Notes: Summary Recently, the Wilson's disease locus (WND) has been mapped to the long arm of chromosome 13. We have analyzed segregation of serveral chromosome 13 markers flanking the WND locus and used multipoint linkage analysis to determine the most likely WND genotype of each of 57 unaffected individuals in 5 Wilson's disease families. Approximately 46% of these could be classified as carrier (heterozygote), homozygous normal, or homozygous affected (not yet symptomatic) with a probability of at least 90%, while 77% could be classified with a probability of at least 80%. Our results demonstrate that even though there is a significant decrease on average in serum copper concentration in Wilson's disease heterozygotes compared to normal homozygotes, other sources of variation in serum copper concentration are much greater and preclude use of serum copper to detect heterozygotes for Wilson's disease. Subsequent analyses showed that a familial component, independent of WND genotype, is the major factor accounting for variation in ceruloplasmin levels among unaffected individuals; age is another factor accounting for more variation in copper levels among unaffected individuals than WND genotype.

Notes: Summary Dyskeratosis congenita is an X-linked recessive disorder with diagnostic dermatological features, bone marrow hypofunction, and a predisposition to neoplasia in early adult life. Linkage analysis was undertaken in an extensive family with the condition using the Xg blood group and 17 cloned X chromosomal DNA sequences which recognise restriction fragment length polymorphisms (RFLPs). No recombination was observed between the locus for dyskeratosis congenita (DKC) and the RFLPs identified by DXS52 (St 14-1) (Zmax=3.33 at Θmax=0 with 95% confidence limits of 0 to 14 cM). Similarly no recombination was observed for the disease locus and F8 (Zmax=1.23 at Θmax=0) nor for DXS15 (Zmax=1.62 at Ήmax=0), but both of these markers were only informative in part of the family whereas DXS52 was fully informative. DXS52, DXS15, and F8 are known to be tightly linked and have previously been assigned to Xq28. Thus the gene for dyskeratosis congenita can be assigned to Xq28. These DNA sequence polymorphisms will be of clinical value for carrier detection and prenatal diagnosis.

Notes: The Raman spectrum of diprotonated 4,4′-bipyridine in the solid state as the dibromide salt is reported. The resonance Raman spectrum of the radical cation derived by one-electron reduction of the dication is also reported. Viscous glycerol was employed as a solvent in order to protect the radical by inhibiting oxygen diffusion. The spectra are interpreted on the basis of a comparison with those of related compounds. A large shift in frequency (+70 cm-1) of the inter-ring stretching vibration between the parent dication and radical monocation is observed. This has been interpreted in terms of a major contribution to the delocalized radical structure of a quinoid-type form.

Notes: Raman spectra are reported for several bridged diquaternary salts of 2,2′-bipyridine. Resonance Raman spectra and some excitation profiles of the coloured radical monocations obtained from the diquaternary salts by one-electron reduction are also reported. Selected frequencies of the parent dications are correlated with changes in the torsional angle between the aromatic rings of the bipyridinium moiety. These correlations, when applied to the spectra of the radical monocations, suggest that the radicals exhibit reduced torsional angle and increased inter-ring bond order, compared with the parent dications. These changes are consistent with predictions of increased inter-ring π-electron delocalization, associated with radical formation. The torsional angle appears to decrease for the radical monocations with increasing number of bridging methylene groups. This is opposite to the trend known to occur in the parent dications.
