Notes: We have observed a persistent photoconductivity effect (PPC) in C70 films well above room temperature. The effect, which consists of a metastable increase in dark conductivity caused by brief exposure to light, exhibits characteristics very similar to the PPC previously observed in C60 films. These characteristics include a magnitude of the effect independent of the light exposure temperature, the existence of a PPC even at 520 K, and the inability of subgap illumination to create or quench PPC. A common origin for the PPC in C70 and C60 seems plausible, and may be related to the distortions the fullerene molecules experience when charged.

Notes: Abstract. Symbiotic germination and development in vitro of Dactylorhiza majalis seeds with a strain of Rhizoctonia is very temperature dependent. Above an optimum at 23–25 °C there is a marked decline in germination percentage. Seeds that did germinate at higher temperatures had only little or no development of mycorrhiza, and developed few or no rhizoids compared with seedlings raised at optimal or lower temperatures. Six-week-old seedlings grown for additional 4 weeks on a range of temperatures had an optimal length increase at 23–24.5 °C mean temperature. At superoptimal temperatures (26 °C), the seedlings contained smaller starch reserves than those at lower temperatures and increased about as much in length as seedlings grown at 13 °C but much less than those grown at optimum. Temperature also influenced the differentiation of the leafy shoot, seedlings growing to a larger size before shoot initiation in the temperature range of optimal growth. Because of the small span between optimal and too-high temperatures, a careful assessment of temperature optimum will be necessary in any orchid/fungus relationship before judging the success of symbiosis. At optimal temperature, symbiotic germination gave a germination percentage about twice that using a good asymbiotic method. The increase in seedling length was about 45% per week in symbiotic culture compared with less than 30% in the asymbiotic culture.

Notes: The localization of fetal antigen 2 (FA2), a recently described basement membrane (BM) associated antigen, was studied by immunohistochemical techniques in 21 basal cell carcinomas (BCC). In both superficial and infiltrating BCC, FA2 was located in a broad diffuse band around the tumor elements and in close contact with the BM. Compared to normal skin, a more extensive distribution of FA2 was seen in BCC. In the infiltrating BCC, FA2 staining was also present in the interstitial stroma between the tumor islands. FA2 was absent in areas with inflammatory cell infiltrates and elastoid degeneration. Epithelial and tumor cells were FA2 negative. The distribution of FA2 was clearly different from that of laminin and collagen type 4. Collagen type 4 and laminin were present as a continuous linear band corresponding to the BM surrounding the tumors. The close contact to the BM and the increased content of FA2 in the reactive stroma around BCC suggest that FA2 is involved in the matrix and/or BM changes taking place during tumor growth and invasion.

Notes: Gastrointestinal pH in 11 healthy subjects with ileostomy was determined with a pH-sensitive, radiotransmitting capsule. Median pH was 7.0 in duodenum, dropped to pH 6.3 in the proximal part, but rose to 7.3 in the distal part of the small intestine. In five subjects the pH of the ileostomy effluents was determined with the pH capsule as well as with a pH meter. Median pH was 7.2 measured with the capsule and 7.4 with the pH meter. The difference between the results obtained with the two methods ranged from 0.1 to 0.3 pH units. The median gastrointestinal transit time of the capsule was 10.5 h (range 6.2–12.8 h). Gastric residence time was 0.6 h (range 0.2–3.8 h), and small intestinal transit time was 10.3 h (range 5.6–11.9 h). Thus the small intestinal transit time in ileostomates is slightly increased compared with values reported from studies on subjects with intact gut. However, no statistically significant correlation was found between the small intestinal transit time and the time elapsed after the creation of the ileostomy. We conclude that colectomy does not alter small intestinal pH but seems to increase the small intestinal transit time of single units.

Notes: Background: Knowledge of the bioavailability of 5-aminosalicylic acid (5-ASA, mesalazine) from the different 5-ASA-containing drugs is important for rational therapy of inflammatory bowel diseases. Methods: The local and systemic bioavailability of 5-ASA from a controlled release 5-ASA preparation (Pentasa—2, 4 or 6 g/day) was investigated and compared with the azo-bond 5-ASA preparation olsalazine (Dipenturn—2 g/day) in 13 healthy volunteers during steady state conditions. Results: The therapeutically relevant parameter of 5-ASA at the rectal level, expressed as the mean concentration in faecal water, showed a significant trend towards higher concentrations with increasing Pentasa dose: 9.2 mmol/L, 19.0 mmol/L and 24.4 mmol/L, respectively. The concentration of olsalazine 2 g/day was 16.0 mmol/L. The concentration of the metabolite N-acetyl-5-aminosalicylic acid (Ac-5-ASA) did not rise with increasing Pentasa dose, indicating saturable presystemic acetylating capacity of 5-ASA. Total urinary excretion of 5-ASA and Ac-5-ASA, as a percentage of the daily ingested 5-ASA dose, remained constant on the three Pentasa doses, but there was a significant increase in the 5-ASA fraction. Mean steady state plasma concentrations of 5-ASA and Ac-5-ASA were significantly higher on Pentasa 4 g/day and 6 g/day than on 2 g/day. Values on Pentasa 2 g/day were comparable with those on olsalazine 2 g/day. Conclusions: The study confirmed that 5-ASA is released from Pentasa in a predictable manner, the amount released increasing with dose. Olsalazine is an excellent generator of 5-ASA in the colon.

Notes: Glucose isomerase from Streptomyces murinus has been crystallized in space group P41212, cell dimensions a = b = 137.65 and c = 132.20 Å. One dimer of the tetrametric molecule is found per asymmetric unit. An initial structure solution was obtained by the molecular replacement method. The crystallographic refinement was performed using molecular dynamics techniques with X-ray restraints. The final crystallographic R value is 21.4% at 2.6 Å resolution including 3023 non-H atoms, two metal ions and two water molecules per monomer.

Notes: Germination percentage of Dactylorhiza majalis (Rchb.f.) Hunt & Summerh. (Orchidaceae) was increased by illuminating surface-sterilized, rinsed and incubated seeds with white light in 16 h photoperiods. Optimal exposures (10–14 days) raised germination from 40 to 75%. Longer light treatments resulted in reduced germination percentage and smaller seedlings. Only about 2% of the seeds could actually germinate in photoperiods; the germination of the rest was delayed by initial light and required about 14 days in constant darkness. Interruption of this darkness period with two consecutive photoperiods increased the germination percentage when the interruption occurred before day 8, but not when occurring later than that.

Notes: Summary Immunohistochemical staining techniques used on an 11-week-old fetus showed that fetal antigen 2 (FA2) was present intracellularly in endochondral and perichondral osteoblasts, and the immunoreaction was extended into the adjacent bone matrix. Osteoclasts and chondroblasts were found to be FA2 negative. A granular perinuclear intracytoplasmic FA2 immunoreaction was found in cultured osteoblasts and osteogenic osteosarcoma cells, and immunoelectron-microscopical examination revealed a granular immunoreaction product in the rough endoplasmic reticulum. These findings indicate that FA2 is synthesized by osteoblasts and osteogenic osteosarcoma cells. A reaction of immunological identity was found between FA2 purified from second trimester amniotic fluid and serum-free supernatants of cultured osteogenic osteosarcoma cells. This shows that an antigen recognized by the anti FA2 antibody is secreted by these malignant cells. Thus, FA2 may represent a marker for altered bone metabolism, and have a potential in the classification of osteogenic osteosarcoma/ chondrosarcoma.
