    Notes: The dynamic behaviour of sediment-laden underflows was examined in Peyto Lake, Alberta, Canada, which contains a midlake sill 7 m high. Sediment-laden underflows are driven by the downslope component of negative buoyant gravity multiplied by the current's thickness. Our measurements of wind, lake currents and water properties indicate that underflows pass over the sill due to the active storage of turbid suspension near the bottom in the deepest proximal region. Sill overflows occurred only when a hydrological threshold of the inflowing river was exceeded, causing quasicontinuous underflow and associated sedimentation in the distal region of the lake basin.

    Notes: Chronic abuse of cocaine or alcohol is associated with structural, neuropathological and cognitive impairments that have been documented extensively. Little is known, however, about neurobiochemical changes in chronic substance abusers.We performed MRI and multi-slice brain proton magnetic resonance spectroscopic imaging (MRSI) to assess neuronal viability (via N-acetylaspartate (NAA)) and white matter metabolite status in 22 4-months-abstinent individuals dependent on crack cocaine only and on both crack cocaine and alcohol. Compared to 11 non-dependent controls we found (1) significantly lower NAA measures in the dorsolateral prefrontal cortex of the combined cocaine-dependent groups; (2) comparable spatial distribution and magnitude of these NAA effects for both cocaine-dependent groups; (3) higher choline-containing metabolites in frontal white matter of individuals dependent on both cocaine and alcohol; (4) absence of brain atrophy in both abstinent cocaine-dependent samples; and (5) partial recovery from prefrontal cortical NAA loss, primarily with abstinence from alcohol. The MRSI findings suggest preferential neuronal damage to the frontal cortex of both cocaine-dependent samples and gliosis in frontal white matter of individuals dependent on both alcohol and cocaine, conditions that persist for more than 4 months of abstinence.

    Notes: This paper celebrates the 21st anniversary of the plane-grating grazing-incidence monochromator, designed at Daresbury in 1974 and in almost continuous operation ever since. The order-sorting principles [Miyake, Kato & Yamashita (1969). Sci. Light (Tokyo), 18, 39–56] are described, along with the detail of the instrument's design, followed by the history of its use on three synchrotron radiation sources (NINA, TANTALUS and SRS). A selection of scientific highlights at the SRS is given, taken from some of the experimental programmes in which the author was involved, concentrating on tunable photon-energy photoemission studies of metallic alloys.     Notes: Abstract A primal - dual simplex variant is presented that incrementally builds up the optimal LPbasis matrix. For network LPs, the incremental primal - dual algorithm can adopt steepest-edgedirections of movement utilizing standard graph data structures without additionalcomputational overhead. Computational results indicate the resulting implementation isuniformly superior to other direction movement schemes, and achieves order of magnitudespeedups versus a primal network simplex code on medium size NETGEN problems. Thesespeedups carry over to the generalized network problem domain as well. Some comparisonsare also made against the relaxation method on NETGEN uncapacitated transportation andtransshipment problems.

    Notes: Summary Elderly patients with end-stage renal disease often remain on dialytic therapy because they are at increased risk for mortality and morbidity. We placed 24 cadaver kidney transplants into 24 patients aged 65–74 years between September 1, 1985, and August 31, 1995. Rates of patient and graft survival were compared with those of 404 concurrent first cadaver-kidney transplant recipients between the ages of 20 and 44 years. The 5-year rates of patient and graft survival were not significantly different (86% versus 92% and 77% versus 63%, respectively; study group presented first). Primary cadaver kidney transplantation can be successfully performed in patients older than 65 years when a selection algorithm is applied to exclude active infection, active malignancy, unsuitable anatomy for technical success, high probability of operative mortality, and noncompliance. Pelvic arteriosclerosis and lower urinary tract abnormalities can cause intraoperative technical problems.

    Notes: Abstract Paclitaxel (Taxol) a clinically active anticancer agent, exerts its cytotoxicity by inducing tubulin polymerization, leading to cellular mitotic block. In contrast, other antimitotic drugs, such as colchicine, podophyllotoxin, and vinblastine, act by depolymerizing microtubules. We report here (a) a semiautomated assay which measures the tubulin-polymerizing activity of paclitaxel analogs and (b) a cellular assay to measure the potential of these compounds to block cells in mitosis. The microtubule-polymerizing assay measured the turbidity of bovine brain microtubule protein (MTP) polymerized by the test compound in a 96-well plate. We maximized the sensitivity of this assay by conducting the polymerization reaction at 20 °C, at which temperature the baseline reaction, i.e. the basic ability of the untreated MTP control to polymerize, was minimal. At 20 °C, the effect of 0.05 μg/ml of paclitaxel on MTP could be detected, whereas at 37 °C, 〉1 μg/ml of paclitaxel was required to detect a significant effect relative to untreated MTP. We describe the analysis of the complex curves of MTP polymerization with varying concentrations of test compounds. The polymerization of microtubules leads to cells being blocked in mitosis. This mitotic blocking effect in intact cells was determined using a cell settling chamber which allowed eight samples to be deposited on a slide. This method required a smaller number of cells (103–105), maintained cell morphology, and allowed for rapid screening of samples. The activity of several new paclitaxel analogs is reported.

    Notes: Abstract We studied intermanual motor transfer for right-to-left or left-to-right direction of transfer between either proximal or distal upper extremity muscle groups. The influence of previously acquired motor engrams (original learning, OL) on learning efficiency of the contralateral side (transfer learning, TL) was examined in 26 right-handed healthy subjects. The task consisted of the drawing of meaningless figures. During TL, OL figures had to be reproduced as vertical mirror reversals. Data revealed a benefit for right-to-left but not left-to-right direction of transfer for time to complete a figure as well as a left-to-right transfer benefit for spatial motor precision. Furthermore, a benefit for intermanual transfer of training between proximal but not distal muscle groups was found when movement time to complete a figure was evaluated. Of special interest was the observation of a disadvantage due to prior contralateral learning for performance at right distal effectors. The asymmetrical transfer benefits with respect to side are in line with previous findings and support the proficiency model and the cross-actiation model. Results further showed that intermanual transfer of training might differ with respect to muscle group involvement and suggest that, although primarily facilitating, previous opposite hand training may lead to inhibitory influences on subsequent contralateral reproduction.
