    Notes: Objective To establish whether the timing of delivery between 37 and 42 weeks gestation influences neonatal respiratory outcome and thus provide information which can be used to aid planning of elective delivery at term.Design All cases of respiratory distress syndrome or transient tachypnoea at term requiring admission to the neonatal intensive care unit were recorded prospectively for nine years.Setting Rosie Maternity Hospital, CambridgeSubjects During this time 33,289 deliveries occurred at or after 37 weeks of gestation.Main outcome measures This information enabled calculation of the relative risk of respiratory morbidity for respiratory distress syndrome or transient tachypnoea in relation to mode of delivery and onset of parturition for each week of gestation at term.Results The incidence of respiratory distress syndrome at term was 2.2/1000 deliveries (95% CI; 1.7–2.7). The incidence of transient tachypnoea was 5.7/1000 deliveries (95% CI; 4.9–6.5). The incidence of respiratory morbidity was significantly higher for the group delivered by caesarean section before the onset of labour (35.5/1000) compared with caesarean section during labour (12.2/1000) (odds ratio, 2.9; 95% CI 1.9–4.4; P 〈 0.001), and compared with vaginal delivery (5.3/1000) (odds ratio, 6.; 95% CI 5.-8.9; P 〈 0.001). The relative risk of neonatal respiratory morbidity for delivery by caesarean section before the onset of labour during the week 37+0 to 37+6 compared with the week 38+0 to 386 was 1.74 (95% CI 1.1–2.8; P 〈 0.02) and during the week 38+0 to 37+6 compared with the week 39+0 to 39+6 was 2.4 (95% CI 1.2–4.8; P 〈 0.02).Conclusions A significant reduction in neonatal respiratory morbidity would be obtained if elective caesarean section was performed in the week 39+0 to 39+6 of pregnancy.

    Notes: Abstract: The coexpression of sulphonylurea binding sites and ATP-sensitive K+ (KATP) channels was examined in the rat motor cortex, an area of the CNS exhibiting a high density of sulphonylurea binding. These channels were not detected on neuronal cell bodies, but sulphonylurea-sensitive KATP channels and charybdotoxin-sensitive, large-conductance calcium-activated K+ BKCa channels were detected by patch clamping of fused nerve terminals from the motor cortex. Subcellular fractionation revealed that high-affinity sulphonylurea binding sites were enriched in the nerve terminal fraction, whereas glibenclamide increased calcium-independent glutamate efflux from isolated nerve terminals. It is concluded that neuronal sulphonylurea receptors and KATP channels are functionally linked in the motor cortex and that they are both selectively expressed in nerve terminals, where the KATP channel may serve to limit glutamate release under conditions of metabolic stress.

    Notes: Abstract: In this report we investigate the isoforms of protein kinase C (PKC) present in cultured adrenal chromaffin cells with respect to their modulation by treatment with phorbol ester and their possible differential involvement in the regulation of responses to histamine and bradykinin. The presence of individual isoforms of PKC was investigated by using eight isoform specific antisera, as a result of which PKC-α, ε, and ζ were identified. To characterize down-regulation of these enzymes, cells were incubated for 6–48 h with 1 µM phorbol myristate acetate (PMA). PKC-ε down-regulated more rapidly than PKC-α. At 12 h, PMA pretreatment, for example, PKC-ε was maximally down-regulated (23 ± 4% of controls), whereas PKC-α was unchanged. PKC-α showed partial down-regulation by 24 h of PMA pretreatment. PKC-ζ did not down-regulate at any of the times tested. Translocation from cytosol to membrane in response to PMA was also more rapid for PKC-ε than for PKC-α. The accumulation of total 3H-inositol (poly)phosphates in response to bradykinin or histamine was essentially abolished by prior treatment with 10-min PMA treatment (1 µM). However, with 12-h exposure to PMA, the bradykinin response was restored to the level seen with no prior PMA exposure. The histamine response showed no recovery by 12 h of PMA, but showed partial recovery by 24 h of PMA pretreatment. These observations showed that the restoration of the response to bradykinin corresponds to the loss of PKC-ε, whereas the restoration of the histamine response corresponds to the loss of PKC-α. This picture was confirmed with further studies on cytosolic Ca2+. The results show that chromaffin cells exhibit an unusual pattern of down-regulation of PKC isoforms on prolonged exposure to PMA, and that there is a differential effect of exposure to PMA on the histamine and bradykinin responses, suggesting that different PLC-linked receptors in chromafin cells are differentially regulated by PKC isoforms.

