    Notes: Widely applied selection strategies for plasmid-containing cells in unstable recombinant populations are based upon synthesis in those cells of an essential, selection gene product. Regular partitioning of this gene product combined with asymmetric plasmid segregation produces plasmid-free cells which retain for some time the ability to grow in selective medium. This theory is elaborated here in terms of a segregated model for an unstable recombinant population which predicts population growth characteristics and composition based upon experimental data for stable strain growth kinetics, plasmid content, and selection gene product stability. Analytical solutions from this model are compared with an unsegregated phenomenological model to evaluate the effective specific growth rate of plasmid-free cells in selective medium. Model predictions have been validated using experimental growth kinetics and flow cytometry data for Saccharomyces cerevisiae D603 populations containing one of the plasmids YCpG1ARS1, YCpG1ΔR8, YCpG1ΔR88, YCpG1ΔH103, YCpG1ΔH200, pLGARS1, and pLGSD5. The recombinant strains investigated encompass a broad range of plasmid content (from one to 18 plasmids per cell) and probability α of plasmid loss at division (0.05 ≤ α ≤ 0.42). Experimental data for all strains considered is inconsistent with the hypothesis that plasmid-free cells are unable to grow in selective medium. For a given value of a, the fraction of plasmid-containing cells in the population decreases with increasing plasmid content and increases for less stable selection gene products. This conceptual framework and mathematical model will aid in strain development for greater effective stability.

    Notes: A segregated model of multicopy plasmid propagation has been formulated which incorporates plasmid replication and partition functions, as well as the effect of plasmid presence on host growth rate. Growth of plasmid-free cells in selective medium is explicitly analyzed. The model parameters can be determined from experimentally measurable quantities. Propagation of a recombinant multicopy plasmid in the yeast Saccharomyces cerevisiae is analyzed using this model.

    Notes: Experiments were performed to evaluate, qualitatively and quantitatively, the adaptation of Escherichia coli to plasmid maintenance and cloned gene expression. Experimental findings indicate that the metabolic response to low plasmid levels is an increase of the biosynthetic capacity of both transcription and translation. At high copy number levels the gene-specific transcription rate continues to increase but the stability of plasmid-derived mRNA drops sharply. Protein levels are maintained, but translation efficiency decreases. These results indicate that cellular biosynthetic capacity may not be limiting productivity in recombinant systems. If macromolecular stability is the bottleneck, then current efforts to increase gene expression that focus on enhancing synthesis rates will be ineffective.

    Notes: An asynchronous bacterial population has been approximated using a finite number of “computer” cells, each based on a complex single-cell model for Escherichia coli. This formulation correctly simulates the transient responses of protein and total cell mass synthesis rate to the sudden increase in the concentration of limiting energy source in the growth medium. Experimentally observed responses of rRNA and mRNA synthesis rates to growth rate shifts are qualitatively mirrored by the model. Simulation trends following those of a rel- mutant suggest that model modifications are needed to describe the dynamics of the stringent response. Simulations of the responses of recombinant populations to plasmid amplification or plasmid promoter induction also result in behavior similar to that determined experimentally. The calculated responses for recombinant populations subjected to constant promoter induction or cyclic induction-noninduction lead to the conclusion that inducible systems give greater productivity than those with fixed promoter strength. This formulation may be utilized as a basis for exploring other aspects of recombinant population dynamics.

    Notes: A restricted diffusion model is developed to study the immobilization of enzyme in porous solid supports. Simulation studies have been carried out for various combinations of process variables and parameters of the immobilization system. The model has also been used to develop a method for estimating the intrinsic rate constant of immobilization when enzyme diffusion into the support is restricted. Results of experiments in which glucose oxidase was immobilized in porous glass supports are consistent with model simulations.

    Notes: The oxidation of CO over aluminum-supported Ag was studied at 147°C using transient experiments. Long-time-scale dynamics were observed that could not be explained by previously postulated kinetic mechanisms for this reaction system. A model is presented with kinetic parameters estimated from steady state and transient data that is able to reproduce a variety of cycled feed stream experiments. The model incorporates a slow reversible step which forms an adsorbed oxygen species that blocks reaction sites.

Sie wurden polymerisiert, um einen Pyruvatester in die Polymerkette einzuführen. Eine Reihe von Spiromethylencyclohexadienen wurde hergestellt, um neben der Carbonylgruppe und gespannten Ringen weitere Gruppen zu finden, die die radikalische Ringöffnungspolymerisation fördern. Bei ihrer Polymerisation wurden Polymere mit einer p-Phenylengruppe in der Kette erhalten.
    Notes: In order to find a series of monomers with high reactivities in copolymerization and still undergo free radical ringopening polymerization, a series of cyclic acrylates were synthesized and polymerized to insert a pyruvate ester into the backbone of the polymer. In order to discover groups other than the carbonyl groups and strained rings that would promote free radical ring-opening polymerization, a series of spiro methylenecyclohexadienes were prepared and polymerized to give a polymer with a p-phenylene group in the backbone.
