Notizen: This paper summarizes recent structural and functional studies on tau protein, its interactions with microtubules, its self-assembly into paired helical filaments (PHF)-like fibers, and its modification by phosphorylation. The structure of tau in solution resembles that of a random coil. Both tau and Alzheimer PHFs have very little secondary structure, making it improbable that the assembly of tau into PHFs is based on interacting beta sheets. Tau's binding to microtubules can be described by a “jaws” effect. The domain containing the repeats binds very weakly, while the flanking regions (jaws) bind strongly, even without the repeats. However, only the combination of flanking regions and repeats makes binding productive in terms of microtubule nucleation and assembly. Although the majority of Alzheimer-like phosphorylation sites are outside the repeats they have only a weak influence on binding, whereas the phosphorylation at Ser262 inside the repeats inhibits binding and makes microtubules dynamically unstable. This site can be phosphorylated by kinases present in brain tissue, and it is uniquely phosphorylated in Alzheimer brain.

Notizen: Abstract 13- and 26-Membered crown ethers have been synthesized based on resorcinol and 1,8-dichloro-3,6-dioxaoctane. The products with substituents in the benzene ring have been prepared by alkylation of 13-membered crown ether. Complexing properties of the macrocycles have been studied with the use of ion-selective membrane electrodes. The structures of 13- and 26-membered crown ethers have been established by X-ray structural analysis.

Notizen: Summary A possibility of extending analytical applications of chemically modified silica gels is described. This involves their utilization for the generation of gaseous standard mixtures consisting of methyl chloride as the analyte and nitrogen as a carrier gas to be used for the calibration of the GC-FID system. N-methylmorpholine was chemically bonded to the propylsilylated surface of silica gel forming chloride of an appropriate immobilized compound which, under certain conditions, undergoes thermal decomposition yielding a single, volatile component (methyl chloride). Such a method of generating specific amounts of a standard substance can be used both for a single point calibration and for checking the accuracy of an analytical instrument in a relatively wide measurement range. It was found that 3.40±0.081 mg of methyl chloride can be generated per 1 g of the modified gel.

Notizen: Abstract Sodium iodide complexes of 13-membered azo-(I)and azoxycrown ethers (II) have been synthesized. Compound I[Na(L1 trans)2]⋅I⋅H2O is triclinic witha = 11.53(2), b = 15.74(2), c = 19.17(2) Å,α = 98.93(9), β = 105.51(9),γ = 89.20(9) deg.; Z = 4, space groupP1. Compound II [Na(L2)2]⋅I is orthorhombic witha = 12.451(2), b = 13.796(3), c = 18.667(4)Å; Z = 4, space group P212121. In bothcomplexes the cation is coordinated tochain oxygen atoms and to one nitrogen atom of the azoor azoxy unit. The ability of bothligands to form complexes in relation to thegeometry of the azo or azoxy subunit of themacrocycle is discussed.

Notizen: Abstract The lithium bromide hydrate complex ofthe 13-membered dibenzoazocrown ether of composition{[LiL13(H2O)]Br}2 has been synthesized. Its structure has been determined by the X-ray method. Thecrystals are monoclinic, space group C2/c, a =12.720(6), b = 19.969(9), c = 13.963(6)Å; β = 104.32(2)°. Selectivities of membraneelectrodes based on 13-membered azocrown ethers arediscussed in terms of complex stabilities and metalcation extractabilities as related to the structuresof the respective complexes.
