    Notes: The aim of this study was to evaluate if continuous treatment with budesonide or salmeterol influences the bronchodilator response to terbutaline in children with asthma; 23 children, aged 7 to 16 years (mean = 11 years), with mild asthma were treated with inhaled budesonide 100 μg b.i.d. and placebo for three weeks in a randomized, double blind crossover study. These treatments were followed by treatment with inhaled salmeterol 50 μg b.i.d. for 3 weeks. On the last day of each period a cumulative dose-response experiment with terbutaline in the doses 50, 100, 250 and 500 μg (cumulative dose 900 μg) was performed. Lung function was measured before and 20 min after each terbutaline inhalation. Baseline pulmonary functions after budesonide treatment were significantly higher than the baseline measured after the two other treatments (p 〈 0.05). After budesonide treatment, the dose-response curve was shifted vertically upwards but otherwise parallel to the dose-response curve after placebo. The increase from baseline after the first cumulative dose of terbutaline was significantly lower after salmeterol treatment than after the two other treatments (p 〈 0.01). Maximal lung functions after 900 μg terbutaline also differed significantly between the three dose-response days; budesonide being significantly higher and salmeterol significantly lower than placebo (p = 0.02 and p 〈 0.001, respectively). It is concluded that budesonide treatment does not enhance the brochodilator response to terbutaline. Further studies are needed to assess if long-term continuous salmeterol treatment reduces the response to terbutaline.

    Notes: Abstract In children with asthma, twice daily administration of salmeterol 25 μg, salmeterol 50 μg and salbutamol 200 μg were compared in two, 3-month, double-blind, parallel group studies, one using metered dose inhalers (MDIs), the other using dry powder inhalers (Diskhaler, DPIs). Both studies were continued for a further 9 months during which time exacerbation rates, lung function at the clinic and adverse events were monitored. Similarities in design and methodology of the two studies justified a combined analysis. Eight hundred and forty-seven asthmatic children aged between 4 and 16 (mean 10.1) years, requiring inhaled beta2-agonist treatment were randomised to treatment. After a 2 week run-in when all bronchodilator therapy was withdrawn, 279 patients received salmeterol 25 μg bd, 290 patients salmeterol 50 μg bd and 278 patients salbutamol 200 μg bd. After 3 months' treatment the change from baseline in daily morning and evening peak expiratory flow (PEF) was significantly greater with salmeterol 50 μg bd than with salbutamol 200 μg bd (P〈0.001). Salmeterol 50 μg bd was also significantly better than salmeterol 25 μg bd at improving mean morning PEF (P=0.017) but both treatments had a similar effect on evening PEF. Analysis of variance showed an interaction between baseline PEF less than 100% predicted normal value and treatment outcome. Analysis of this sub-set of patients with lower lung function revealed similar results to the total population although the improvements in PEF from baseline were greater. Data from both studies, showed that the improvement in lung function was maintained throughout 12 months' treatment. Patients receiving salmeterol 50 μg bd had significantly more symptom-free nights (P〈0.01) and a higher percentage of rescue bronchodilator-free days (P=0.01). The incidence of asthma exacerbations was evenly distributed between the three treatment groups and there was no evidence of any change in the rate of occurrence of exacerbations over the 12 month period. Adverse events were no different across treatment groups or across age groups and were primarily related to the patients' disease state.

This paper considers systems of Laguerre-type orthogonal polynomials for which the corresponding Jacobi matrices represent unbounded self-adjoint operators which are bounded above or below. There are twoa priori candidates for an associated orthogonal harmonic analysis: (i) the existence of some subset Λ inR n such that the exponentials {eiλ·x}Λ form anorthogonal basis forL 2(μ); and (ii) the existence of a certaindual pair of representations of theC *-algebraO N wheren is the cardinality of the setB. (For eachN, theC *-algebraO N is known to be simple; it is also called the Cuntz algebra.) We show that, in the “typical” fractal case, the naive version (i) must be rejected; typically the orthogonal exponentials inL 2(μ) fail to span a dense subspace. Instead we show that theC *-algebraic version of an orthogonal harmonic analysis, namely (ii), is a natural substitute. It turns out that this version is still based on exponentialse iλ·x, but in a more indirect way. (See details in Section 5 below.) Our main result concerns the intrinsic geometric features of affine systems, based onR andB, such that μ has theC *-algebra property (ii). Specifically, we show that μ has an orthogonal harmonic analysis (in the sense (ii)) if the system (R, B) satisfies some specific symmetry conditions (which are geometric in nature). Our conditions for (ii) are stated in terms of two pieces of data: (a) aunitary generalized Hadamard matrix, and (b) a certainsystem of lattices which must exist and, at the same time, be compatible with the Hadamard matrix. A partial converse to this result is also given. Several examples are calculated, and a new maximality condition for exponentials is identified.

