    Notes: The electronic structures of mononuclear Fe–S complexes with a FeIIS4 core and of binuclear Fe–Mo–S complexes containing the FeS2Mo core have been calculated by a semiempirical molecular orbital method (iterative extended Hückel theory), followed by a spin-orbit coupling calculation on the five highest occupied iron-like molecular orbitals. Fine structure and hyperfine structure tensors and parameters (g, D, E, A, and electric field gradient) have been calculated and compared with data from spin-Hamiltonian analysis of Mössbauer measurements. For the mononuclear complex anions [Fe(SPh)4]2− and [Fe(dts)2]2− it was found that Vzˆzˆ is negative, D positive, and that the magnetic anisotropy places the preferred direction of the hyperfine magnetic field perpendicular to the Vzˆzˆ direction in agreement with spin-Hamiltonian results. The similarity of parameters of [Fe(SPh)4]2− and reduced rubredoxin (Rdred) confirms the suggestion that this anion has a ground electronic state practically identical to Rdred. The complex anion [Fe(dts)2]2− shows smaller anisotropy, and due to the fact that the orbital ground state is energetically not well separated from higher states in this case a strong temperature dependence of the quadrupole splitting is observed. For the binuclear complex anions [(SPh)2FeS2MoS2]2−, [S5FeS2MoS2]2−, and [Cl2FeS2MoS2]2− it was found that d is negative and Vzˆzˆ is positive. A specific feature of these binuclear Fe–Mo–S complexes is that Vzˆzˆ is directed perpendicular to the Fe–Mo line. (This theoretical result is confirmed by single crystal Mössbauer studies on [Cl2FeS2MoS2]2−; see the following paper in this journal.) The preferred direction of the magnetic hyperfine field is close to the Vzˆzˆ axis. The correlation of calculated values of ρ(0) and isomer shifts for mononuclear and binuclear compounds confirms the role of MoS2−4 as a charge withdrawing ligand.

    Notes: A single crystal of [(C6H5)4P][(C6H5CH2)–(CH3)3N][Cl2FeS2MoS2] has been investigated by Mössbauer spectroscopy with various orientations of the crystal with respect to the γ beam. From this investigation we derive that the main component of the electric field gradient tensor Vzˆzˆ is positive and oriented perpendicular to the Fe–Mo direction. This result was confirmed by additional single crystal measurements applying an external magnetic field, and by magnetically perturbed spectra of polycrystalline samples. The determination of the orientation of the electric field gradient tensor with respect to the molecular axes is a prerequisite for a decisive check of molecular orbital calculations on this type of binuclear bio-inorganic systems.     Notes: Secretoneurin is a recently described peptide derived by endoproteolytic processing from secretogranin II, previously named chromogranin C. In this study, we have investigated the distribution of secretoneurin-like immunoreactivity in the human hippocampus in controls and in Alzheimer's disease patients, and compared the staining pattern to that of calretinin. Secretoneurin-like immunoreactivity is present throughout the hippocampal formation. At the border of the dentate molecular layer and the granule cell layer, a band of dense secretoneurin immunostaining appeared. In this part, as in the area of the CA2 sector, the high density of secretoneurin-immunoreactivity coincided with calretinin-like immunoreactivity. The mossy fibre system displayed a moderate density of secretoneurin-immunoreactivity. In the entorhinal cortex, a particularly high density of secretoneurin-immunoreactivity was observed. The density of secretoneurin-like immunoreactivity was significantly reduced in the innermost part of the molecular layer and in the outer molecular layer of the dentate gyrus in Alzheimer's disease. For calretinin-like immunoreactivity, a less pronounced decrease was found in the innermost part of the molecular layer. About 40–60% of neuritic plaques were secretoneurin-immunopositive.This study shows that secretoneurin is distinctly distributed in the human hippocampus and that significant changes of secretoneurin-like immunoreactivity occur in Alzheimer's disease, reflecting synaptic loss.

