    Notes: Acylated B-ring substituted anthocyanins with a 41-keto structure may be of interest as food colorants because at pH values above 4.0 they show an extra absorption band at 583 nm. This makes the pigments highly colored at pH 〉4.0. The extra band also provides a simple method of analysis based on the difference between the absorption at 583 nm at pH 5.5 and 2.0. The Delta E 1% 1 cm for the major anthocyanin in Tradescantia pallida was 97.

    Notes: The acylated, B-ring substituted anthocyanins of Tradescantia pallida (TPA) were extracted from the leaves with acidic water, purified on a Duolite-861 resin and incorporated into a model beverage. The beverage was composed of a citrate-phosphate buffer at pH values 3.5, 4.5 or 5.5, and 15% sucrose. Comparison samples contained cyanidin-3-glucoside from blackberries and a commercial sample of enocyanin from grapes. The TPA samples were much more stable than the other two, particularly at the higher pH values as judged by pigment retention and tristimulus color readings. The extra absorption band of the TPA samples at pH 5.5 indicated the samples were well colored. Addition of tannin and/or ascorbic acid resulted in marked pigment and color loss.

    Notes: Tradescantia pallida (syn. Setcreaseapurpurea) contained two major anthocyanins. One was cyanidin-3,7,31-triglucoside with 3 molecules of ferulic acid and an extra terminal glucose. The other pigment was similar but without the terminal glucose unit. The nonacyl portion was indentified by comparison with the pigment of Zebrina pendula. The acyl/aglycone and acyl/sugar ratios were determined by using internal standards and response factors with HPLC. The pigments may have potential as food colorants.

    Notes: Abstract The relative contributions of the pre- and postsynaptic components of the myoneural blocking effect of different antibiotics were studied using: (a) a radio-active method that measures selectively the Ca2+-dependent, stimulation evoked, quantally released, 3H-acetylcholine (3H-ACh) from the mouse in vitro phrenic nerve-hemidiaphragm preparation without cholinesterase inhibition; (b) measurement of the force of contraction of the indirectly or directly stimulated muscle. The antibiotics studied (neomycin, polymyxin B and lincomycin), reduced the release of 3H-ACh evoked by stimulation (18 trains of 40 shocks at 50 Hz) in a concentration dependent manner. While the inhibitory effect of neomycin was inversely related to [Ca2+] o , that of lincomycin was moderately and that of polymyxin B was not affected by increasing [Ca2+] o from 0.75 to 5.0 mM. Similarly, the d-tubocurarine (d-Tc)-induced inhibition of the release of 3H-ACh was independent of [Ca2+] o . The K-channel blocking agent, 4-aminopyridine (4-AP), enhanced the release of ACh in a concentration dependent manner and prevented the neuromuscular effect of neomycin. However, the neuromuscular effect of polymyxin B and of lincomycin was not affected by 4-AP. Atropine, enhanced the release of 3H-ACh. Antibiotics, however, were still able to reduce the release of ACh when the negative muscarinic feedback mechanism of ACh release was eliminated by atropine. Our findings indicate that the antibiotics studied possess both pre- and postsynaptic effects. Presynaptically they reduce the evoked release of ACh; postsynaptically they inhibit muscle contractility. The rank order of presynaptic action is neomycin 〉polymyxin B 〉lincomycin.     Notes: Abstract We report ab initio cluster computations of deuterium atoms in a tetrahedral palladium site, performed at the UHF level with extended basis sets. An interstitial deuteron is found to be energetically favored over an interstitial deuterium atom. Computations with one interstitial deuterium atom or with one deuteron reveal an increase in electron density near the deuteron. Not only valence electrons but also inner core electrons of the palladium atoms are present in the vicinity of the deuteron. A potential energy curve is calculated with two deuterium atoms approaching each other, with one deuterium fixed at the center of a tetrahedral site. With regard to the environment provided by molecular deuterium, our results show that the tetrahedral site does not favor a closer deuteron encounter. No metastable dimer geometry is found. The two deuterons repel each other despite the screening. These computations represent more than 250 CPU h of a new-generation biprocessor vector computer.

    Notes: Abstract We have screened fourteen kindreds with X-linked hypophosphataemic rickets with four microsatellite markers, viz. AFM163yh2, DXS999 (AFM234yf12), DXS443 and DXS365, in order to refine the genetic map flanking the gene, and to define a close flanking interval for the construction of a yeast artificial chromosome (YAC) and cosmid contig. The genetic data were enhanced after the isolation of a large 1.2-megabase YAC derived from AFM163yh2, in which marker DXS274 was present but not DXS365 or DXS443. Against HYP, DXS365, AFM163yh2 and DXS443 showed no recombinants (Z max = 18.1, Z max = 9.9, and Z max = 16.0 respectively). DXS999 gave Z max = 9.6 at 4% recombination and lies distal to HYP but proximal to DXS197 and DXS43. The disease gene and markers AFM163yh2 and DXS365 are flanked by DXS443 and DXS274. Combining the genetic and physical data, we are able to propose the following gene marker order: Xptel-DXS43-DXS197-DXS999-DXS443-[(DXS365-AFM163yh2), HYP]-DXS274-DXS41-Xcen.
