Notes: Abstract: α-d-Mannosidase (EC 3.2.1.24.) was purified to homogeneity from adult rat brain. The enzyme, of apparent molecular weight 397,000, appears to be formed of subunits of molecular weight 120,000 made of two protomers (62,000) bound by disulfide bridges. Isoelectric focusing gives two bands, of pi 5.40 and 5.15. Both isoenzymes seem to have the same pH curve (a small peak of activity at pH 4.5 and a maximum of activity around pH 6.0). These two isoenzymes are immunologically related.

Notes: Crystalline SiGe alloy sheet polymers were topotactically prepared from epitaxially grown calcium germanosilicide Ca(Si1−xGex)2 precursor films in the whole composition range. These polygermanosilynes are found to be a well-defined mixture of the known siloxene and polygermyne sheet polymers with the OH groups exclusively bonded to silicon. The optical properties determined by photoluminescence and optical reflection measurements identify the mixed SiGe sheet polymers as direct semiconductors with efficient luminescence tunable in the energy range between 2.4 and 1.3 eV. © 2001 American Institute of Physics.

Notes: The level of glutathione (GSH) in plants is important in defence reactions against biotic and abiotic stresses and can place considerable demand of the sulphur assimilation pathway. Enzymes involved in sulphur assimilation and GSH metabolism are not evenly distributed between different subcellular compartments or between different cell types in leaves or roots; however, there is little information on the effect that such asymmetries have on the actual GSH concentration in each compartment or cell type. In the present study in situ labelling with monochlorobimane (MCB) in combination with confocal laser scanning microscopy was used to quantify GSH in each of the main cell types of poplar leaves from fluorescence of the GSB conjugate formed. Comparison of results from the in situ approach with total GSH levels measured in vitro by high-performance liquid chromatography suggested that only the cytosolic GSH pool was labelled using this approach. This suggests that an appropriate GST was not present within the chloroplasts to catalyse the conjugation reaction and that chloroplastic GSH does not rapidly exchange with the cytoplasmic pool under the conditions of the assay. Cytosolic GSH levels were between 0.2 and 0.3 mm for both photosynthetic and non-photosynthetic (epidermal) cell types in wild-type poplar leaves. Cytosolic levels increased by around two-fold in transgenic poplars over-expressing bacterial γ-glutamylsynthetase (γ-ECS) in the cytosol of all cell types, but there was no concomitant increase in the chloroplastic GSH pool.

Notes: A method of measuring the difference between the lattice parameter of a region of an unknown sample and that of a region of a standard reference crystal to a sensitivity of 1 part in 109 is presented. Problems inherent in multiple-beam arrangements due to sample strains and non-uniformity have been overcome by the use of a new double-source arrangement in which the two X-ray beams sample the same spot on a crystal under study. Ways of identifying and preventing errors from significant mechanical and thermal effects arising in the sensitivity region explored are indicated.

Notes: Abstract  One of the main advantages of digital imaging is the possibility of altering display options for improved image interpretation. The aim of the present study was to evaluate the subjective image quality of direct digital panoramic images and compare the results with those obtained from conventional images. Furthermore, the effect of various filter settings on image interpretation was assessed. Panoramic images were obtained with three different types of panoramic equipment (one direct digital and two conventional units) from three groups of 54 patients with a natural dentition in all quadrants. The first series of panoramic images consisted of 54 unprocessed digital images; conventional film images (n=108) comprised the second and third series. A final series consisted of the digital images treated with three different filters (”smoothening,””sharpening,” and ”contrast enhancement”). All images were scored randomly by four experts in oral radiology on a 4-point rating scale. The results showed a statistically significant difference in scorings between the conventional and digital panoramic units. The main reason for poor image quality appeared to be a combination of blurring and overlapping in the panoramic image. The premolar region in the upper jaw was the region where most additional radiographs were needed.

