    Notes: malR of Streptomyces coelicolor A3(2) encodes a homologue of the LacI/GalR family of repressor proteins, and is divergently transcribed from the malEFG gene cluster, which encodes components of an ATP-dependent transport system that is required for maltose utilization. Transcription of malE was induced by maltose and repressed by glucose. Disruption or deletion of malR resulted in constitutive, glucose-insensitive malE transcription at a level markedly above that observed in the parental malR+ strain, and overproduction of MalR prevented growth on maltose as carbon source. Consequently, MalR plays a crucial role in both substrate induction and glucose repression of maltose utilization. malR is expressed from a single promoter with transcription initiating at the first G of the predicted GTG translation start codon.

    Notes: Abstract Hereditary neuralgic amyotrophy (HNA) is an autosomal dominant, recurrent focal neuropathy characterized by episodes of painful brachial plexus neuropathy with muscle weakness and atrophy, as well as sensory disturbances. Single episodes are commonly preceded by unspecific infections or immunization, or are associated with parturition. Minor facial dysmorphic features are present in some pedigrees but do not clearly segregate with the disease. To confirm the recently described HNA locus on distal chromosome 17q, we performed a genetic linkage study in an extended Turkish pedigree. We were able to refine the HNA locus on chromosome 17q24–q25 in a 16-cM region.

    Notes: Abstract In this paper we explore the dominant position of a particular style of scientific modelling in the provision of policy-relevant scientific knowledge on future climate change. We describe how the apical position of General Circulation Models (GCMs) appears to follow ‘logically’ both from conventional understandings of scientific representation and the use of knowledge, so acquired, in decision-making. We argue, however, that both of these particular understandings are contestable. In addition to questioning their current policy-usefulness, we draw upon existing analyses of GCMs which discuss model trade-offs, errors, and the effects of parameterisations, to raise questions about the validity of the conception of complexity in conventional accounts. An alternative approach to modelling, incorporating concepts of uncertainty, is discussed, and an illustrative example given for the case of the global carbon cycle. In then addressing the question of how GCMs have come to occupy their dominant position, we argue that the development of global climate change science and global environmental ‘management’ frameworks occurs concurrently and in a mutually supportive fashion, so uniting GCMs and environmental policy developments in certain industrialised nations and international organisations. The more basic questions about what kinds of commitments to theories of knowledge underpin different models of ‘complexity’ as a normative principle of ‘good science’ are concealed in this mutual reinforcement. Additionally, a rather technocratic policy orientation to climate change may be supported by such science, even though it involves political choices which deserve to be more widely debated.

    Notes: The interaction of biotinylated human interleukin 5 ([BT]hIL5) with immobilized receptor was measured with a real-time biosensor, and these results were used as a basis for configuring as ELSIA for screening antagonists of hIL5-receptor binding. The recombinant proteins used, hIL5 and shIL5Rα-Fc (chimeric fusion receptor constructed by linking the soluble component of the hIL5 receptor α subunit to the constant domain (Fc) of immunoglobulin G), were produced by the expression of cloned vectors in Drosophila schneider (S2) cells. Initial attempts to develop a screening assay by direct immobilization of soluble IL5 receptor to microtiter plates proved unsatisfactory and led to use of theFc chimera attached by oriented immobilization via protein A. Hence, shIL5Rα-Fc was bound to protein A covalently immobilized on a carboxymethyl dextran (CM-5) biosensor chip. Specific binding was demonstrated of [BT]hIL5 to protein A/shIL5Rα-Fc receptor complex. The binding was high affinity (Kdapp=6 nM), reversible and saturable. The affinity of [BT]hIL5 was similar to that determined with the biosensor assay for unmodified hIL5. The observed kinetics of the interactions of Fc chimera with protein A (slow dissociation) and of [BT]hIL5 with immobilized Fc chimera (faster dissociation) were favorable for subsequently establishing plate based ELISA assay. In the latter, Fc chimera was immobilized to the plate via protein A as in the biosensor experiment. Binding of [BT]hIL5 to immobilized Fc chimera in the ELISA was concentration dependent and was competed by both hIL5 and shIL5Rα. The results demonstrate a novel use of real-time biosensor technology as a macromolecular interaction analysis tool to help develop other assay using immobilized ligands, including high throughout screening assays to identify antagonist leads. The biosensor technology also offers a means to evaluate the mechanism of action of lead compounds by competition binding, as well as by direct binding if the leads are large enough or can be immobilized to the sensor surface.
