    Notes: Abstract: We have investigated which α2-receptor subtypes are expressed in cultured cortical astroglia, and their coupling to second messengers. Binding assays using [3H]rauwolscine showed a very low number of α2 receptors in the astrocytic cultures. Treatment of cultures with dibutyryl cyclic AMP (dBcAMP) increased significantly the number of receptors. The RNase protection assay was used to investigate which receptor subtype the cells express. The α2B message was expressed at a low level in both treated and untreated cells, the levels of mRNA for the α2A/D subtype were up-regulated significantly in cells treated with dBcAMP and no expression of mRNA for the α2C subtype was detected. The α2 agonist dexmedetomidine inhibited forskolin-induced increases in cyclic AMP both in treated and untreated cultures in a pertussis toxin-dependent manner. This effect was abolished by the α2-receptor antagonist rauwolscine. Selective α2-receptor agonists dexmedetomidine, clonidine, and UK14,304 all increased intracellular calcium only in dBcAMP-treated cells. The antagonist rauwolscine abolished this effect. Ca2+ responses were also seen in the absence of extracellular Ca2+ and they were inhibited by the phospholipase C inhibitor U-73122, suggesting that astroglial α2 receptors are coupled to the inositol phospholipid pathway. We therefore also tested the effect of dexmedetomidine directly on inositol 1,4,5-trisphosphate accumulation. A significant increase was seen that was blocked by the antagonist rauwolscine and, as expected, by U-73122. In short, the results demonstrate that the α2 receptors in astroglia are coupled to multiple second messenger pathways. They are up-regulated in cells treated with dBcAMP, which simultaneously assume a process-bearing morphology. If this morphological change reflects some in vivo process such as reactive gliosis, the up-regulation of α2-receptor expression could mean an adaptive change in astrocytic responses to a common neurotransmitter, noradrenaline.

    Notes: Abstract: In examining steroid synthesis in the CNS, expression of the mRNAs encoding for cytochrome P450 side-chain cleavage enzyme (P450SCC) and 3β-hydroxysteroid dehydrogenase/Δ5-Δ4 isomerase (3β-HSD) has been studied in the rat brain. P450SCC transforms cholesterol into pregnenolone and 3β-HSD transforms pregnenolone into progesterone. PCR was used to amplify cDNA sequences from total RNA extracts. Classical steroidogenic tissues, like adrenal and testis, as well as the non-steroidogenic tissue lung have been used as controls. The expression of P450SCC and 3β-HSD have been demonstrated by PCR in cortex, cerebellum, and spinal cord. In addition, primary cultures of rat cerebellar glial cells and rat cerebellar granule cells were found to express P450SCC and 3β-HSD at comparable levels. Furthermore, three of the four known isoenzymes of 3β-HSD were identified, as determined using selective PCR primers coupled with discriminative restriction enzymes and sequencing analysis of the amplified brain products. Using RNA probes, in situ hybridization indicated that P450SCC and 3β-HSD are expressed throughout the brain at a low level and mainly in white matter. Enrichment of glial cell cultures in oligodendrocytes, however, does not increase the relative abundance of P450SCC and 3β-HSD mRNA detected by PCR. This discrepancy suggests that the developmental state of cultured cells and their intercellular environment may be critical for regulating the expression of these enzymes. These findings support the proposal that the brain apparently has the capacity to synthesize progesterone from cholesterol, through pregnenolone, but that the expression of these enzymes appears to be quite low. Furthermore, the identification of these messages in cerebellar granule cell cultures implies that certain neurons, in addition to glial cells, may express these steroidogenic enzymes.

    Notes: The spatial and temporal evolution of the components of a plasma surrounding a 90° convex and a 90° concave electrode whose potential is suddenly decreased in a three-component plasma is numerically investigated. The quasineutral plasma consists of positive ions and various proportions of negative ions and electrons. The results are compared and contrasted with those that are obtained in a normal two-component plasma. The transient or ion matrix sheath is identified and its subsequent evolution is investigated.     Notes: We have performed ab initio calculations on the model cluster (OH)3Si–O–Si(OH)3 with lithium or sodium cations coordinated to the central oxygen in order to refine the relationships between 17O quadrupolar coupling parameters and the local structure around the bridging oxygen in the case of alkali silicates. We have also used a point charge model to derive approximate expressions to describe the dependence of the 17O electric field gradient tensor on the orientation of the alkali cation-bridging oxygen internuclear vector(s). From these calculations we predict that the previously established trend in 17O quadrupolar coupling constant, Cq, with Si–O–Si angle is systematically shifted to lower magnitudes with increasing number and field strength of coordinating alkali cations and that Cq will be relatively insensitive to variations in the alkali cation-bridging oxygen internuclear vector orientation. The previously established trend in 17O quadrupolar coupling asymmetry parameter, ηq, with Si–O–Si angle is systematically shifted to higher values by the presence of one coordinating alkali cation, and only slightly shifted to higher values by the presence of two coordinating alkali cations. As with the quadrupolar coupling constant, the magnitude of the shift in asymmetry parameter increases with increasing field strength of the coordinating alkali cation(s). In contrast to Cq, ηq is additionally dependent on variations in the alkali cation-bridging oxygen internuclear vector orientation. With two or more coordinating alkali cations, however, the magnitude of these variations are reduced, and for certain alkali cation configurations are eliminated. Finally, the parametric dependence of Cq on ηq via the Si–O–Si angle can be used to distinguish between bridging oxygen environments with differing number of coordinating alkali cations. © 1998 American Institute of Physics.

    Notes: Reflection of modified Korteweg–de Vries solitons from the sheath in front of a negatively biased metal disk is experimentally investigated in a negative ion plasma. Both rarefactive and compressive solitons are reflected and the polarity of the reflected solitons is the same as that of the incident solitons. The points of reflection for the rarefactive and compressive solitons are at different locations. As the magnitude of the negative bias potential that is applied to the reflector is increased, the two reflection points move away from the reflector. An interpretation in terms of the sheath characteristics is presented. © 1996 American Institute of Physics.

    Notes: The expansion of a plasma consisting of positive ions, negative ions, and electrons into a vacuum is solved numerically. In addition to the expected self-similar expansion found in a quasineutral plasma expansion model, a burst of positive ions is accelerated into the vacuum and a burst of negative ions is accelerated into the plasma.     Notes: Experiments on the excitation of ion acoustic solitons using a fine mesh grid in a normal two component plasma are described. The excitation is novel in that a modulated high-frequency sinusoidal wave voltage signal is applied to the grid. The carrier frequency of the high-frequency sinusoidal wave is above the ion plasma frequency. An interpretation of the velocity modulation and bunching of free streaming ions that pass through the grid to which the signal is applied is given. © 1997 American Institute of Physics.

    Notes: Abstract: The effect of ethanol on the intracellular Ca2+ concentration response to NMDA in rat cerebellar granule cells grown in low or high KCI concentrations has been studied using image analysis. The cells grown in low KCI displayed high sensitivity for glycine. The subtype-selective antagonist ifenprodil inhibited the response with high (in the low micromolar range) and low (in the high micromolar range) potency. Ethanol affected the high-potency component in these cultures. In cells grown in high KCI the glycine sensitivity was lower, and a low potency for ifenprodil (high micromolar) dominated. These cells were not significantly sensitive to ethanol. The results indicate that the component displaying potency for ifenprodil in the low micromolar range with properties of the NR2B subunit is the target for ethanol action on the NMDA receptor.
