    Notes: Theoretical models describing the dynamics of multicomponent polymer mixtures are reviewed. Some detailed derivations are shown to make the basic assumptions clear and to ease the comparison between these models. The effects of random noise and memory functions on the time evolution of dynamical correlation functions are examined. The long-range hydrodynamic interactions or mode-coupling effects are also included. A possible extension of these models to the kinetics of spinodal decomposition in solutions of polymer mixtures is briefly discussed.

    Notes: The time evolution of the static structure factor following a sudden temperature jump ΔT is investigated in the case of a symmetric mixture of two polymers and a solvent in the optical theta conditions. The two reverse processes corresponding to ΔT 〉 0 and ΔT 〈 0 are considered, and the kinetics of approach towards the final states are investigated both in the Rouse limit and in the presence of hydrodynamic interactions.

    Notes: We present new results on the behaviour of single chains in homo-and copolymer blends which are beyond the rendom phase approximation (RPA). The radius of gyration of a chain in the blend was found to depend strongly on the interactions as well as the effective diffusion constant of a single chain in the blend or copolymer melt. Note that these results cannot be obtained in the original formulation of the RPA. They are based on the effective Hamiltonian of the chain in the medium. In the equation found for the self diffusion constant of a single chain, Ds, the second term is directly the value of the effective potential at zero wave vector. Thus the measurement of Ds would detect the screened potential at q = 0. Experimental verification of these results is in progress.

    Notes: The static scattering properties of weakly charged copolymers in solution are investigated. The cases of symmetrical and nonsymmetrical diblocks are considered and various structure factors calculated. The electrostatic interaction is introduced via a combination of the random-phase approximation and the Debye-Hückel approximation. Several cases of charge distributions are considered. In particular, diblock copolymers made of neutral and charged monomers and oppositely charged monomers are considered. All the cases deal with systems in the homogeneous disordered phase where no mesophase structures or micelle formations can be observed.

    Notes: The use of plasma deposition to introduce sulfonate groups to the surface of a polyurethane was attempted. In previous work, the bulk incorporation of sulfonate groups was found to improve the blood contacting properties of the base polyurethane but physical properties in the hydrated state were adversely affected. Plasma deposition schemes involving ammonia and sulfur dioxide were utilized in an attempt to incorporate sulfonate groups. Surface characterization by X-ray photoelectron spectroscopy (XPS) and contact angle measurements was used to follow polymer surface rearrangement dynamics and to address the issue of plasma chemistry specificity. Concerns of reaction specificity were alleviated by using the plasma as a pretreatment which is followed by a chemical surface derivatization.

    Notes: A combined theoretical and experimental study of the binding and interaction of valproic acid (VPA) with the bacterial cytochrome P450cam enzyme and the determination of regio- and stereoselective hydroxylation product distribution was performed. From the experiments, C4—;OH VPA was found to be the predominant hydroxylation product with a small amount of C5—OH VPA formed. The experimental stereoselectivity of hydroxylation was 2R4S 〉 ∼ 2S4R 〉 2R4R 〉 ∼ 2S4S and 2S5 〉 ∼ 2R5. The overall goals of the theoretical study were twofold: (1) to characterize as completely as possible, using energy optimization and molecular dynamics simulations, the interactions of flexible ligands with their target proteins, and (2) to determine the extent to which these results could be used to develop criteria to predict or explain the experimentally observed regio- and stereoselectivity of hydroxylation of the flexible ligands. Among the useful insights into the behavior of flexible ligands upon binding to their traget proteins obtained are (1) a large change in conformation occurs for many conformers of VPA upon binding to P450cam, (2) low- energy conformers of VPA do not necessarily lead to optimum interactions with the target protein, and (3) the most favorable mode of interaction of this flexible ligand with the protein binding site has been identified and found to be a result of strong electrostatic interactions between VPA and both Tyr96 and Asp297. For the study of the hydroxylated VPA products, the challenging aspect of this problem was to determine criteria for weighing the contribution of each of the possible protein-ligand complexes. To this end, various possibilities were examined and compared with the experimental results. No single complex was found to reproduce the observed experimental regio- and stereoselectivity. This result indicates that more than one bioactive form of VPA contributes to its oxidation. Results most consistent with experiment are obtained by using the interaction energy of the protein-ligand complex as a criterion for including its contribution to product formation. Although there are remaining disparities between predicted and observed product distributions, the combined theoretical and experimental effort has led to insights into the modes of interaction of this flexible ligand that lead to its observed product specificity. © 1993 John Wiley & Sons, Inc.