Notes: Abstract  Oxidative protection of mayonnaises with 16% fish oil was studied during cold storage (5  °C) after supplementation with different tocopherol systems: the ternary antioxidant system ascorbic acid, lecithin and tocopherol (A/L/T), and two commercial mixtures, an oil-soluble (Toco 70) preparation and a water-soluble (Grindox 1032) preparation. The physical structure of the fish-oil-enriched mayonnaise was manipulated by adding extra emulsifier (Panodan TR) with the purpose of investigating whether or not this affected the antioxidative activity of the tocopherol mixtures. A number of different analytical techniques HPLC (high-performance liquid chromatography, gas chromatography mass spectrometry (GC-MS), sensory analysis, confocal laser scanning microscopy and rheological measurements) were employed to elucidate the chemical, sensory, structural and rheological aspects of the oxidation process. Discriminant partial least squares regression was used to analyse the data obtained. The three tocopherol preparations not only affected the oxidative stability of the mayonnaises differently, they also influenced the rheological and structural properties of the mayonnaises in different ways. The rheological and structural properties of the mayonnaise were also affected by the addition of extra emulsifier, but this did not influence the formation of fishy and rancid off-flavours. Addition of the A/L/T system caused the immediate formation of distinct fishy and rancid off-flavours in the fresh mayonnaises. The volatile compounds trans-2-heptenal, 4-octen-3-one, 1-octen-3-ol, trans,cis-2,4-heptadienal, trans,trans-2,4-heptadienal, trans-2-octenal, nonanal and trans,cis-2,6-nonadienal were thought to contribute to the fishy and rancid flavours. Addition of Toco 70 did not affect the sensory perception of mayonnaise nor the development of volatile off-flavour compounds as evaluated by GC-MS, but the peroxide values were slightly increased in mayonnaise containing Toco 70 as compared to the other mayonnaises. Mayonnaise with Grindox 1032 seemed to have fewer fishy and rancid off-flavours than mayonnaises without antioxidant. This flavour-protective effect of Grindox 1032 was correlated to an increase in the size of the droplet diameter of mayonnaises supplemented with Grindox 1032.

Description / Table of Contents: Abstract  In a retrospective study we examined whether it is possible to achieve better results in carotid artery surgery with a new operative technique called ”no-touch-isolation technique” in combination with intraoperative angiography than with conventional operative techniques. Patients and methods: In 1997 and 1998, 80 patients who were operated on using this new operative technique were compared with a group of 144 patients who had undergone conventional carotid endarterectomy. The new aspect of the no-touch-isolation technique is that the whole operation strategy aims to avoid possible intraoperative and postoperative embolism. At first, the internal carotid artery is exposed and clamped distally to the stenotic plaque very atraumatically. The operation was then continued without an intraluminal shunt. At the end of the operation the result was checked by means of an intraoperative angiography. If any suggestion of a possible embolism, such as tears of the intima, were seen, an intraoperative revision was carried out until all thrombembolic material was removed. Results: The two groups were not different regarding age, sex, symptoms, degree of stenosis and risk factors nor did any significant differences exist in the operative technique used for endarterectomy (with or without patch, eversions-TEA, etc.). The group operated on with the no-touch-isolation technique had significantly longer operation- times (110 min vs 90 min) and also significantly longer clamping times (38.7 min vs 28.2 min) than the group operated on using conventional technique. The postoperative results, however, were still better. There were significantly fewer neurological deficits (4.1% vs 9.3%). Conclusion: The no-touch-isolation technique in combination with intraoperative angiography is a very safe method of treatment in patients with symptomatic stenosis of the internal carotid artery with a more than 70% degree of stenosis.

Notes: Summary Methodic numerical experiments with a non-trivial 10-level prediction model in a zonal plane are carried out in order to demonstrate the efficiency of a normal mode initialization method for an one-way nested situation. The results demonstrated in the mode space and the physical space support the approach to be promising, particularly in the early stage of the forecast when the vertical motion field is developing.

Notes: Summary A physiologically based steady-state model of whole leaf photosynthesis (WHOLEPHOT) is used to describe net photosynthesis daily time courses in Prunus armeniaca. Net photosynthesis rates are calculated in response to incident light intensity, leaf temperature, air carbon dioxide concentration, and leaf diffusion resistance measured at five minute intervals. The steady-state calculations closely approximate the observed net photosynthesis rates for a broad range of weather conditions and leaf stomatal behavior.
