    Notes: Abstract Glioneuronal malformations with a striking histological resemblance to cortical tubers of tuberous sclerosis, but no extracerebral stigmata of this phacomatosis, are frequently encountered in patients with chronic pharmacoresistant epilepsies. It is controversial as to whether these lesion represent a forme fruste of tuberous sclerosis or a distinct entity. The recently reported loss of heterozygosity (LOH) at the regions of the TSC1 or TSC2 locus in hamartomas obtained from different organs of patients with established tuberous sclerosis, including cortical tubers, stimulated us to examine epilepsy-associated tuberous sclerosis-like glioneuronal malformations with respect to LOH at the TSC1 and TSC2 loci of chromosomes 9q34 and 16p13.3, respectively. The analysis was carried out on DNA derived from paraffin-embedded brain tissues of 11 patients. For 5 patients, peripheral blood leukocytes were also available for DNA extraction. We performed microsatellite analysis with five markers on chromosome 9 and four markers on chromosome 16. In addition, polymerase chain reaction-restriction fragment length polymorphism (RFLP) analysis was performed using a polymorphic EcoRV restriction site in exon 40 of the TSC2 gene. No LOH was identified in any of the cases. These findings do not support a relationship between the epilepsy-associated glioneuronal lesions and tuberous sclerosis. However, tuberous sclerosis is genetically heterogeneous and microsatellite and RFLP analysis cannot exclude small deletions or point mutations. Thus, given the histopathological similarity of glioneuronal malformations in epilepsy patients to cortical tubers, further molecular genetic studies will be needed as our understanding of the molecular basis of tuberous sclerosis increases to completely clarify the relationship of these lesions to tuberous sclerosis.

    Notes: Abstract Tuberous sclerosis (TSC) is an autosomal dominant condition characterised by tumour-like malformations (hamartomas) in the brain and other organs. A proportion of hamartomas from patients with TSC show loss of heterozygosity (LOH) for DNA markers in the region of either the TSCI gene on chromosome 9834 or the TSC2 gene on 16p13.3. This implies that these lesions are clonal. We have studied X-chromosome inactivation, as a marker of clonality, in 13 hamartomas from females with TSC. The hamartomas comprised five renal angiomyolipomas, three fibromas and seven other lesions. In previous studies, four of the lesions showed LOH. A polymerase chain reaction assay was used to analyse differential methylation of anHpalI restriction site adjacent to the androgen-receptor triplet-repeat polymorphism on Xg11-12. In 12 of the lesions, there was a skewed inactivation pattern with one X chromosome being fully methylated and the other unmethylated. Normal tissue showed a random pattern of inactivation. These data confirm that most TSC hamartomas are clonal in origin. This is an intriguing finding, since these lesions are composed of more than one cell type.

    Notes: Abstract This article argues that existing concepts of the civil society–democracy relationship are misleading guides for analyzing political development because they incorporate limited perspectives on the nature and activities of civil society, particularly the nonprofit sector. First, three contemporary conceptualizations of civil society are critiqued, and a contradiction is noted between the importance assigned to ideational functions and the inadequacy of reliance upon elections and party systems. Particular emphasis is placed on civil society's involvement in policy formation, for it is there that a weak state–society connection may be discovered, with significant ramifications for democratic development. Second, recent political instability in the Czech Republic is related to negative trends in political attitudes, indicating a weak connection. Third, the general nature of Czech policy-making processes is examined, and then a closer look is taken at the formation of nonprofit legislation and environmental policy. Finally, implications for the future development of Czech democracy are drawn.

    Notes: Abstract Strategies considered and studied for achieving ammonium excretion in nitrogen fixing bacteria include 1) inhibition of ammonium assimilation and 2) interference with the mechanisms by which ammonium inhibits either nitrogenase synthesis or activity. These aspects of nitrogen fixation have been best studied in diazotrophic Proteobacteria and Cyanobacteria and those of the former are reviewed in this paper. Ammonium assimilation by glutamine synthetase (GS) can be diminished or prevented by treatment of bacteria with chemicals that inhibit GS activity and in some diazotrophs, such treatment results in excretion of up to 15mM ammonium into liquid growth medium. Also, mutants with altered GS activity, isolated by selection for resistance to GS inhibitors, often excrete ammonium. In Proteobacteria, ammonium inhibits nitrogenase activity and/or synthesis, the latter by preventing activity or expression of NifA, a transcriptional activator required for expression of other nif genes. In Azotobacter vinelandii, ammonium inhibits NifA activity but not its synthesis; NifL mediates this effect by interacting directly with NifA causing its inactivation. In nifL insertion mutants, NifA is constitutively active and up to 10 mM ammonium is excreted during nitrogen fixation. GlnD insertion/deletion mutations are unable to be stably maintained in A. vinelandii wild type but are stable and viable in a mutant that produces constitutively active GS (cannot be adenylylated). This confirms the hypothesis that GlnD is required for activity of GS, an essential enzyme in A. vinelandii. In addition, the stable glnD mutants are Nif, supporting also the previous conclusion that GlnD is involved in mediating NifL/NifA interaction. Mechanisms of inhibition of synthesis or activity of NifA by ammonium in other diazotrophs are discussed and compared.

    Notes: A high-frequency inductive power distribution (HID) technology has been developed that generates sinusoidal magnetic fields at a frequency of 10 kHz. In typical industrial applications, field intensities in the order of 0.2 mT can be expected between the current-carrying coils. Because the possible health effects of 10 kHz sinusoidal magnetic fields of this type had never been investigated, a broad evaluation of possible effects on animal health was made in a preliminary 14 day acute study and in a 90 day subchronic study using male and female B6C3F1 mice. Exposures were at 0.08, 0.28, and 1.0 mT vs. a background exposure of 3.7 μT and were essentially continuous.These studies failed to demonstrate any health effects that can be clearly related to the magnetic field exposure. No changes in animal behaviour or indications of morbidity were detected during the initial exposure to the fields. There were no significant differences in body weight between exposed and unexposed (control) mice at any time in the study, and the clinical chemistry and hematology parameters were essentially unchanged. Although minor differences in some clinical chemistry and hematology parameters were seen between control and exposure groups, the lack of exposure dependence, the lack of consistency between sexes, and the lack of correspondence with the results of the two studies all suggest that these were chance associations. Even if the changes were real, the magnitude of the changes was very small and does not indicate serious biological effects. Finally, all organs were macroscopically and microscopically normal except for isolated, generally mild, histological lesions and lesions that were ascribed to fighting among males. There was no obvious association with field intensity. © 1996 Wiley-Liss, Inc.
