    Notes: Abstract The thermotolerant methylotroph Bacillus sp. C1 possesses a novel NAD-dependent methanol dehydrogenase (MDH), with distinct structural and mechanistic properties. During growth on methanol and ethanol, MDH was responsible for the oxidation of both these substrates. MDH activity in cells grown on methanol or glucose was inversely related to the growth rate. Highest activity levels were observed in cells grown on the C1-substrates methanol and formaldehyde. The affinity of MDH for alcohol substrates and NAD, as well as V max, are strongly increased in the presence of a M r 50,000 activator protein plus Mg2+-ions [Arfman et al. (1991) J Biol Chem 266: 3955–3960]. Under all growth conditions tested the cells contained an approximately 18-fold molar excess of (decameric) MDH over (dimeric) activator protein. Expression of hexulose-6-phosphate synthase (HPS), the key enzyme of the RuMP cycle, was probably induced by the substrate formaldehyde. Cells with high MDH and low HPS activity levels immediately accumulated (toxic) formaldehyde when exposed to a transient increase in methanol concentration. Similarly, cells with high MDH and low CoA-linked NAD-dependent acetaldehyde dehydrogenase activity levels produced acetaldehyde when subjected to a rise in ethanol concentration. Problems frequently observed in establishing cultures of methylotrophic bacilli on methanol- or ethanol-containing media are (in part) assigned to these phenomena.

    Notes: Abstract The thermal stability of unsupported alumina membrane top-layers was studied by determining the pore structure (mainly pore size) change of alumina gels, prepared by sol-gel methods, after sintering at different temperatures ranging from 450 to 1200 °C. The average pore size of the pure alumina membranes and PVA-added membranes increased sharply after sintering at temperatures higher than 1000 °C. Addition of 3% lanthanum, either by mixing lanthanum nitrate in the alumina sol or impregnating lanthanum nitrate into calcined alumina gel, followed by a second heat treatment, can considerably stabilize the pore structure of the alumina membrane top-layers. The pore diameter for the lanthanum-doped membranes was stabilized within 25 nm after sintering at 1200 °C for 30 h, about one-sixth of that for the pure alumina membranes after sintering at 1200 °C for 30 h. The substantial increase in the pore size for the pure alumina membranes at the sintering temperature of 1000 to 1200 °C was accompanied by the phase transformation from γ-to α-alumina. The addition of lanthanum can raise this phase transformation temperature by about 200 °C.

    Notes: Abstract We have investigated positron scattering from the alkali atoms lithium, sodium, potassium and rubidium, focusing on the calculation of differential cross sections for elastic scattering and impact excitation. The results obtained with the two-state and five-state close-coupling method as well as the first-order distorted wave Born approximation are compared with each other and with some selected results for electron scattering. Numerical problems due to lack of, or numerically inaccurate, calculation of partial waves with large angular momenta are solved through semi-empirical fitting/extrapolation procedures.
