    Description / Table of Contents: Summary   Design patterns and frameworks, two quite new concepts, allow the reuse of design and of individual design experience. A design pattern embodies local design experience that is related to a few classes, whereas a framework embodies a global kind of experience that covers essential aspects of an application. This article describes experiences with these advanced object-oriented concepts in computer-science education. Concrete examples from projects show how design patterns were successfully used by inexperienced students for designing parts of frameworks. The very positive experiences strongly encourage the inclusion of design patterns and frameworks into computer science education.
    Notes: Zusammenfassung   In den letzten Jahren wurden zwei Konzepte entwickelt, welche die Wiederverwendung von Entwurfserfahrung erlauben. Dies sind zum einen Entwurfsmuster, welche lokale, sich jeweils auf wenige Klassen beziehende Entwurfserfahrung zur Wiederverwendung durch andere Entwickler verfügbar machen, zum anderen Frameworks. Diese erlauben die Wiederverwendung einer globalen Art von Entwurfserfahrung, welche wesentliche Aspekte einer Anwendung umfaßt. Dieser Artikel erläutert Erfahrungen mit Entwurfsmustern bei der Frameworkentwicklung. An einem Beispielprojekt erläutert er die erfolgreiche Arbeit von Studenten mit diesen Methoden. Das Ergebnis ermuntert zur Aufnahme entsprechender Methoden in die Informatiker-Ausbildung.

    Notes: Zusammenfassung Mit einer dünnwandigen Druckionisationskammer und einem Röhrenelektrometer wurden die Sto\verteilungskurven und Sto\auslösekurven für Stö\e 〉 136000 EQ (etwa 〉 10 Korpuskeln entsprechend) für die Sto\strahler Blei, Eisen und Aluminium aufgenommen. Für die Sto\auslösekurven von Fe und Al ergaben sich jeweils zwei Maxima; für diejenigen von Pb stellte sich in dem erfa\ten Bereich nur ein Maximum heraus, jedoch deutet sich bei den grö\ten erfa\ten Stö\en die Ausbildung eines zweiten Maximums an. (Lagen der Maxima: 2,25 cm Pb; 4,5 cm Fe; etwa 10 cm Fe; etwa 6,5 cm Al; etwa 35 cm Al). Das erste Maximum tritt besonders im Gebiet der kleinen Stö\e hervor, das zweite im Gebiet der grö\eren. Eine kontinuierliche Verlagerung des Maximums in Abhängigkeit von der Sto\grö\e ist nicht vorhanden. Eine Deutung wird mit der überlagerung zweier Prozesse versucht. Der eine verursacht das erste Maximum und besteht in einer Multiplikation der Luftschauer, der zweite ist als eigentlicher Sto\proze\ anzusehen und ist wahrscheinlich durch Mesotronen hervorgerufen.

    Notes: Zusammenfassung Blasenbauchfell eignet sich gut zur Deckung von serosaentblößten Stellen im kleinen Becken. Meist genügt es, eine Falte des in stets ausreichendem Maße vorhandenen Blasenbauchfells über die Nahtlinie am Gebärmuttergrunde oder am Gebärmutterstumpfe bis auf die hintere Gebärmutterwand oder bis in den Douglas zu ziehen. Wenn aber eine große Wundfläche an der Hinterwand des Gebärmutterhalses und im Douglas nach Entfernung von ausgebreiteten Endometriosisherden bei einer jüngeren Frau mit gesunden Eileitern nicht restlos gedeckt werden kann, so läßt sich das Blasenbauchfell als gestielter Lappen nach Drehung um 90° gut in den Bauchfelldefekt einnähen. Bei 12 derart operierten Frauen ist nach störungsfreiem Verlaufe auch ein gutes Dauerergebnis erzielt worden.

    Notes: Abstract The 20S proteasome (prosome) is a highly organized multiprotein complex with approximate molecular weight of about 700 kDa. Whilst the role of the proteasome in the processing and turnover of cellular proteins is becoming clearer, its relationship with RNA remains still obscure. Here we focus on the nature and function of proteasome associated endonuclease activity. Thus the involvement of a proteasome α-type subunit in RNA-degradation, the catalytic requirements, the interaction of proteasomes with their RNA-substrate and the identification of a well defined cleavage site in the 3-UTR of short-lived cellular mRNAs will be described in detail. All data indicate that proteasomes associated endonuclease activity could be involved in post-transcriptional gene control at the level of translation. Over the last decade the possibility of association of proteasomes with specific RNAs or mRNPs have been particularly controversial. Proteasomes were reported to inhibit translation of viral mRNAs and to be tightly associated with RNase activity. It is possible that proteasomes are also involved in cellular RNA breakdown and RNA processing like prokaryotic RNase E.

    Notes: Abstract The interrelationships between proteasomes and viral gene products are very complex. 20S proteasomes associate with a number of viral mRNAs which are cleaved by proteasome's associated endonuclease activity. In addition proteasome's endopeptidase activities are involved in the presentation of viral antigens. Viral proteins of different origin associate with the 20S and 26S complexes and interfere with their enzymatic activities. A major part of this review deals with the interactions between 20S proteasomes and the gene products of the human immunodeficiency virus (HIV) which has been studied in detail by our group.

    Notes: Abstract The effects of columns (Nucleosil C18ODS, MZ-PAH, YMC-PACK C30), column properties (inner diameters of 4 mm, 3 mm and 2 mm, pore-width 10 nm and 30 nm) and eluents (methanol, acetonitrile, acetone, water) were tested on the separation of algal pigments. The length of columns was 250 mm and particle size was 5 μm. Flow rates and gradients were adjusted to optimize peak separation; remaining chromatographic conditions were kept constant. The resolution of chromatographic systems was tested with pigment standards and various algal cultures. Total flow rate and retention times decreased with decreasing inner diameter, whereas pressure, sensitivity and peak-width increased. Pore width had negligible effects on the chromatographic separation of pigments under the test conditions. Only with acetonitrile as eluent were all the taxonomically important pigments resolved adequately: zeaxanthin (Cyanophyceae), lutein (Chlorophyceae), fucoxanthin (Bacillariopyceae), alloxanthin (Cryptophyceae), peridinin (Dinophyceae).

    Notes: High resolution two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) is a powerful research tool for the analytical separation of cellular proteins. The qualitative and quantitative pattern of polypeptides synthesized by a cell represents its phenotype and thus defines characteristics such as the morphology and the biological behavior of the cell. By analyzing and comparing the protein patterns of different cells it is possible to recognize the cell type and also to identify the most typical features of these cells. In applied pathology it is often difficult to identify the tissue of origin and the stage or grade of a neoplasia by cellular morphology analyzed by classical or immunostaining procedures. The protein pattern itself is the most characteristic feature of a cell and should therefore contribute to the identification of the cell type. For this reason we separated protein fractions originating from different lung tumor cell lines using 2-D PAGE and we compared the resulting patterns on a multivariate statistical level using correspondence analysis (CA) and ascendant hierarchical clustering (AHC). The results indicate that (i) protein patterns are highly typical for cells and that (ii) the comparison of the protein patterns of a set of interesting cell types allows the identification of potentially new marker proteins. 2-D PAGE is thus a unique and powerful tool for molecular cytology or histopathology, unveiling the protein expression level of tissues or cells.