Stimulation of Ehrlich ascites tumor cells with leukotriene D4 (LTD4) within the concentration range 1–100 nm leads to a concentration-dependent, transient increase in the intracellular, free Ca2+ concentration, [Ca2+] i . The Ca2+ peak time, i.e., the time between addition of LTD4 and the highest measured [Ca2+] i value, is in the range 0.20 to 0.21 min in ten out of fourteen independent experiments. After addition of a saturating concentration of LTD4 (100 nm), the highest measured increase in [Ca2+] i in Ehrlich cells suspended in Ca2+-containing medium is 260 ± 14 nm and the EC50 value for LTD4-induced Ca2+ mobilization is estimated at 10 nm. Neither the peptido-leukotrienes LTC4 and LTE4 nor LTB4 are able to mimic or block the LTD4-induced Ca2+ mobilization, hence the receptor is specific for LTD4. Removal of Ca2+ from the experimental buffer significantly reduces the size of the LTD4-induced increase in [Ca2+] i . Furthermore, depletion of the intracellular Ins(1,4,5)P3-sensitive Ca2+ stores by addition of the ER-Ca2+-ATPase inhibitor thapsigargin also reduces the size of the LTD4-induced increase in [Ca2+] i in Ehrlich cells suspended in Ca2+-containing medium, and completely abolishes the LTD4-induced increase in [Ca2+] i in Ehrlich cells suspended in Ca2+-free medium containing EGTA. Thus, the LTD4-induced increase in [Ca2+] i in Ehrlich cells involves an influx of Ca2+ from the extracellular compartment as well as a release of Ca2+ from intracellular Ins(1,4,5)P3-sensitive stores. The Ca2+ peak times for the LTD4-induced Ca2+ influx and for the LTD4-induced Ca2+ release are recorded in the time range 0.20 to 0.21 min in four out of five experiments and in the time range 0.34 to 0.35 min in six out of eight experiments, respectively. Stimulation with LTD4 also induces a transient increase in Ins(1,4,5)P3 generation in the Ehrlich cells, and the Ins(1,4,5)P3 peak time is recorded in the time range 0.27 to 0.30 min. Thus, the Ins(1,4,5)P3 content seems to increase before the LTD4-induced Ca2+ release from the intracellular stores but after the LTD4-induced Ca2+ influx. Inhibition of phospholipase C by preincubation with U73122 abolishes the LTD4-induced increase in Ins(1,4,5)P3 as well as the LTD4-induced increase in [Ca2+] i , indicating that a U73122-sensitive phospholipase C is involved in the LTD4-induced Ca2+ mobilization in Ehrlich cells. The LTD4-induced Ca2+ influx is insensitive to verapamil, gadolinium and SK&F 96365, suggesting that the LTD4-activated Ca2+ channel in Ehrlich cells is neither voltage gated nor stretch activated and most probably not receptor operated. In conclusion, LTD4 acts in the Ehrlich cells via a specific receptor for LTD4, which upon stimulation initiates an influx of Ca2+, through yet unidentified Ca2+ channels, and an activation of a U73122-sensitive phospholipase C, Ins(1,4,5)P3 formation and finally release of Ca2+ from the intracellular Ins(1,4,5)P3-sensitive stores.

    Notes: Abstract  Cephalometry was used to detect patterns of cranial growth in fetal bats that have been differentially stained for bone and cartilage. Rhinolophoid bats exhibit elaborate nasal cavities with coincidental distortions of the maxilla. The expansion of these cavities creates paired auxiliary fontanels among the nasal, maxillary, and frontal bones. This distortion of the rostrum is also associated with the loss of the lacrimal bones and the modification of the infraorbital foramen into a shallow canal. The use of the head as an acoustical horn is discussed with reference to the ontogeny of echolocation.

    Notes: Abstract The toxic effect of copper to coastal phytoplankton communities was studied in 6 m3 enclosures during 15 days using the PICT (Pollution-Induced Community Tolerance) methodology. PICT, primary production, algal biomass, species composition and diversity in phytoplankton communities were measured in control enclosures and enclosures with 1, 3, 6 and 15 μg Cu l-1 added. Increased copper tolerances were induced in the enclosures with added copper and, related to the pooled mean value of the controls, the mean values for copper tolerance were significantly higher in enclosures with 1, 6 and 15 μg Cu l-1. In enclosures with 6 and 15 μg Cu l-1, photosynthetic activity was significantly depressed. Fundamental changes in taxonomic diversity and species composition occurred in enclosures with 15 μg Cu l-1. Algal biomass was not affected by the addition of copper. The distribution and concentration of copper in the enclosures showed that copper exposure was almost constant over time in the enclosures and was proportional to the amount of copper added to them. Our studies indicate that the PICT methodology is sensitive enough to detect even minor effects of copper on phytoplankton communities. The PICT measurements indicate effects from concentrations as low as 1 μg Cu l-1 in highly eutrophic coastal areas, where the bioavailability of copper is assumed to be low. Copper concentration in the fjord and control enclosures was high, about 0.5 μg Cu l-1, only a factor two below the effect concentration measured by PICT. These results are significant in view of the increased release of copper to the coastal marine environment as results of the substitution of copper-based ship paints for tri-n-butyltin (TBT) antifouling paints.