    Notes: Abstract: Phosphoinositide-specific phospholipase C (PLC) is a key enzyme in signal transduction. We have previously demonstrated that an antibody to an isozyme of PLC, PLC-δ, produced intense staining of neurofibrillary tangles in the brains of patients with Alzheimer's disease. In the present study, we investigated the protein level and activity of this enzyme in control and Alzheimer brains. Western blot analysis using a specific antibody for PLC-δ showed that the concentration of PLC-δ protein was significantly higher in the cytosolic fraction of Alzheimer's disease cortical tissue than in control brains. The activity of PLC-δ, which hydrolyzes phosphatidylinositol, was also investigated, and we found that PLC-δ activity was not significantly different in the Alzheimer and control cytosolic fractions. These results indicate that the specific activity of PLC-δ is decreased in Alzheimer brains and suggest that inactivation of PLC-δ might be related to the pathophysiology of this disease.

    Notes: The near ultraviolet (UV) and visible photodissociation dynamics of BrCl have been explored using the technique of photofragment ion imaging at 26 wavelengths in the range 235 to 540 nm. Ion images of the Cl(2P3/2), Cl(2P1/2), Br(2P3/2) and Br(2P1/2) photofragments reveal both the angular distributions of photofragment velocities (characterized by anisotropy parameters, β) and which of the four possible photofragment pathways are active at different wavelengths. The anisotropy parameters show extensive variation both with wavelength and for the different fragmentation channels, and these variations are interpreted largely in terms of excitations to the A 3Π(1), B 3Π(0+), C 1Π(1) and D(0+) states as the wavelength is reduced. At wavelengths between 235 and 262 nm, the Br(2P1/2)+Cl(2P3/2) channel is dominant and β=2.0±0.1 at 235 nm, characteristic of a parallel parent transition (ΔΩ=0) and supporting previous assignments of the absorption in this wavelength range being due to the D(0+)–X 1Σ+(0+) transition. A minor channel forms Cl(2P1/2)+Br(2P3/2) with an anisotropy indicative of the involvement of an underlying perpendicular absorption (ΔΩ=±1) to a state with Ω=1. Br(2P3/2)+Cl(2P3/2) and Br(2P1/2)+Cl(2P1/2) fragmentation channels are not observed. Excitation in the wavelength range 320 nm to 410 nm results in Cl(2P3/2)+Br(2P3/2) products with an anisotropy parameter of β=−1.0±0.1, consistent with assignment of the strong parent absorption to the C 1Π(1)–X 1Σ+(0+) transition. For photolysis wavelengths longer than 400 nm, the Cl(2P1/2)/Cl(2P3/2) branching ratio increases [with β∼1.0 to 1.4 for the Cl(2P1/2)], β for Cl(2P3/2) becomes less negative (and for λ≥450 nm, values lie in the range 0 to −0.2) and Br(2P3/2) β-parameters also increase. No formation of Br(2P1/2) is observed. These observations are, in part, consistent with absorption via the B 3Π(0+)–X 1Σ+(0+) transition, although the nonlimiting β-parameter values imply a significant perpendicular contribution to the absorption spectrum. The measured anisotropy parameters for λ≥410 nm are interpreted in terms of excitation both to an Ω=0 state [B 3Π(0+)] and an Ω=1 state [A 3Π(1) or C 1Π(1)], together with transfer of dissociating flux between states during the dissociation. © 1998 American Institute of Physics.

    Notes: Molecular dynamics simulations of many degree of freedom systems are often comprised of classical evolutions on quantum adiabatic energy surfaces with intermittent instantaneous hops from one surface to another. However, since quantum transitions are inherently nonadiabatic processes, the adiabatic approximation underlying the classical equations of motion does not hold in the regions where quantum transitions take place, and the restriction to classical trajectories for adiabatic quantum states is an approximation. Alternatives which employ classical paths that account more fully for nonadiabaticity can be computationally expensive and algorithmically complicated. Here, we propose a new method, which combines the surface hopping idea with the mean-field approximation for classical paths. Applied to three test systems, the method is shown to outperform the methods based on an adiabatic force without significant extra effort. This makes it an appealing alternative for modeling complex quantum–classical processes. © 1997 American Institute of Physics.

    Notes: Abstract: Many Gs-linked receptors have been reported to use multiple signalling pathways in transfected cells but few in their normal cell environment. We show that the adenosine A2a receptor uses two signalling pathways to increase the release of acetylcholine from striatal nerve terminals. One pathway involves activation of Gs, adenylyl cyclase, protein kinase A, and P-type calcium channels; the other is mediated by a cholera toxin-insensitive G protein, protein kinase C, and N-type calcium channels. The effects of these two pathways are not additive, the second pathway being inhibited by the first; but they are equally sensitive to the A2a receptor antagonist KF17837. This demonstrates that the A2a receptor activates two signalling systems in striatal cholinergic neurons.
