Notes: Abstract  A histidine-2′-deoxyguanosine hybrid, Phac-Hse(p5′dG)-His-OH (I), was synthesized, and its reaction with cisplatin was monitored by reversed-phase HPLC and 1H NMR. Two new compounds, II and III, were observed to be simultaneously formed, II in larger proportion than III. These products were isolated after HPLC purification and extensively characterized. Both II and III contained platinum, had the same mass and showed a bathochromic displacement of their absorption maxima with respect to that of I. Both remained undegraded upon enzymatic digestion and yielded I when treated with NaCN. From these data and the information provided by 1H NMR analysis, we inferred that II and III were constitutional isomers, in particular chelates in which platinum was coordinated to the N7 of guanine and either the Nπ or the Nτ imidazole nitrogens, respectively. No preference of the metal for either of these N-donors was observed.

Notes: Abstract  To evaluate levels of essential (zinc and copper) and non-essential (mercury and cadmium) heavy metals, 34 species of organisms from different areas close to the Antarctic Peninsula were analysed. These included algae, filter-feeders, omnivorous invertebrates and vertebrates. Mercury was not detected, while cadmium was found in the majority of organisms analysed (detection limit was 0.05 ppm for both metals). The highest cadmium concentration was observed in the starfish Odontaster validus. Anthozoans, sipunculids and nudibranchs showed maximum levels of zinc, while the highest copper level was found in the gastropod Trophon brevispira. Mercury and cadmium levels in fishes were below the detection limit. Concentrations of essential and non-essential metals in birds were highest in liver followed by muscle and eggs. Cadmium and mercury levels in muscle of southern elephant seals were above the detection limit, whereas in Antarctic fur seals they were below it. The objective of the study was to gather baseline information for metals in Antarctic Ocean biota that may be needed to detect, measure and monitor future environmental changes.

Notes: Abstract The effect of 6-benzylaminopurine (6-BA) alone or in combination with naphthaleneacetic acid or indoleacetic acid on the morphogenetic response of cotyledon explants of Citrullus colocynthis (L.) Schrad. was tested. The best results were obtained with a medium containing 25 µm 6-BA, which yielded organogenic calli at a frequency of 81.8%. When these organogenic calli were transferred to elongation medium (basal medium supplemented with 0.5 µm 6-BA), 80% produced well-developed shoots. These shoots rooted normally when cultured on rooting medium containing indolebutyric acid at 2.5 or 5.0 µm. Plants grew to maturity under greenhouse conditions and gave normal fruits. Cotyledon explants were transformed by cocultivation with Agrobacterium tumefaciens LBA4404 carrying the binary vector pBI121 which bears the reporter gene β-glucuronidase (gus) and the marker gene neomycin phosphotransferase (nptII). Transformants were selected for growth capacity on medium with 100 mgl–1 of kanamycin. On the basis of β-glucuronidase expression, the transformation frequency was 14.2%. Molecular characterization by polymerase chain reaction confirmed the presence of the two genes transferred (gus, nptII) in the transgenic plants. Sexual transmission of both genes was also confirmed by studying their expression in progenies from several transgenic plants.

Notes: Summary Physiological and histochemical studies have recently supported the notion that nitric oxide (NO) is the transduction signal responsible for the non-adrenergic, non-cholinergic relaxation of the vasculature as well as the airways of the mammalian lung. We report the presence of immunoreactivity to NO synthase (NOS) in nerve cell bodies and nerve fibres in the neural plexus of the buccal cavity and lungs of the frog, Rana temporaria, using the indirect immunocytochemical technique of avidin-biotin and the NADPH-diaphorase technique. The neural ganglia located next to the muscle layer and within the connective tissue of the buccal cavity were partially immunoreactive for NOS. In the lungs, NOS immunoreactivity occurred in nerve cell bodies, as well as in both myelinated and unmyelinated nerve fibres. Fine nerve fibres immunoreactive to NOS were observed within the muscle fibre bundles and next to the respiratory epithelium. Both the presence of NOS immunoreactivity and the positive histochemical reaction for NADPH-diaphorase in the neural plexus of amphibian respiratory tract suggests a broad evolutionary role for NO as a peripheral neurotransmitter.

Notes: Abstract The aim of the present study is to analyze the morphologic changes occurring along the period of a woman's life thatareknown as climacterium. Our sample consists of 648 women from different provinces of Cuba, but who lived in Havana for at least 15 years. Morphological variables such as height, weight, biliocristal and biachromial diameters and six subcutaneous fat skinfolds were measured. With regard to the menopausic condition, the sample was divided into three subpopulations: premenopausic, naturally menopausic and surgically menopausic (when the two ovaries had been removed). Age at menopause was calculated by both the “retrospective” (49.45±0.49) and “status quo” (48.83±0.02) methods. The results obtained point towards morphological differences between pre- and postmenopausic women. We observed greater accumulations of fatty tissue in the suprailiac regions of postmenopausal females. Lean body mass represents a bigger fraction of the total body weight in women who are still menstruating. In the case of surgically menopausic women, they are at a mid-distance between the premenopausic and naturally menopausic subpopulations. A discriminant analysis was carried out which confirmed the results previously observed. From this analysis it appeared the body mass index and the subcutaneous skinfolds are the variables that discriminante, that is, separate, the three subsamples.
