salinity induced an increase in the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione-dependent peroxidase (GPX). Lpa peroxisomes exhibited increased SOD, APX, MDHAR and catalase activity and their lipid peroxidation and H2O2 levels were not affected by the salt treatment. The activities of all these enzymes remained at control level in peroxisomes of salt-treated Lem plants. The salt-induced increase in the antioxidant enzyme activities in the Lpa plants conferred cross-tolerance towards enhanced mitochondrial and peroxisomal reactive oxygen species production imposed by salicylhydroxamic acid (SHAM) and 3-amino-1,2,4-triazole (3-AT), respectively.

    Notes: Mitochondria require robust antioxidant defences to prevent lipid peroxidation and to protect tricarboxylic acid cycle enzymes from oxidative damage. Mitochondria from wild, salt-tolerant tomato, Lycopersicon pennellii (Lpa) did not exhibit lipid peroxidation in response to high salinity (100 mm NaCl), whereas those isolated from cultivated tomato, L. esculentum (Lem), accumulated malondialdehyde. The activity, intraorganellar distribution and salt response of mitochondrial ascorbate peroxidase (mAPX) differed dramatically in the two species. In Lem mitochondria, the majority (84%) of mAPX was associated with membranes, being located either on the inner membrane, facing the intermembrane space, or on the outer membrane. Total mAPX activity did not increase substantially in response to salt, although the proportion of matrix APX increased. In contrast, 61% of Lpa mAPX activity was soluble in the matrix, the remainder being bound to the matrix face of the inner membrane. Salt treatment increased the activity of all mAPX isoforms in Lpa, without altering their intramitochondrial distribution. The membrane-bound isoforms were detected in mitochondria of both species by western blotting and found to be induced by salt in Lpa. These observations suggest that matrix-associated APX isoforms could act in concert with other mitochondrial antioxidants to protect against salt-induced oxidative stress.

    Notes: Composite resin bite-raising splints were fabricated on both sides of the upper jaw in rats. EMG activity of the deep masseter muscle was monitored continuously for 24 h before and up to 4 weeks after treatment. Together with the EMG activity, measurements of the neuromuscular junction area and muscle fibre histochemistry were made. EMG activity showed great variability immediately after splint fabrication. Some of the animals showed increased EMG activity, while others exhibited less activity 48 h after splint application. Overall, the fluctuation lasted for about 4 days, and after approximately 7 days all the animals exhibited decreased EMG activity, followed by a gradual increase in activity towards baseline levels. The neuromuscular junction territory increased significantly 5–10 days after splint application. Histochemical characterization of the deep masseter muscle revealed that it contained the three main types of muscle fibre: slow oxidative (SO), fast-oxidative-glycolytic (FOG) and fast glycolytic (FG). The frequency distribution of fibre types did not change after bite-raising splint treatment. These findings indicated that splint treatment in rats is effective in reducing EMG activity, while the muscle type characteristics remain stable. The effect of lower activity on the motor end-plates suggested that the neuromuscular junctions are quite sensitive to functional changes.

    Notes: Transport processes in plasticized PVC membranes containing the H+ chromoionophore ETH 5294 and additional tetraphenylborate-type mobile sites were analyzed by employing the potential-step method. Using chronoamperometry, impedance spectroscopy, and UV/VIS absorbance measurements, it is shown that the ionophore behaves as a carrier for protons, when the membranes are immersed between two aqueous solutions. It was possible to document the changes in the concentration profiles of both free ionophores and ionic species in some of the membranes. There is no evidence of proton hopping between the ionophores.

    Notes: The fabrication and electrochemical characterization of a variety of self-supported thermorevrsible organic electrolyte gels prepared using l,3:2,4-dibenzylidene sorbitol (DBS) as a gelling agent are reported. Impedance analysis and cyclic voltammetry were used to examine these gel systems, which exhibit ionic conductivities similar to corresponding liquid electrolyte solutions.
