    Notes: Summary A second prenatal diagnosis of severe haemophilia B was carried out in a family with no prior history of the disease. The first prenatal diagnosis was based on linkage analysis and showed the male fetus not to be affected because he had inherited the same X-chromosome as his healthy brother. Carrier status in the female at risk could not be assessed by restriction fragment length polymorphisms (RFLPs). She was found to have inherited the same marker constellation as her affected brother. However, due to the fact that a pedigree with no prior history of haemophilia B has been examined diagnosis was impossible. In addition factor IX coagulant and antigen values gave no definitive clue to a haemophilia B carriership. The problems with RFLP analysis in this pedigree were circumvented by polymerase chain reaction (PCR) based direct sequencing of the factor IX gene. A previously unknown mutation could be detected in patient haemophilia B (Kleve) and the carrier status in the female at risk could be confirmed. The second prenatal diagnosis showed that the male fetus had inherited the mutation and will therefore be afflicted with haemophilia B.

    Notes: A simple, rapid sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) method is presented for isolating the α, α' and β subunits of rabbit muscle phosphorylase kinase. The SDS-PAGE procedure can yield milligram amounts of α and β from a single preparative gel and also allows isolation of the α' isozyme free of α. Notably the method provides the purified subunits in a form amenable to structural analysis. Edman degradation of α and α' reveal identical NH2-terminal structures. Amino acid analysis of the electrophoretically purified α and β subunits are in good agreement with their deduced primary structures. The amino acid sequence of 488 residues in α and 713 residues in β were determined by gas phase Edman degradation. The data support the recently deduced primary structures of α (Zander et al., Proc. Natl. Acad. Sci. USA 1988, 85, 2929-2933) and of β (Kilimann et al., Proc. Natl. Acad. Sci. USA, 1988, 85, 9381-9385).

Die Suspension der Fasern in Nujol resultierte in einer verbesserten Auflösung der Spektren. Eine näherungsweise Quantifizierung des Substitutionsgrades wurde durch die Anwendung einer Entfaltungsfunktion im Absorptionsgebiet der OH-Valenzschwingungen erreicht. Die relative Bandenhöhe bei 3500 bis 3535 cm-1 erwies sich als ein Maß für den Nitrierungsgrad der Cellulosefasern. Der Einfluß der Ausgangscellulose sowie der Nitrierungsmethode und von Vorbehandlungen auf die Spektren von Einzelfasern wurde untersucht.
    Notes: Cellulose nitrate fibres of different degrees of substitution were analyzed by micro-FTIR technique. A suspension of the fibres in Nujol resulted in improved recording conditions. An approach to quantification of the degree of substitution was reached by applying a deconvolution function to the absorbance range of the OH valency vibrations. The relative band height at 3500 to 3535 cm-1 was found to be a measure of the nitration degree of cellulose fibres. The influences of the original cellulose as well as the kind of nitration and pretreatment on the spectra of individual fibres were studied.