    Notes: Abstract: The properties of the catecholamine-storing organelles from transplantable rat phaeochromocytoma and rat adrenal glands were compared by density gradient centrifugation. It was shown that tumour granules are more heterogeneous and less dense than adrenal granules. Both granule preparations can take up catecholamines and nucleotides by a process driven by an electrochemical proton gradient. Dopamine β-hydroxylase and glycoprotein III were analysed by immunological techniques. Glycoprotein III was shown to be a specific component of chromaffin granules. Tumour tissue (average weight 700 mg) contains amounts of these antigens comparable to those in 210 adrenals. The biosynthesis of granules in the tumour apparently occurs at a low rate, making turnover studies difficult. The transplantable rat phaeochromocytoma is very useful for studies on the uptake properties and the immunological characteristics of rat catecholamine storage granules because one tumour provides an amount of material that could otherwise be obtained only from a large number of adrenal glands.

    Notes: Abstract: A glycoprotein (s-GP III) was isolated from the soluble lysate of chromaffin granules by chromatography with immunoaffinity and lectin columns. An identical protein (m-GP III) was shown to be present in the granule membranes. The apparent molecular weight of these glycoproteins as determined by the electrophoresis system of Laemmli (1970) was 43,000 under reducing conditions. In the absence of mercaptoethanol they aggregated to dimers. Antisera were raised against both the soluble and the membrane-bound forms of this glycoprotein. With these antisera GP III was further characterized: Immunoreplicas were obtained after two-dimensional electrophoresis of soluble and membrane-bound proteins of chromaffin granules. GP III was identified as a protein with a rather broad pI (4.6-5.3), indicating microhetero-geneity. As shown by subcellular fractionation, m-GP HI is specifically confined to chromaffin granules. GP III can therefore be used as a marker for the membranes of these organelles. The soluble form is secreted from adrenal medulla during stimulation with carbamylcholine chloride. An immunologically identical antigen was detected in adeno- and neurohypophysis. The physiological function of GP III is still unknown. It does not demonstrate any of the enzymatic activities so far known to occur in chromaffin granules.

    Notes: An antiserum was raised against the peptide PE-11 whose sequence is present in the chromogranin B molecule. The antiserum reacts only with the free C-terminal end of this peptide. PE-11 immunoreactivity in brain was characterized by molecular size exclusion high performance liquid chromatography. Only the free peptide and a N-terminally elongated peptide were detected, indicating that proteolytic processing of chromogranin B in brain is quite extensive. In immunohistochemistry PE-11 immunoreactivity was found in varicosities, fibres and perikarya throughout the brain. Strong staining was detected in the shell sector of the nucleus accumbens, in the lateral septum, in subregions of the extended amygdala, in some areas of the hippocampus and of the hypothalamus, in the locus coeruleus, in the Purkinje cells of the cerebellum and in the dorsal horn of the spinal cord. Our results, which demonstrate significant processing of chromogranin B in brain and its widespread distribution, can be taken as an indication that chromogranin B represents a precursor of peptides with functional relevance for this organ.

    Notes: Abstract– The mechanisms leading to spatial segregation along optical habitat dimensions in closely related, co-occurring fishes were investigated. In two experimental series, six laboratory-reared cyprinid species were faced to six novel optical patterns. These were positioned either laterally, at the bottom, or at the water surface, in one of three compartments of a large tank. When a bottom pattern was presented, the primarily bottom-dwelling species Vimba elonguta and Abranzis brama chose the test compartment at high frequencies, indicating a significant preference. More surface-oriented species like Chalcalburnus chalcoides mento and Scardinius erythrophthalmus developed similar preferences in response to surface patterns. V. elongata, A. brama, and Rutilus rutilus showed significant preferences for a photograph of submerged plants. Also, V. elongata developed preferences for several optical patterns. These data confirm earlier studies on V. elongata habitat selection and foraging behaviour which suggest a highly exploratory lifestyle. S. erythrophthalmus and Leuciscus cephalus showed significant avoidances for laterally positioned optical patterns associated with reduced interindividual distances, indicating marked neophobic responses. The establishment of species-specific preferences or avoidances during the two experimental series was influenced by learning processes.

    Notes: The influence of temperature on the embryonic development of three European cyprinid fishes, Abramis brama, Chalcalburnus chalcoides mento and Vimba vimba, was studied with respect to hatching efficiency and the functional relationship with the duration of development. The functional dependance of the duration of development on temperature is well described by a power function with a correcting constant for temperature. Comparison with published data for other cyprinids shows that the response of embryos to temperature, together with the various temperature controlled responses of adult fishes, is a valuable aid for the ecological characterization of the different species.
