    Notes: In contrast to the multiaperture–multigrid concept of the International Thermonuclear Experimental Reactor (ITER) injector reference design in which D− acceleration to 40 A, 1 MeV is achieved through seven grids containing 1280 apertures each, the European SINGAP (single gap–single aperture) concept proposes acceleration of 1280 preaccelerated (20–50 keV) beamlets to 1 MeV in a single step. During acceleration the beamlets are merged into 16 groups of 80 beamlets. These 16 so-called hyperbeamlets then emerge from an exit grid containing only 16 very large apertures. This concept is expected to reduce cost and increase the tolerance to mechanical errors and stray magnetic fields. The SINGAP physics is described here. The SINGAP beam optics is calculated to provide good beam transmission (86%) to the ITER. © 2002 American Institute of Physics.

    Notes: Since heroin is delivered to a selected group of drug addicts under supervision of nurses in the Netherlands, we reported about several nurses who presented with work-related eczema and positive patch tests to heroin. To investigate the prevalence of heroin contact allergy among all workers in this heroin delivery project, a study was started using questionnaires. Altogether 31 nurses reported work-related complaints out of 100 who returned questionnaires. Besides reports of eczema, mainly of eyelids (probably airborne) and hands, there were mucosal and respiratory complaints. Patch tests were performed in 25 nurses with complaints; in 9 of them a heroin contact allergy could be confirmed. In 6 out of these 9 nurses this was combined with mucosal or respiratory complaints. There were also 6 nurses with mucosal or respiratory complaints without a contact allergy. Contact dermatitis from opioids, such as morphine and codeine, has been documented among opioid industry workers, nurses, doctors, pharmacists, and in patients. In conclusion heroin appears to be a potent contact allergen, causing contact dermatitis. Mucosal and respiratory complaints however, cannot be explained by this contact allergy; they might be caused by a type-1-allergy to heroin, or by a direct histamine liberating effect. Opioids are known histamine liberators causing urticaria, rhinitis and anaphylactoid reactions; therefore intracutaneous tests with heroin are unreliable. In an ongoing research project it will be attempted to detect specific IgE to heroin in the 12 workers with mucosal or respiratory complaints; within the next few months results will be available.

    Notes: The target performance for the KAMABOKO ion source on the MANTIS test bed in Cadarache is to accelerate a beam of D− with a current density of 200 A/m2 and 〈1 extracted electron per accelerated D− ion, at an injected power ranging between 1 and 2 kW per liter of source volume, at a source pressure of 0.3 Pa. For ITER, a continuous neutral beam must be assured for pulse lengths of 500 s, but beams of up to 3600 s are also envisaged. During the last campaign, continuous beam pulses of duration up to 1000 s were demonstrated both in hydrogen and in deuterium. In this article, the source performance, the effect of the plasma grid temperature for long pulse operation, and the limits of the present experimental setup are described. Additionally, the effect of changing the strength of the magnetic filter in the ion source on the extracted ion and electron currents and the beam transmission is reported. © 2002 American Institute of Physics.

    Notes: The oxygen adsorption on the unreconstructed Rh(110) surface has been studied by scanning tunnelling microscopy (STM), by imaging the evolution of the surface reconstruction in situ during oxygen uptake at 390 K. The dynamics terminate with the formation of a strained (10×2)–O structure with high local oxygen coverage. Initially, kink atoms are detached from the steps and diffuse over the surface, most probably as Rh–O units, as inferred by comparison to x-ray photoelectron spectroscopy (XPS) data, until they condense in zigzag structures characteristic for the (2×2)p2mg–O structure (0.5 ML). At oxygen doses above 0.12 L, missing rows begin to be dug along the [11¯0] direction into the steps, detaching further Rh atoms. The reconstruction proceeds as a missing/added row reconstruction. With ongoing surface reconstruction, a second reconstruction mechanism of local nature sets in: Rh atoms start to be ejected out of the terraces and condense at the ends of the so formed troughs. Dosing further oxygen on the (2×2)p2mg–O structure leads to the formation of the strained (10×2)–O structure. The ejection of two of every 10 Rh atoms proceeds only if the Rh rows are not neighbored by troughs. © 2001 American Institute of Physics.

    Notes: Recent research has demonstrated a crucial role for autoantibodies in the pathogenesis of Sjögren's syndrome (SS)-like disease in the non-obese diabetic (NOD) mouse, but it remains to be determined which antibody species among all those present are directly related to the various aspects of pathology. To identify autoantigens in the NOD mouse system, we have taken the approach of using immunoglobulin (Ig)G purified from sera of NOD mice exhibiting SS-like symptoms to screen cDNA expression libraries derived from exocrine gland mRNA. Here we report the identification of a novel autoantigen, designated LGP10, expressed in lacrimal and submandibular glands. Autoantibodies to this protein are prominent in the sera of NOD mice starting at 11–12 weeks of age, but not in control nonautoimmune mice. LGP10 has no known function, but bears similarities to various other proteins produced by epithelia. Interestingly, all of these similar proteins have been linked to immunosuppression and/or steroid binding, both processes that could have a significant impact on pathological features of SS.

    Notes: Background The existence of a third B7-1/B7-2 receptor was postulated in a recent study using a novel mouse strain lacking both CD28 and CTLA4 (CD28/CTLA4−/−).Objective In the present study, it was investigated if T cell co-stimulation via the putative B7-1/B7-2 receptor plays a role in the induction of Th2-mediated asthma manifestations in mice.Methods BALB/c wild-type, CD28/CTLA4−/− and B7-1/B7-2−/− mice were sensitized and aerosol challenged with ovalbumin (OVA).Results At 24 h after the last aerosol, wild-type mice showed airway hyper-responsiveness in vivo and up-regulated levels of serum OVA-specific IgE compared with the situation shortly before OVA challenge. In addition, eosinophil numbers and IL-5 levels in the broncho-alveolar lavage fluid and Th2 cytokine production by lung cells upon OVA re-stimulation in vitro were observed. In agreement with an earlier study, we failed to induce any of the asthma manifestations in B7-1/B7-2−/− mice. Importantly, also CD28/CTLA4−/− mice showed no asthma manifestations upon OVA sensitization and challenge.Conclusion These data clearly demonstrate that T cell co-stimulation via the putative B7-1/B7-2 receptor appears to have no role in the induction of Th2-mediated asthma manifestations in this murine model and, conversely, that CD28 signalling is crucial.

    Notes: The structural and optical properties of the gel-forming photochromic switch 1,2-bis(2’-methyl-5’-{{((R) phenylethyl)amino}carbonyl}thienyl-3’-yl)cyclopentene are studied by means of the linear absorption and holographic grating techniques. The use of diffractive optics enables recording of holographic gratings with high long-term phase stability. The diffraction efficiency of the recorded holographic gratings approach values of 30% for low writing beam energies when diffusion is the rate determining factor. At higher writing pulse energies the competition between the diffusion and photodecomposition processes causes lower diffraction efficiencies. At irradiation doses above 10 mJ the spatial profile of the recorded gratings is strongly influenced by saturation effects. Because of the well-determined grating profile the holographic grating technique is potentially applicable for the quantitative characterisation of the diffusion process in photochromic gels
