    Notizen: Summary Phenotypic variation for parameters of seed yield were examined in 57 buffalo gourd (Cucurbita foetidissima HBK.) plants cultivated as annuals. Among the 52 individuals which bore fruit, seed weight/plant was highly variable (cv=106%); the majority of plants exhibited seed yields below that of the mean (431 g/plant). Values of seed weight/plant were more highly influenced by fruit/plant (r=+0.81) than by seed weight/fruit (r=+0.19). A weak relationship existed between fruit/plant and seed weight/fruit (r=−0.28), suggesting the possibility of their simultaneous improvement through selection. Seed weight/fruit was positively associated with fruit size, seed/fruit and 100-seed weight; 4 carpellate fruit displayed significantly greater seed weight/fruit and seed/fruit than 3 carpellate fruit.

    Notizen: Abstract Three specific enzymes are involved in the cerebral synthesis of 7,8-dihydrobiopterin from GTP. These were isolated, purified, and characterized. The first enzyme, also catalyzing the rate-limiting step, is GTP-cyclohydrolase A-I or Mg2+-dependent A-II, which hydrolyze the GTP to the specific product 2-amino-6-(5-triphosphoribosyl)-amino-5-or-6-formamido-6-hydroxypyrimidine (FPyd-P3). FPyd-P3 is cyclized by a synthetase tod-erythro-7,8-dihydroneopterintriphosphate (NPTH2-P3). The new enzyme,d-erythro-7,8-dihydroneopterintriphosphate synthetase (enzyme B) is a basic protein of 9177 daltons containing three free SH groups, isoleucyl-seryl- as N- and valyl-glutamyl- as C-terminals. This enzyme of 69 amino acid residues from rat and 68 residues (one less aspartic acid) from guinea pig brain contains no hydroxyproline, methionine, or tryptophan. The enzyme from rat brain will gradually convert its product NPTH2-P3 to BH2, whereas the enzyme from guinea pig brain lacks this property. 2,4-amino-6-hydroxypyrimidine and dFPyd-P3 are effective inhibitors of this enzyme. The synthesis of BH2 from NPTH2-P3, but not from 7,8-dihydroneopterin, is catalyzed byl-erythro-7,8-dihydrobiopterin synthetase (enzyme C), which was purified to electrophoretic purity. This enzyme does not require pyridine nucleotides or Mg2+ for its catalysis.

    Notizen: Abstract The results of a three-dimensional photoelastic analysis of a propellant-grain model are compared with the results obtained from a three-dimensional finite-element analysis of the same model. The loading considered was differential thermal expansion between the case and the grain model. The model analyzed was the Stage I Minuteman propellant grain which consists of a six-point-star configuration with a head-end web. Stresses and strains determined from the two analyses in the star valley are compared. The excellent correlation between the stresses determined from the photoelastic analysis and the stresses calculated by the three-dimensional finite-element computer program established the validity of the computer program.

    Notizen: The suitability of using annually grown, carrot-sized buffalo gourd (Cucurbita foetidissima) roots as a feedstock for alcoholic fermentation was explored. Roots grown in 1982 and 1983 were slurried, dextrinized and saccharified using Takatherm™ and Diazyme™ (commercial enzymes manufactured by Miles Laboratories), and fermented by the action of Saccharomyces cerevisiae. These processes were monitored in detail and results were compared with those displayed by controls formulated using potato tubers. The preparation of gourd root slurries with suitable viscosity characteristics for enzymatic digestion required the addition of water (at least 50% by weight) which reduced the proportion of fermentable sugars in the resulting saccharified suspensions. The resulting slurries were well-suited to enzymatic conversion of starch to sugar. Estimates of enzymatic efficiency in gourd root suspensions did not suggest the presence of naturally occurring amylase or glucosidase inhibitors in these plant materials. Saccharified gourd root mashes supported yeast growth well and produced ethanol yields at 82.2-86.5% of the theoretically maximum efficiency.
