    Notes: Summary Meningiomas of the third ventricle are rare intracranial neoplasms. We reported such a case in a 42 years old man without clinical evidence of increased intracranial pressure. Computed tomography (CT) and magnetic resonance imaging (MRI) demonstrated the tumour sitting in the superior and anterior part of the third ventricle, bulging into the lateral ventricles. CT was more effective than MRI in the demonstration of calcifications whereas MRI proved to be superior in delineation of the tumour and its relation with surrounding structures.

    Notes: Abstract Fourteen hypertensive patients hospitalized in a paediatric intensive care unit were studied to evaluate safety and hypotensive efficacy of intravenous nicardipine. Systolic and diastolic blood pressure significantly decreased 1 h after the beginning of the treatment (1 μg/kg per minute). Mean decrease in systolic blood pressure during the first 24 h was between 9.9% and 13.4% of the initial value. Mean lowering of diastolic blood pressure was between 16.7% and 25.6%. Nicardipine did not significantly affect heart rate with dose of 1 μg/kg per minute. No clinical side-effects were observed. Nicardipine could be a first line drug for the treatment of hypertension in paediatric intensive care units.

    Notes: Abstract A NDO approximate procedure based on the indirect intrinsic ab initio localization method of von Niessen is developed. It is shown that only when the NDO approximations are introduced at the two electron level, expressions are obtained which are the charge density counterpart of those found in the approximate energy localization methods. The results of these two methods are quite similar both in the CNDO and INDO approximations. The indeterminacies observed in the CNDO localization for unsaturated systems and for molecules with two or three lone pairs on the same atom, are removed by localizing up to an INDO level. The approximate charge density localization is however computationally much easier than the approximate energy localization method and should be more appropriate in LMO studies of large organic molecules.

    Notes: Abstract The use of single determinantal approximate molecular wavefunctions of the LCAO MO NDO type for the calculation of the momentum densityρ(p) and the radial momentum density distributionJ(p) is discussed. In each case, these expressions should be orientationally invariant and the momentum density should be normalized. Combining these two requirements, it is shown that only two approximations are physically significant: (1) NDO wavefunctions are used andρ(p) andI(p) are approximated respectively up to an INDO and a CNDO level; (2) Overlap integrals are explicitly taken into account when solving the Roothaan SCF equations or deorthogonalized NDO functions are employed, together with the unapproximated expressions.

    Notes: Abstract In this study we have compared the characteristics of six labelling kits for the preparation of technetium-99m labelled human serum albumin (99mTc-HSA) and evaluated the usefulness of the various 99mTc-HSA preparations as blood pool tracer agents. The amount of the principal ingredients, i.e. HSA and stannous ions, varies largely between the studied kits and this is probably a reason for the observed differences in the labelling rate. Analysis of the reaction mixtures after labelling of the respective kits with 99mTc showed in each preparation the presence of four to five radioactive components in variable relative amounts. The retention time of the main component on size-exclusion high-performance liquid chromatography (SEC-HPLC) was identical for all preparations. Biodistribution of the HPLC-isolated fractions was studied in mice. The components with the shortest and longest retention times on HPLC show poor retention in the plasma. The three intermediate fractions, including the principal peak, are initially retained relatively well in the blood (60%–70% of the injected dose after 10 min), but clearly to a lower degree than iodine-125 labelled HSA. Moreover, they diffuse out of the vascular compartment at a much higher rate than 125I-HSA. The biological behaviour of the main component of the various preparations was clearly different, despite the identical retention time on SEC-HPLC. Study of the total preparations in mice and a rabbit showed that two of them are cleared rapidly from the blood and cannot be considered valuable blood pool tracers. Diffusion of the other preparations out of the blood is slower but also considerable and compromises their use for ventriculography.

