    Notes: Abstract: Syntaxin, a protein required for the docking of synaptic vesicles, may be involved in the manifestation of synaptic plasticity. The possible involvement of syntaxin in epileptogenesis was investigated by assessing the expression levels of syntaxin1B/GR33 mRNA by in situ hybridization at different stages of hippocampal kindling epileptogenesis and after the induction of generalized seizures. Densitometric analysis of the autoradiograms revealed that the expression was not changed in pyramidal and granular neurons of the hippocampal formation 24 h after the first kindling stimulation. However, the mRNA levels in CA1, CA3, and fascia dentata neurons were bilaterally enhanced after six afterdischarges and remained at this elevated level during the whole period along which afterdischarges were elicited. An immunoassay was unable to reveal a clear significant increase of syntaxin1B/GR33 protein levels in hippocampus homogenates of fully kindled animals. The use of syntaxin1B-specific antibodies is necessary to draw definite conclusions on the changes at the protein level. At long term, 4 weeks after the last kindling-elicited generalized seizure, no significant alterations in transcript levels could be detected. The results suggest that the induction of kindling epileptogenesis is associated with an enhanced expression of syntaxin1B/GR33, but this enhanced expression is not necessary for persistence of kindling-induced synaptic plasticity.

    Notes: Abstract: Muscimol-stimulated radiotracer 36Cl− uptake in synaptoneurosomes was used to investigate the function of the GABAA receptor complex in the CA1-3 area and fascia dentata (granular and molecular layers and hilus) of rats kindled by stimulation, twice a day, of the Schaffer collateral fibers. Two kindled groups were studied: (a) 24 h after the last generalized tonic-clonic seizure [fully kindled (FK) stage] and (b) 28 days after the last generalized seizure (long-term stage). Synaptoneurosomes were prepared in parallel from subslices of the CA1-3 area and fascia dentata. In FK animals, the muscimol-stimulated 36Cl− uptake was significantly reduced by 21% in the CA1-3 area in comparison with nonstimulated controls, whereas a significant increase of 29% was found in the fascia dentata. Significant changes were no longer present at 4 weeks after the last generalized seizure. The observed changes in muscimol-stimulated 36Cl− uptake at the FK stage closely parallel the recently observed changes in [3H]muscimol binding in the CA1 area and fascia dentata. These results indicate that kindling causes a transiently decreased GABAA receptor-mediated function in the CA1-3, in contrast to an increased GABAA receptor-mediated function in the fascia dentata.

    Notes: Summary A survey is given of the principles underlying the diagnosis of brain tumours. Traditionally diagnosis and localization of brain tumours have been based upon morphological criteria. Currently unsurpassed levels in imaging of anatomical details and topographical relations by the techniques of computed tomography (CT) and magnetic resonance imaging (MRI) have been achieved. The techniques of positron emission tomography (PET) and of magnetic resonance spectroscopy (MRS), which depict also metabolic and blood flow aspects, provide a refinement of our knowledge on the metabolism, structure and pathophysiological relations of a tumour to the surrounding parenchyma. Recent advances in the recording of function-related changes of the cerebral electro-magnetic field allow a better definition of critical functional areas.

    Description / Table of Contents: Sommario Mediante stimolazione elettrica periodica della via commissurale/collaterale di Schaffer nell'area CA1 dell'ippocampo di ratto è stata indotta una epilessia legata al meccanismo del “kindling”. La natura progressiva del fenomeno è dimostrata da una dettagliata descrizione di segni comportamentali e del progressivo incremento della durata della scarica durante il “kindling”. In aggiunta sono state considerate: l'evoluzione delle alterazioni dei potenziali di campo evocati da coppie di pulsi, le modificazioni del legame recettoriale per i GABAA, l'espressione dell'mRNAs che codifica per le subunità recettoriali del GABAA e Glutamato. Vengono qui presentate le evidenze secondo cui durante il kindling avvengono in CA1 e nella fascia dentata modificazioni opposte in termini di bilancio tra eccitazione ed inibizione dovuti a cambiamenti contrastanti della funzione inibitoria GABA-mediata.
    Notes: Abstract Kindling epileptogenesis was induced by periodic electrical stimulation of the Schaffer collateral/commissural pathway in the CA1 area of the rat hippocampus. The progressive nature of hippocampal kindling is demonstrated by a detailed description of the behavioral signs and the progressive increase of the after-discharge duration in the course of kindling acquisition. Furthermore, the evolution of the alterations in the paired-pulse local evoked field potentials and the modifications of the GABAA receptor binding and of the expression of mRNAs encoding for the subunits of the GABAA and glutamate receptors are considered. Evidence is presented that during kindling opposite changes occur in the CA1 and the fascia dentata in terms of the balance between excitation and inhibition due to contrasting changes in GABA-mediated inhibitory function.
