    Notes: Electroblotting proteins separated by gel electrophoresis provides a suitable support for further manipulations and analysis of small amounts of relatively pure samples. On-membrane digestion, peptide mapping by mass spectrometry, and database searching offer sensitive and fast tools to identify the analyte. By providing sequence information, tandem mass spectrometry can go a step further, confirming the database identification, solving problems connected with post-translational modifications and sequence variations, or supplying the stretches of internal sequence necessary to synthesize an oligonucleotide probe for gene isolation. The viability of this approach was successfully evaluated using different tandem mass spectrometric techniques: metastable decomposition in a matrix-assisted laser desorption/ionization (MALDI) time-of-flight instrument with a curved-field reflectron; low energy collision-induced dissociation in a MALDI quadrupole ion trap mass spectrometer; and high energy collision-induced dissociation in a high-performance four-sector mass spectrometer with massive cluster-impact ionization.

    Notes: This paper describes the results of a study on the deactivation of the surface of glass capillary columns by high temperature silylation (HTS). The different steps, leaching, washing, dehydration and HTS were optimized. A practical procedure yielding a high percentage of very good columns is given. The influence of leaching and HTS on the temperature stability and coating efficiency of capillary columns coated with OV-1 was studied. The inertness of the glass surface after HTS is demonstrated by several examples.

    Notes: The ion/molecule reactions of allyl cations and 2-methoxyallyl cations with vinyl methyl ether, 1-chloro-2-ethoxyethene and 1,2-dimethoxyethene are discussed in terms of [2+3] cycloaddition reactions. Deuterium labelling of the cations has been used for the study of the reaction mechanisms. The appearance of various product ions in these ion/molecule reactions lead to the suggestion that in reactions of allyl cations with alkenes non-cyclic [C5H5]+ product ions are formed preferentially, but that in reactions of 2-methoxyallyl cations with alkenes a significant part of the product ions are methoxycyclopentadienyl cations. These observations are ascribed to the stabilizing effect of the methoxy group with regard to the positive charge in the product ions.

    Notes: The 1J(15N13C) values for a series of ring-substituted N,N-dimethylaniline-15N derivatives and a series of nitrobenzene-15N derivatives were measured from the 13C spectra. In the nitrobenzenes, small changes in 1J(15N13C) are attributed to the inductive effect of the substituents, since steric inhibition of conjugation has little effect on the magnitude of the coupling. In contrast, steric inhibition of nitrogen lone-pair delocalization in N,N-dimethylaniline derivatives markedly reduces the value of 1J(15N13C). Theoretical calculations of 1J(15N13C) values for the two series of compounds were made using standard INDO parameters and a ‘sum-over-states’ perturbation approach. Fair agreement between the calculated and experimental values is found.

    Notes: A simple but effective GC approach to odour evaluation and fraction collection with capillary columns is described. An allglass device splits the column effluent, with one splitter arm going to an FID. For odour evaluation, the other splitter arm is led via teflon tubing to the nose. Off-line heart-cutting is carried out by collecting the split effluent on a glass capillary microtrap, the inner walls of which are coated with OV-101. This allows recovery of nanogram amounts of material. Multipass operation or repeated collection leads to preparative capillary GC for further spectroscopic investigation. These techniques do not involve sophisticated equipment or valves. Applications of this type of smell analysis, heart-cutting and enrichment in the analysis of Rhododendron fragrantissimum essential oil, black pepper essential oil and hop essential oil, illustrating the versatility of the systems, are presented.

    Notes: A simple method is described for the quantitative determination of organochlorine pesticides and polychlorinated biphenyls (PCB's) in water at the sub-ppb level. A micro gas-phase extractor advantageously replaces other preconcentration and purification techniques. The extract is analyzed by capillary gas chromatography without further enrichment. The recovery at the ppb level was nearly 100% for organochlorine pesticides and more than 80% for PCB's. The complete procedure including sample preparation, steam distillation-extraction, and capillary gas chromatographic analysis is carried out in less than four hours.

    Notes: Changes of fluorescence intensity at the far side (transmission) and near side (remission) in cases where a spot of a fluorogen lies in different sublayers are calculated using a multilayer model. Examples shown are: the relative intensity of fluorescence in the layer as a function of the absorption coefficient of a sorbent at the excitation wavelength; the relative intensity of fluorescence at the far and near sides of a TLC plate; the influence of using too broad a monochromatic filter at the emission side on the results of fluorescence measurements. Advantages of near-side scanning of fluorescence compared to far-side scanning are discussed. The most important source of errors in fluorodensitometric measurements is the effect of secondary chromatography.

The emission and excitation spectra of the reserpine and the absorption spectrum of the ajmaline were scanned. The linear range between peak area and concentration is given.
