    Notes: Time-average values from particle simulations of a collisionless, single-emitter plasma device modeling single-ended Q machines or thermionic converters with a negatively biased collector are presented. These results quantitatively confirm the predictions of collisionless, kinetic plane-diode theory for spatial potential profiles that decrease monotonically. However, simulations of negative-bias potential profiles with a single internal maximum differ significantly from previous theoretical predictions which assumed electron phase space to have either (i) no trapped electrons or (ii) trapped electrons isothermal with the passing electrons. A more general class of trapped-electron model distributions is introduced from which new equilibrium potential values can be recovered that closely match the simulations. These simulations clearly demonstrate the sensitive role that trapped electrons play in shaping the potential profiles of the equilibrium (or slowly evolving) states of the simulated systems. The trapped-electron distributions in these simulations are themselves shown to be controlled critically by fluctuations whose levels are varied by the choice of particle injection scheme. These effects, although found and discussed here in the context of a particular model, are believed to be important in many bounded plasma systems where electrons can be trapped in potential wells.

    Notes: The efficacy of low-dose dopamine as a renal protective agent was compared with that of dopexamine in patients who underwent orthotopic liver transplantation. Twelve patients who received a continuous infusion of dopexamine (1–3 μg/kg/minute) were matched for age, diagnosis, pre-operative creatinine clearance and blood loss with 12 patients who received a low-dose infusion of dopamine (2 μg/kg/minute). The catecholamine infusion was started after induction of anaesthesia and continued for 48 hours after surgery. Patients in the dopexamine group had less evidence of renal impairment and failure than those in the dopamine group during 7 days after the operation, although the differences between groups did not achieve statistical significance. Similarly there were no significant differences between the two groups in peri-operative urine output, urine/plasma osmolality ratio or creatinine clearance. Dopexamine is at least as effective as dopamine for renal protection in patients who undergo liver transplantation.

    Notes: Abstract Cystic encephalomalacia occurred in two preterm infants who had proven intrauterine herpes simplex virus type 2 infection. Calcification was evident in the basal ganglia. Follow-up scans indicated that the cysts had resolved over a period of two months in one infant, while in the case of the other, the cysts became progressively larger with significant ventricular dilatation developing. While cystic changes in the brain of preterm infants are usually due to periventricular leukomalacia intrauterine infection needs to be considered as a possible cause. Antiviral treatment may be of benefit to infants with herpes simplex virus infection.

    Notes: Summary The use of bioelectrical impedance (BI) analysis as a non-invasive approach for individualising gentamicin therapy in newborn infants has been investigated in a two phase study. In Phase I, 1/impedance and length2 were identified as statistically significant predictors of the distribution volume of gentamicin (Adj R2=0.78, CV=12.42%), and length2/impedance and post-conceptual age were predictors of total systemic clearance (Adj R2=0.83, CV=14.5%), following the administration of 2.5 mg·kg−1 gentamicin to 17 neonates (gestational age (GA) 27 to 36 weeks). In a prospective validation of these relationships in an independent (Phase II) group of 27 infants (GA 26 to 41 weeks), predicted serum gentamicin concentrations were close to those achieved. Several instances of high prediction errors (predicted minus achieved levels) were observed in infants with known or suspected renal impairment and they caused significant (P〈0.05) perturbation in the bias and accuracy of the models. Daily BI measures over a four to five day period were able to detect individual changes in the fat-free body compartments, which were translated into alterations in gentamicin regimens. This simple, non-invasive and relatively inexpensive bedside technique provides a potentially valuable means to individualise gentamicin therapy without relying on the measurement of serum gentamicin concentration.

    Notes: Abstract The expression levels of recombinant human follicle stimulating hormone (r-hFSH) were studied in several CHO-K1 derived cell lines. The cell lines varied in the promoter used to control r-hFSH expression and in the subunit gene copy ratio. FSH is a heterodimeric molecule, with 2 N-glycosylation sites per peptide chain, and shares a common alpha subunit with the other gonadotropins. Serum stimulated FSH production in the beta actin promoter cell lines 2–3 times over the 7–10 ng/106 cells/h levels obtained in protein-free medium. Serum seemed to have roles other than purely at the transcriptional level judging by the increased free alpha to dimer ratio secreted from cell cultrured in serum-free medium. Zinc induced FSH expression in metallothionein cell lines, with a 3-fold induction at 50μM concentrations compared to 0μM zinc, giving specific productivities of about 7–10 ng/106 cells/h, but the induction kinetics were complicated, and suggested other roles for zinc in addition to activation of the metallothionein promoter. Evidence suggested significant post-transcriptional regulation of FSH expression.

    Notes: Abstract A recombinant CHO cell line in which the expresison of human follicle stimulating hormone (hFSH) was under the control of the β actin promoter was maintained in steady state perfusion cultures on a protein free medium. The level of expression of the hFSH was controlled by varying the steady state level of dissolved oxygen (10–90% of air saturation) and of sodium butyrate (0–1.5mM). Under these conditions, the specific productivity of hFSH (qFSH) varied from 0.7 to 4.8 ng hFSH/106 cells/h. As the specific productivity of hFSH increased, there was a shift in the FSH isoforms to the lower pI fractions, corresponding to increased sialic acid content. As the specific productivity of hFSH increased, shifting the isoform distribution towards the lower pI isoforms, that the sialyltransferase enzymic activity also increased.

    Notes: Abstract A model mammalian cell system for the production of recombinant proteins was investigated. Murine myeloma cells which had lost the ability to produce both heavy and light chain immunoglobulin molecules were transfected with a vector containing the immunoglobulin heavy chain promoter and enhancer elements linked to the human growth hormone gene. The growth kinetics of G32, a clonal isolate, were found to be similar to both the parent myeloma and hybridomas. However, production of hGH by G32 was growth associated, rather than as a secondary metabolite as is the case for hybridomas. In addition, G32 produced hGH at molar levels greater than most hybridomas.
