    Notes: In this review I will outline several chemogenetic approaches used to determine the chemical basis of large ribozyme function and structure. The term chemogenetics was first used to describe site-specific functional group modification experiments in the analysis of DNA-protein interactions. Within the past few years equivalent experiments have been performed on large catalytic RNAs using both single-site substitution and interference mapping techniques with nucleotide analogues. While functional group mutagenesis is an important aspect of a chemogenetic approach, chemical correlates to genetic revertants and suppressors must also be realized for the genetic analogy to be intellectually valid and experimentally useful. Several examples of functional group revertants and suppressors have now been obtained within the Tetrahymena group I ribozyme. These experiments define an ensemble of tertiary hydrogen bonds that have made it possible to construct a detailed model of the ribozyme catalytic core. The model includes a functionally important monovalent metal ion binding site, a wobble-wobble receptor motif for helix-helix packing interactions, and a minor groove triple helix. © 1998 John Wiley & Sons, Inc.     Notes: The major photoproduct from irradiation of triphenylsulfonium salts is a Brönsted acid. This photochemical process has been used in several polymer film applications where the acid is used for crosslinking of films or other acid catalyzed reactions. Despite the widespread application of these materials, very little is known about the efficiency of acid generation in polymer films or the extent of the catalytic chain. This paper describes the use of a merocyanine dye technique to analyze for acid production in polymer films. The amount of acid produced on irradiation can be determined by this method and the extent of the catalytic chain then be determined.

    Notes: The pressure loss between the mold and the nozzle in the injection molding of bar and box moldings has been monitored. The pressure drop observed during filling of the mold is reduced during the packing stage but remains finite. This has been attributed in the literature to solidification of polymer across the cavity transducer and to melt relaxation phenomena. Experiments have been carried out with hot molds to prolong the packing stage at the expense of the ‘cooling’ stage. Under these circumstances the pressure drop is reduced but not eliminated. The observed pressure drop may be related to the viscosity of the melt and its dependence on pressure and temperature although strain-induced crystallization and the pressure dependence of the melting point can confer effects similar to the cooling stage.

    Notes: Styrene-butadiene multiblock copolymers were examined with both newly introduced and established rheological techniques and by transmission electron microscopy (TEM) to evaluate shear-induced structural changes in these polymers. Transient rheological tests (based on superposed flow principles) were developed which probed structural changes that occur in the copolymers during and at the cessation of steady shear. Data from these tests indicated that for the cylindrical morphology copolymer (SB1) there were structural changes occurring during steady shear that were recovered upon cessation of shear. The recovery process took place on time scales that could be significant in processing. The lamellar morphology material (SB2) did not exhibit this recovery behavior. Longer-term structure changes were investigated using established techniques and showed differences between the cylindrical and lamellar copolymers. When tested at 210°C, peaks in tan δ occurred at 30 rad/s for SB1 and at 0.5 rad/s for SB2 with saturation strain levels of 150 strain units (SU) for SB1 and 80 SU for SB2. TEM analysis of SB2 indicated that, although rheological changes are significant up to 80 SU, better alignment of the domain morphology can be achieved at moderately low strains of 7 SU. This indicates that the copolymers' rheological changes, which occur as a result of steady shearing, may not be due entirely to domain alignment, but may also be due to more local molecular rearrangements (e.g., chain disentanglement). © 1996 John Wiley & Sons, Inc.

    Notes: The sensitivity that can be obtained with the quadrupole ion trap for on-line CE analysis using a sheathless electrospray interface and gold-tipped columns is examined. Capillaries of different i.d. are compared in continuous infusion experiments and the effects of sheath solvents on the signal for some peptides and proteins are illustrated. The sheathless CE/ITMS system using 20 μm i.d. columns is shown to provide mid-attomole detection levels for leucine enkephalin in total ion electropherograms under full scan conditions. Good quality MS/MS spectra are also obtained for low femtomole injections of this peptide.

    Notes: Segregated composite electrodes mimic microelectrode ensembles. In this preliminary report, the use of a rotating gold-polychlorotri- fluoroethylene (or Kel-F, a 3M Company polymer) composite electrode in combination with pulsed amperometric detection (PAD) is described for the detection of glucose. Comparisons are made with results obtained at a solid gold disk electrode. The composite electrode exhibits a higher signal and a lower background than does the solid gold electrode. In terms of current density, the enhancement of the signal above the background is over 3-fold, similar to that observed with segregated graphite composite electrodes used in a constant potential mode. Little or no glucose signal is observed at either the solid gold or the gold composite electrode when employed in the constant potential mode. In the PAD mode, the signal is stable for periods in excess of an hour.

    Notes: Tandem mass spectra of a variety of adenine-containing deoxynucleic acid anions obtained using collisional activation in a quadrupole ion trap are described. Data are reported for singly charged mono-, di- and trinucleotide species, and also for multiply charged anions derived from tri-, tetra-, penta- and hexanucleotides. Attention is focused on the first step in the unimolecular decomposition of these species, namely the loss of the nucleobase. Specifically, the competition between the loss of the nucleobase as a neutral versus its loss as an anion is addressed within the context of a proposed proton-bound intermediate consisting of the nucleobase and a phosphodiester linkage. The data are interpreted on the basis of the energy surface involved in the break-up of the proton-bound intermediate, which can explain why the competition between loss of a neutral versus charged base is strongly dependent upon the charge of the parent ion.

    Notes: The purpose of this study was to determine the effects of endothelial cell seeding and graft internodal distance upon the performance of 4-mm-ID e-PTFE grafts during acute reduced blood flow conditions. PTFE grafts especially manufactured with three different mean internodal distances (28, 40, and 52 μm) were evaluated. Fifteen dogs (n = 5 for each design of PTFE graft) underwent bilateral carotid artery replacements with 6 cm lengths of 4-mm-ID PTFE grafts. In each dog one graft was seeded with enzymatically derived endothelial cells; the contralateral graft was nonseeded. All grafts were evaluated 5 weeks postoperatively. Dogs with bilaterally patent grafts were subsequently subjected to flow conditions through the graft that were maintained at 30% of the initial flow rates for 4 hr. Following controlled low flows the grafts were excised and assessed for patency, thrombus-free surface area, inner capsule thickness and prostacyclin production. Endothelial cell seeding of these small-diameter e-PTFE vascular grafts improved patency and thrombus-free surface areas in grafts of all pore sizes, with these parameters being greatest in the 40-μm grafts. Inner-capsule healing in these grafts was controlled and related to the pore size. PGI2 production was improved in endothelial cell seeded grafts of all pore sizes. However, neither endothelial cell seeding nor graft pore size affected the performance of these e-PTFE grafts under conditions of reduced blood flows.
