    Notes: An interferometer previously developed for measuring instantaneous film thickness of the order of 10 μm–1 mm [T. Ohyama, K. Endoh, A. Mikami, and Y. H. Mori, Rev. Sci. Instrum. 59, 2018 (1988)] has been improved with respect to both facility of handling and accuracy of thickness determination. The improved version of the interferometer uses an expanded laser beam focused aslant on the subject film. The rays reflected from the front and rear surfaces of the film form interference fringes on a screen, which are then recorded in the form of a single snapshot interferogram or as a set of serial interferograms to be geometrically analyzed. Experiments with thin glass plates as sample films verified the expected advantages of the new version over the original one. The improved version of the interferometer was also tested on a liquid film asymptotically thinning as it drained down a vertical plate. The test indicated that the accuracy and precision of the new improved version far exceed those of other transient film-thickness measuring techniques. © 1996 American Institute of Physics.

    Notes: Nondestructive analysis of water distribution and structural changes in cooked rice grains, Oryza sativa cv Koshihikari, was performed with Nuclear Magnetic Resonance (NMR) Micro imaging of protons (1H). Using a specially designed holder, high quality images of cooked rice grains were obtained, even when up to 10 grains were studied simultaneously. Internal hollows were observed in all examined cooked rice grains, and we propose a mechanism to explain their formation. The origin of these hollows was hypothesized to be cracks or fissures, and hollows resulted from sealing of such lacerations by gelatinized starch in the peripheral layer in combination with expansion of the grain during cooking.

    Notes: We previously reported that nuclear magnetic resonance (NMR) micro imaging detected the hollows in cooked rice but required a specialist to operate equipment. Convenient detection of hollows is useful in the characterization of cooked rice. X-ray imaging and light transmittance photography were studied as alternatives to NMR micro imaging. X-ray imaging detected about 50% of all hollows. Light transmittance photography detected almost all hollows in cooked rice (Oryza sativa cv. Koshihikari) effectively and rapidly, relatively quantifying hollow sizes and numbers.

    Notes: Calprotectin is a calcium binding protein produced by leukocytes, macrophages and epithelial cells, and its levels in several tissues increase during infections and in many inflamed areas, suggesting that it may be an indicator of inflammatory activity. Osteopontin is a prominent phosphorylated glycoprotein in bone matrix, having calcium binding capacity. Recently, it has been reported that calprotectin and osteopontin are present in urinary stones (pathological mineralized masses in the body), and that these proteins may be involved in their formation. Dental calculus formed by mineralization of dental plaque is an inflammatory factor which may contribute to periodontal disease. It contains many organic components involved in mineralization. We recently found osteopontin molecules in human dental calculus and suggested that the components of its matrix may be similar to those of urinary stones. In this study, we investigated the presence of calprotectin in human dental calculus by immunohistochemical and immunoblotting analyses using a specific antibody for calprotectin. After fixation and demineralization of dental calculi adhered to tooth roots, sections embedded in paraffin were immunoreacted with the antibody for calprotectin and positive immunostaining for calprotectin was observed. Dental calculus proteins were then extracted with EDTA and separated by electrophoresis on 15% polyacrylamide gels. By immunoblotting analysis, 3 or 4 bands were observed at 11, 14.5, 22–25, 28 or 36.5 kDa and these patterns corresponded to those of calprotectin subunits. When non-immune rabbit serum was used instead of calprotectin-specific antibody as a negative control, no immunoreactivity was observed. These findings indicate that calprotectin is associated not only with antibacterial action but also with calcium binding capacity during dental calculus formation.

    Description / Table of Contents: Zusammenfassung Der Einsatz und die Kombination von GC, GC/MS und Headspace-Variante ermöglicht die Routine-Analyse von Tetrahydrofuran (THF) in biologischem Material. Probleme mit der Headspace-Methode, die auf die beträchtliche Wasser- und Lipidlöslichkeit dieser Substanz zurückgehen, konnten durch eine Aussalz-Technik gelöst werden. Bei Konzentrationen von über 5 μg pro Probe wurde die Identifizierung massenspektrometrisch durchgeführt. Im Rahmen quantitativer Bestimmungen kam bei der GC Tetrahydropyran (Pentamethylen-oxid) bzw. bei der GC/SIM octadeuteriertes THF (TDF) als innerer Standard zum Einsatz. THF konnte bis zu 1 μg pro Probe mittels GC und bis zu 0,1 μg pro Probe mittels GC/SIM nachgewiesen werden. Bei Labortieren wurden Maximalspiegel von THF im Blut ungefähr 1 h nach oraler Applikation beobachtet; die Halbwertszeit lag bei etwa 5 h. Das Verteilungsverhältnis zwischen Gewebe und Blut war etwa 1,5–2 bei Fettgewebe und Niere bzw. praktisch übereinstimmend bei Gehirn, Leber, Milz und Muskulatur.
    Notes: Summary Routine analysis of tetrahydrofuran (THF) in biologic materials has become feasible using GC and GC/MS and the headspace method. Problems of the headspace method and this substance which has a high water-and lipid-solubility were overcome by using the salting-out technique. Identification was made by mass spectral examination, in case of concentrations over 5 μg per sample. For quantitative determinations, tetrahydropyran (pentamethylene oxide) was used as an internal standard in GC, and a stable isotopic substance, octadeuterated THF (TDF) in GC/SIM. THF was detected in 1 μg per sample by GC, and 0.1 μg per sample by GC/SIM. THF blood levels in laboratory animals reached their highest values about 1 h after the oral administration, and the half-life was about 5 h. Ratios of tissue levels to blood were ca. 1.5–2 in the adipose tissue and kidney, and fairly equal in the brain, liver, spleen, and muscle.
