    Notizen: The response to host genotype-directed selection for specific virulence in Sclerospora graminicola was studied in the pearl millet-downy mildew system. In greenhouse experiments, seedlings of resistant pearl millet genotypes MBH 110 and 852B were inoculated with sporangia of a field population of S. graminicola maintained on susceptible genotypes NHB 3 and 7042. Sporangia from infected seedlings of MBH 110 and 852B were used to inoculate seedlings of MBH 110 and 852B, respectively, in succeeding generations. Within 12 generations of selection on MBH 110 and five generations of selection on 852B, respective host-specific virulences were identified. Selection for 852B-specific virulence was much faster than that for MBH 110-specific virulence. Symptom type, and pathogenic fitness parameters such as latent period, infection index, sporangial and oospore production were measured, and composite fitness index (CFI) was calculated for each selection generation. In general, CFI and individual fitness parameters were positively correlated with selection generation. Virulences of the host-specific pathotypes artificially selected through asexual generations were comparable to the naturally selected host-specific pathotypes AB, an MBH 110-specific pathotype from Aurangabad, and MYS, an 852B-specific pathotype from Mysore. The host genotype x pathogen population interaction was highly significant (P≤0-001). The results indicate that genetic variation for host genotype-specific virulence exists within field populations of the pathogen, and that selection through asexual pathogen generations can rapidly increase the quantitative virulence of the population to resistant host genotypes. This implies that resistance and virulence in the pearl millet-downy mildew system are controlled by relatively few genes.

    Notizen: ‘BJ 104’ was the most widely grown pearl millet hybrid in India until it became susceptible to downy mildew (DM) in 1984—85. Residual variability for resistance was found in both parental lines, 5141 B (maintainer of 5141 A) and J 104, and through four generations of pedigree selection under intense disease pressure in the DM nursery, two lines, IC-MA841 (from 5141 B) and ICMP 84814 (from J 104), were selected resulting in a reconstituted DM resistant hybrid (‘ICMH 84814’) which was equal in yield to the original ‘BJ 104’. The reconstituted hybrid, though phenotypically similar, can be distinguished from “BJ 104” being slightly taller, flowers later, has heavier heads, and 1000-seed weight, but tillers less.A similar exercise was attempted on Tifton 23 B, the female parent of the first widely grown hybrid (‘HB 3’— the male parent was also J 104), but no variability for resistance to DM was found.

    Notizen: Recent progress in the analysis of mutants of the Eschenchia coli lactose carrier function is reviewed, with special emphasis on the structural basis for energy barriers which prevent ‘forbidden’ con-formational changes. Mutations which break down the barriers to forbidden isomerizations involving the binary carrier:sugar (CS) and carrier:proton (CH) complexes have been obtained in several laboratories. These mutants allow uncoupled transport of H+ or galactoside in the lactose carrier which normally couples cation and sugar movement in a 1:1 stoichiometry. These uncoupled mutants appear to be associated with changes in both sugar and cation recognition, suggesting that the physical interactions forming the basis for co-substrate recognition and uncoupling are not independently variable.By postulating that translocation involves transformation of the stable intermediate of the co-transport cycle to unstable transition state conformations of the carrier, it is possible to consider the consequences of mutagenesis in terms of transition state theory. Consistent with several experimental observations, the analysis predicts in each mutant the occurrence of more than one abnormality in the transport cycle (such as changes in sugar recognition, cation recognition or the coupling reaction). We have called the general phenomenon a ‘mutational double-effect’ because any mutation which alters the Gibbs free energy change of one reaction in the transport

    Notizen: Abstract  The protein binding of weakly acidic and basic drugs has been shown to be altered in cancer patients. Brequinar is a weakly acidic, low-clearance, and highly protein-bound (〉98% bound) antitumor agent. The pharmacokinetic parameters of brequinar are subject to large interpatient variability. This large interpatient variability may be related to brequinar’s plasma protein-binding capacity (assuming no change in the intrinsic clearance of the unbound drug). The objectives of this study, therefore, were (a) to characterize brequinar’s protein binding in the plasma of healthy donors and cancer patients and (b) to examine the relationships between brequinar’s plasma protein binding and its pharmacokinetics in patients. Brequinar protein binding was determined in human serum albumin (HSA) solution, drug-free donor plasma, and brequinar-free, predose plasma samples obtained from a phase I cancer trial. Pharmacokinetic results from this study were used to examine relationships between plasma protein binding and drug disposition. In HSA solution and healthy donor plasma, brequinar’s protein binding as determined using spiked samples was concentration-dependent. The unbound brequinar fraction increased by a factor of 3 (from 0.3% to 0.9% free) in 4% HSA solution and by a factor of 4 (from 0.4% to 1.6% free) in donor plasma as the brequinar concentrations increased from 0.1 to 2.3 mM in the HSA solution and from 0.076 to 1.5 mM in the donor plasma. Analysis of brequinar binding characteristics using the binding ratio and Rosenthal binding plots showed that albumin was the primary protein for brequinar binding in human plasma. The addition of various concentrations of α1-acid glycoprotein to 4% HSA solution did not affect the protein binding of brequinar to HSA. The protein binding determined in the plasma of cancer patients was not quantitatively different, except for variability, from that observed in the plasma of healthy donors. Examination of relationships between the unbound brequinar fraction and pharmacokinetics suggested that plasma protein binding was not a major determinant of brequinar disposition in cancer patients.

    Notizen: Abstract Penumatosis Intestinalis (gas in the bowell wall) is uncommon in children and is associated with a number of clinical conditions [1] most notably necrotizing enterocolitits in sick neonates. We observed portal venous gas on sonography and benign pneumatosis intestinalis on abdominal raiiography in two children who had recently undergone liver transplantation. Asymptomatic portal venous gas and pneumatosis intestinalis has not been described previously in pediatric liver transplant patients. It is important to recognize the sonorgaphic appearance of portal venous gas since sonography is the imaging modality most frequently utilized for monitoring the allograft post liver transplantation.