    Notes: Abstract Technetium-99m labelled red blood cells (99mTc-RBCs) are far superior to 99mTc-labelled human serum albumin (99mTc-HSA) for radionuclide ventriculography, but their labelling is more complex, time consuming and risk bearing (in vitro labelling) or suffers from interference by some medications (in vivo labelling). We have now modified HSA by the introduction of mercapto groups with the purpose of preparing stable and practical 99mTc-mercaptoalbumin with long retention in the vascular system, that could replace 99mTc-RBCs. HSA was incubated with N-succinimidyl S-acetylthioacetate (SATA) or N-succinimidyl 2,3-di(S-acetylthio) propionate (SATP) to introduce a chain containing one or two protected sulfhydryl groups on some of the lysine amino groups. After purification by size-exclusion chromatography (SEC), the mercapto groups were deprotected by incubation at alkaline pH or by treatment with hydroxylamine. The reaction products were used with or without SEC purification for direct or exchange labelling experiments with 99mTc at neutral pH. SEC-HPLC was used to determine labelling yields and to isolate pure 99mTc-mercaptoalbumin. Stable 99mTc-mercaptoalbumin complexes could be formed in 90%–95% yield after coupling albumin with SATA or SATP in all molar ratios used followed by deacetylation in one of the mentioned conditions. The most favourable results were obtained after reaction of SATA or SATP with HSA in a 25: 1 ratio and deprotection with NH2OH. The stability of the resulting 99mTc-mercaptoacetyl-albumin (99mTc-MAHSA) and 99mTc-dimercaptopropionyl-albumin (99mTcDMP-HSA) and their retention in vivo in plasma of mice and rabbits are clearly higher than that of conventional 99mTc-HSA preparations. 99mTc-DMP-HSA approaches the behaviour of 125I-HSA quite well in both animal species. A preliminary study with 99mTc-DMP-HSA in a volunteer showed a retention in the vascular compartment almost identical to that of 99mTc-RBCs and clearly higher than that of a common 99mTc-HSA preparation. The results indicate that these 99mTc-mercaptoalbumins and especially 99mTc-DMP-HSA are very promising as a practical alternative to 99mTc-RBCs.

    Description / Table of Contents: Zusammenfassung Die von Wurziger in den Wachsauflagerungen von Rohkaffee gefundenen Carbonsäure-5-hydroxytryptamide (CHT) dienen als Indicatorsubstanzen für eine erfolgte Behandlung der Rohbohnen, die zu einem sogenannten bekömmlichen, coffeinhaltigen Röstkaffee führt. Ihre Analytik wird erörtert und eine verbesserte Methode, anwendbar auf Roh- und Röstkaffee, vorgestellt. Die für die Aufstellung einer Eichkurve erforderlichen Substanzen wurden aus Kaffee gewonnen und die Eichgerade durch eine analoge Mischung synthetischer CHT-Homologen bestätigt. Die massenspektroskopischen Ergebnisse dieser CHT-Substanzen und ihrer Benzyläther werden diskutiert. Es zeigte sich, daß die Analysenwerte gegenüber der bisher üblichen Methode um ca. 40% bei sogenannten reizarm veredelten Kaffees niedriger liegen. Damit sinkt die bisher in der Praxis übliche Grenze für behandelte Kaffees von 400 auf 250 ppm CHT. Die aus Wiederhol- und Vergleichsanalysen ermittelten Werte der Fehlerstreuung werden angegeben.
    Notes: Abstract The carboxylic acid 5-hydroxytryptamides found by Wurziger in the wax of the surface layer of green coffee beans are indicator compounds for detecting a treatment of the green beans, leading to an easily digestible coffee beverage with full caffeine content. The analytical methods are discussed and an improved method applicable to green and roasted coffee is presented. The standard compounds, necessary for calibration have been isolated from coffee wax and the calibration curve was confirmed with a mixture of some synthetic CHT-homologues. The mass spectrometric data of these CHT and their benzyl ethers are discussed. Contrarily to the usual methods the analytical results were reduced to about 40% when applied to treated coffee. Thus, the usual limit for processed coffees decreases from 400 to 250 ppm CHT. The errors of the analytical methods (repeatability and reproducibility) are presented.