    Notes: The adhesion and detachment of platelets were studied on glass coatings of a series of copolymers of hydroxyethyl methacrylate (HEMA) and ethyl methacrylate (EMA). Observations of the interactions of mepacrine labelled washed platelets with these surfaces from a flowing (500 s-1 wall shear rate) suspension in Tyrode's solution containing albumin and red cells were made with epifluorescent video microscopy (EVM). Total platelet adhesion, including platelets which adhere on first contact and platelets which attach temporarily before adhesion, and the number of detaching platelets were minimal for the 0 and 20% EMA copolymers, reached a maximum for the 50% EMA copolymer and showed reduced values for the 80% and 100% EMA copolymers. For the 50, 80, and 100% EMA copolymers, the adhesion values expressed, as a percentage of total contacting platelets, were not different. Albumin adsorption to these copolymers shows a continuous increase from the 0% to the 100% EMA copolymer. It is likely that the peak in platelet adhesion at the 50% EMA composition is related to: low protein adsorption on the 0 and 20% EMA copolymers, too little albumin adsorption to block adhesion on the 50% EMA copolymer, and full-scale blocking on the 80 and 100% EMA copolymers due to greater albumin adsorption.

    Notes: The glow discharge plasma deposition (GDPD) of tetraethylene glycol dimethyl ether is introduced as a novel method for obtaining surfaces that are resistant to protein adsorption and cellular attachment. Analysis of films by x-ray photoelectron spectroscopy and several biological assays indicate the formation of a foulingresistant, PEO-like surface on several substrata (e.g., glass, polytetrafluoroethylene, polyethylene). Adsorption of 125I-radiolabelled proteins (fibrinogen, albumin and IgG) from buffer and plasma was very low (typically 〈20 ng/cm2) when compared to the untreated substrata, which exhibited much higher levels of protein adsorption. Not all coated substrata adsorbed equal amounts of protein (e.g., coated glass samples typically adsorbed more protein than coated polyethylene or coated polytetrafluoroethylene samples), suggesting that the substratum used may affect the amount of protein adsorbed. Measurement of dynamic platelet adhesion, using epifluorescent video microscopy, and endothelial cell attachment further demonstrates the short-term nonadhesiveness of these surfaces.

Observations of the interactions of acridine-orange-labeled washed platelets with these surfaces from a flowing (500 s-1 wall shear rate) suspension in Tyrode's solution containing albumin and red cells were made with epifluorescent video microscopy (EVM). In some cases preadsorbed materials were incubated for 24 h, during which little or no loss of protein occurred. Protein surface concentration, by itself, was a poor indicator of expected cell adhesion and morphology. Surface chemistry was a second important factor which must be considered. A third observation is that for the 100% EMA copolymer, 24 h of incubation led to a large reduction in platelet adhesion when compared to the 100% EMA material without incubation. For the 0% and 100% EMA polymers, the percentage of contacting platelets which adhere and detach is greater for the 24-h incubation cases than for those not incubated. These results led to the conclusion that our most hydrophilic surface favors adhesion with detachment, transient cell contact, over long-term adhesion, as does incubation of adsorbed protein. A brief discussion is presented of a possible connection between this behavior and platelet consumption in viuo for hydrogels.
