    Differential regulation of ACC synthase genes in cold-dependent and -independent ripening in pear fruit (2004)

    Notes: Late pear cultivars such as Passe-Crassane (PC) require a long chilling treatment before they are capable of ripening. Early cultivars such as Old-Home (OH) have no cold prerequisite. The regulation of 1-aminocyclopropane-1-carboxylic acid synthase (ACS) genes was studied in OH, PC and in OH × PC hybrids in order to determine the role of this gene family in the cold requirement. Of the seven Pc-ACS cDNAs isolated, four (Pc-ACS1a/b and Pc-ACS2a/b) showed differential expression associated with the cold requirement. Pc-ACS1a transcripts accumulated throughout the cold treatment and, with Pc-ACS2a, during ripening of cold-dependent cultivars. Pc-ACS1b and Pc-ACS2b were detected only during ripening of cold-independent genotypes. Furthermore, Pc-ACS2a transcript accumulation was negatively regulated by ethylene, whereas Pc-ACS2b was positively regulated by the hormone. Pc-ACS3, 4 and 5 transcript accumulation was similar in all genotypes. Genetic analyses of OH, PC, and 22 OH × PC progenies demonstrated that late, cold-dependent cultivars were homozygous for Pc-ACS1a and 2a whereas early, cold-independent cultivars were heterozygous for Pc-ACS1(a/b) and homozygous for Pc-ACS2b. A model is presented in which differences in Pc-ACS alleles and gene expression between cold- and non-cold-requiring pears are critical in determining the ripening behaviour of the cultivars.

    Notes: Physiological characterization of ethylene-suppressed Cantaloupe Charentais melons (Cucumis melo var. cantalupensis Naud cv. Védrantais) revealed that some ripening-associated events, like degreening of the rind and cell separation in the peduncular abscission zone, are totally dependent on ethylene. By contrast, some other ripening events, like softening and membrane deterioration, depend only partially on ethylene and display some ethylene-independent components. Application of increasing levels of exogenous ethylene on these antisense 1-aminocyclopropane-1-carboxylic acid oxidase fruits enabled the determination of the gradual sensitivity of various ripening pathways to the hormone. The threshold level of ethylene capable of physiological activity varied from 1 ppm for degreening of the rind to 2.5 ppm for softening, membrane deterioration and cell separation in the peduncular abscission zone. Up to a saturating dose of 5 ppm, the extent of rind degreening was proportionally related to the level of applied ethylene. The saturating levels of ethylene for flesh softening (2.5 ppm) and for membrane deterioration and cell separation in the peduncular abscission zone (5 ppm) were much lower than the internal ethylene found at the climacteric peak of wild-type fruit (over 100 ppm). The cessation of ethylene treatment resulted in a complete arrest of the rind degreening and peduncular cell separation indicating that both ripening pathways are completely dependent on ethylene. On the contrary, softening and membrane deterioration, though significantly slowed upon removal of ethylene treatment, continued to proceed in the absence of the hormone, thereby unmasking the ethylene-independent component of softening and membrane deterioration. The presence of ethylene-independent components in the regulation of individual pathways indicates that the ripening of climacteric fruit involves a substantial portion of non-climacteric regulation.

    Notes: To better understand the possible role of deoxygenation of haemoglobin in the pathogenesis of exophthalmos in the West Australian dhufish, Glaucosoma hebraicum Richardson, some oxygen transport properties of haemoglobin from this species were determined and compared with haemoglobin from black bream, Acanthopagrus butcheri (Munro), snapper, Pagrus auratus (Bloch & Schneider) and King George whiting, Sillaginodes punctata (Cuvier). Dhufish had a single haemoglobin with a pronounced Root effect. Snapper and black bream blood had six and five types of haemoglobin, respectively. The magnitude of the Root effect of haemoglobins in the haemolysate was demonstrated by comparing oxygenation and total deoxygenation at pH 8 with relative deoxygenation at pH 6.9. Dhufish haemoglobin demonstrated a large Root effect, with the Root effect of King George whiting, snapper and black bream being of lesser magnitude. Deoxygenation of all haemolysates was more pronounced in the presence of adenosine triphosphate (ATP). Seasonal changes in haematological parameters and ATP content of dhufish blood were not evident. The Root effect is discussed in the context of its possible role in oxygen supply to the retina and adaptation to the fish's habitat and the environment.

    Notes: The West Australian dhufish, Glaucosoma hebraicum Richardson (family Glaucosomatidae) is a potentially valuable aquaculture species, but spontaneous exophthalmos is common in freshly caught and cultured dhufish. To investigate the epidemiology and pathogenesis of exophthalmos in tanks of wild-caught and captive-bred dhufish, records of culture conditions, the prevalence of lesions and pathological examinations were used to study the disease and associated environmental and management factors. Naturally occurring cases of exophthalmos that were examined were mostly unilateral and occurred more frequently in summer months during periods of increasing water temperature. In 15 affected eyes that were examined histologically gas bubbles and haemorrhages were consistently present in the choroid. In some, gas bubbles and haemorrhage were also present in retrobulbar tissues associated with perforation of the sclera. Oxygen concentrations were measured in 12 exophthalmic eyes and concentrations of 50–73% were recorded in gas bubbles in the anterior chamber of three of these that were acute cases. Very low oxygen tensions were recorded at the retinal–vitreal junction of four eyes with retrobulbar haemorrhages indicating that there may have been disruption of arterial blood supply to the choroid in chronic lesions or with perforation of the sclera. The results of experiments to determine the significance of exercise, high water temperature, sudden increases in light intensity, fish handling or tank cleaning indicate that exercise and/or high water temperature (25.5 °C) may be important risk factors for the development of exophthalmos in dhufish. Possible mechanisms for the formation of gas bubbles and haemorrhage in the choroid are discussed.
