    Notes: The integro-differential growth model of Eakman, Fredriekson, and Tsuehiya has been employed to fit cell size distribution data for Schizosaccharomyces pombe grown in a chemostat under severe product inhibition by ethanol. The distributions were obtained with a Coulter aperture and an electronic system patterned after that of Harvey and Marr. Four parameters - mean cell division size, cell division size standard deviation, daughter cell size standard deviation, and a growth rate coefficient - were calculated for models where the cell growth rate was inversely proportional to size, constant, and proportional to size. A fourth model, one where sigmoidal growth behavior was simulated by two linear growth segments, was also investigated. Linear and sigmoidal models fit the distribution data best. While the mean cell division size remained relatively constant at all growth rates, standard deviation of division size distribution increased with increasing holding times. Standard deviation of the daughter size distribution remained small at all dilution rates. Unlike previous findings with other organisms, the average cell size of Schizosaccharomyces pobme increased at low growth rates.

    Notes: Positive and negative thermospray mass spectra have been recorded for a number of phenazines at the submicrogram level. The spectra are dominated by molecular ion species ([M + H]+, [M - H]-) with minimal fragmentation. The gas chromatographic/electron impact mass spectrometric behaviour of the phenazines was also investigated. A number of phenazines were not amenable to gas chromatographic/mass spectrometric analysis without derivatization; in contrast, thermospray mass spectrometric data were obtained for each phenazine. Thermospray liquid chromatography/mass spectrometry has been applied to identify phenazine-1-carboxylic acid and 2-hydroxyphenazine in culture filtrates of Pseudomonas aureofaciens and 1-amidophenazine from Pseudomonas aeruginosa.

    Notes: Glucoamylase, industrially derived from Aspergillus niger, was chromatographically separated into forms I and II and purified to near homogeneity. Preparations were proved to be free of D-glucosyltransferase by electrophoretic and differential inhibition tests. Maximum rates and Michaelis constants were obtained for both glucoamylases I and II with maltooligosaccharides from maltose to maltoheptaose and with isomaltooligosac-charides from isomaltose to isomaltohexaose. Subsite maps were calculated from these kinetic data and were not significantly different for the two forms. Subsites in both forms had lower affinities for D-glucosyl residues contained in isomaltooligosaccharides than for D-glucosyl residues in maltooligosaccharides.

    Notes: The penetration of ultrasonic waves through opaque media and the large difference in the acoustic properties between air bubbles and the fermentation broth were used to measure the energy attenuation of pulsed ultrasound by the bubbles as the waves passed through the broth. This leads to an on-line determination of the specific interfacial area provided information is available about the holdup or bubble mean diameter. This article gives the principle of the method and demonstrates how the measured interfacial area may be used in evaluating the mass transfer coefficient of a fermentation system in a bubble column.

    Notes: Near-homogeneous forms of glucoamylases I and II, previously purified from an industrial Aspergillus niger preparation, were used to hydrolyze a number of di- and trisaccharides linked by α-D-glucosidic bonds. Maximum rates and Michaelis constants were obtained at various temperatures and pH values with glucoamylase I for the disaccharides β,α-trehalose, kojibiose, nigerose, maltose, and isomaltose and the trisaccharides panose and iso-maltotriose, and with glucoamylase II for maltose, maltotriose, and isomaltotriose. Maximum rates were highest and energies of activation were lowest for maltose, maltotriose, and panose, the only three substrates containing α-D-(1, 4)-glucosidic bonds. Michaelis constants were lowest and standard heats of binding were most negative for maltose and maltotriose. The variation of maximum rates and Michaelis constants with varying pH values suggested that two carboxyl groups were involved in substrate binding.

Kinetics and equilibria of the formation of α,β-trehalose, kojibiose, nigerose, maltose, isomaltose, panose, and isomaltotriose, which with isomaltotetraose were the only products formed, were determined. There was no difference in the abilities of GA I and GA II to form these products. Activation energies for the formation of maltose and panose were lower than those of the other Oligosaccharides. Relative rates of oligosaccharide production based on glucoamylase hydrolytic activity did not vary significantly between pH 3.5 and 4.5 but were lower at pH 5.5. Maltose was formed much faster than any other product. Equilibrium concentrations at higher dissolved solids concentrations decreased in the order isomaltose, isomaltotriose, kojibiose, nigerose, maltose, α, β-Mrehalose, panose, and isomaltotetraose. They were not appreciably affected by changes in temperature or pH. A kinetic model based on adsorption of D-glucose and the seven di- and trisaccharides by the first three glucoamylase subsites was formulated. Oligosaccharide formation was simulated with the model, using equilibrium data gathered for this article and subsite binding energies and kinetic parameters for oligosaccharide hydrolysis measured earlier. Agreement of simulated and actual oligosaccharide formation data through the course of the reaction was excellent except at very high solid concentrations.

    Notes: The bacterium Acetobacter suboyxdans and the yeast Saccharomyces carlsbergensis have been grown together on a synthetic medium in a chemostat. Mannitol, the only carbon source fed to the fermenter, is oxidized by the bacteria to fructose. The yeast, which cannot attack mannitol, breaks down the fructose nearly completely. Eight steady states and five transitory periods after changes in flow rate have been analyzed to study the kinetics of the mixed culture. Separate cell concentrations were determined by a modified Coulter counter apparatus. Both sugars were monitored. Both bacteria and yeast may be modeled using Monod's equation, the latter with some deviations. The yeast is unable to grow beyond the washout point of the bacteria, even though its maximum growth rate is much higher. The yield of both organisms decreases with increasing dilution rate, as does their average cell size. After step changes in dilution rate, repeated oscillations of both sugar and cell concentrations usually occur before steady-state conditions are reattained. They are generally in phase, with no definite sign of a lag. Oscillations of yeast and fructose concentrations are more pronounced. Periods average about 6 hr and are not correlated with fermentation conditions or equipment variables. Repeated oscillations are not found after step-downs in pure cultures of A. suboxydans, leading to the conclusion that the instability in mixed cultures may be caused by a feedback mechanism from the yeast to the bacteria.

    Notes: The response of two-species commensalistic systems in a chemostat has been investigated after perturbations in steady state conditions and after step changes in dilution rate. The system is inherently stable with not more than three overshoots and undershoots possible. More complicated commensalistic systems are less stable, with limit cycle response occurring after dilution rate changes when feedback inhibition and feedforward activation occurs. In general variation of feedback parameters is more effective in changing the behavior of the systems than variation of feedforward parameters. Limited agreement with the experimental data of Chao and Reilly was obtained.
