Summary The choice of method for determining nitate levels depends on the accuracy of the test and the nature of the task, i.e. whether exact scientific analysis or rapid semiquantitative appraisal. Four methods for analysis the nitrate content of potato tubers, which required little by way of equipment and reagents were tested for their accuracy and applicability: (1) the Merckoquant® test strip (Merck, Darmstadt, FRG) (2) the ion-sensitive electrode (Kolbe & Müller, 1984) (3) microbiological method (Kücke & Przemeck, 1983) (4) the 1,3,4-xylenol method (vielemeyer & Seyfarth, 1965). Juice was extracted for fresh samples either from 10 whole tubers (2.5 kg–5.0 kg fresh weight (fw) per sample) or from three cores taken with a cork borer from one tuber. If dried material were used then 2 g of inely milled samples were shaken in 50 ml distilled water (Merckoquant®, microbiological method). or with 1% alum solution (electrode, xylenol method). There was a close correlation between results obrained by applying the Merckoquant® test to single tubers and to mixed samples (Fig. 1), but the variation about the line at 14.5–20% was considerable, and nitrate levels above 200 mg NO 3 − /kg fw were often overestimated (Fig. 2). Although this rapid method is best used semiquantitatively, the results demonstrate that it can give more than simple appraisals, especially if numbers of samples appropriate to this test are analysed (r=0.982; Fig. 2). The coefficient of variation of measurements by the electrode method ats=2.2%. was generally comparable with other exact methods. The ion-ssensitive electrode provides rapid and precise analyses of nitrate contents of 50 mg NO3/−/kg fw and upwards, and is therefore suitable for rapid and large scale analyses. Under 50 mg NO 3 − /kg fw the readings were clearly too high when compared with the microbiological method (Fig. 3). The recovery rates for nitrate added in standard amounts to smaples were on average 103% and 99.1% for the potentiometric and microbiological method respectively (Table 1). Nitrate measurements obtained by the microbiological method were 10% higher than by the 1,3,4-xylenol method for nitrate contents of about 500 mg NO 3 − /kg fw, but at low concentrations there was very good agreement overall. The microbilogical method was especially good for exact quantitative measurements and for very low nitrate levels, whereas the xylenol method was unsuitable for analysing the nitrate contens threshold at 100 mg NO 3 − /kg fw, and considerable demands on time and labour. Table 2 assesses in summary form each of the tested methods.

Résumé Selon la précision de l'analyse du nitrate (méthode d'analyse scientifique ou méthode rapide semi-quantitative), les exigences diffèrent suivant les méthodes. Pour étudier la précisoin et l'intérêt des méthodes d'analyse de nitrate sur tubercules, 4 méthodes relativement peu exigeantes en équipement et en réactifs ont été examinées: (1) la bande test Merckoquent® (Fa. Merck, Darmstadt, RFA) (2) électrode spécifique (Kolbe & Müller, 1984) (3) un procédé microbiologique (Kücke & Przemeck, 1983) (4) la méthode xylénol-1,3,4 (Vielemeyer & Seyfarth, 1965) Pour l'analyse à partir de la matière fraîche (échantillon de 2,5 à 5.0 kg), environ 10 tubercules entiers ou 3 carottes par tubercule extraites avec une sonde cylindrique et pressées sont examinés. Avec du matériel déshydraté, un échantillon finement moulu de 2 g est mélangé avec 50 ml d'eau distillée (Merckoquant® méthopde microbiologique), ou avec une solution d'alun à 1% (électrode spécifique, méthode au xylénol). Entre les deux méthodes du test Merckoquent® (soit bonne corrélation est observée (Fig. 1). En revanche, la variation des résultats de la méthode avec bande test est considérable (14.5–20%). Les teneurs en nitrate de plus de 200 mg/kg de poids frais sont souvent surestimées (Fig. 2). Bien que ce test rapide ne soit qu'une méthode semi-quantitative, les résultats obtenus avec un nombre important d'échantillons, et confrontés à ceux d'autres méthodes, démontrent que les valeurs sont comparable (r=0,982; fig. 2). Avec une variation des=2,2% la méthode à l'électrode spécifique est comparable à d'autres méthodes exactes dans la plage principale des déterminations. L'électrode spécifique permet une analyse rapide et preécise de la teneur en nitrate lorsque la quantité de NO 3 − atteint environ 50 mg ou plus par kilo de matière fraîche. Par conséquent, cette méthode se prête particulièrement pour les analyses de routine et de séries. En dessous de 50 mg NO 3 − /kg de matière fraîche, les valeurs sont surestimées par rarpport à la méthode microbiologique (Fig. 3). Le taux de récupération du nitrate additoinné est d'environ 103,0% pour la méthode à l'électrode spécifique et de 99,1% pour la méthode microbiologique (Tab. 1). Pour des teneurs en nitrate d'environ 500 mg/kg de matière fraîche, la méthode microbiologique donne des valeurs d'environ 10% supérieures à la méthode au xylénol-1,3,4. En revanche, pour des cioncentrations plus faibles, les valeurs obtanuesa par les deux méthodes correspondent bien (Fig. 4). Selon les résultats obtenus, la méthode microbiologique se prête bien pour une analysie quantitative exacte de teneurs très faibles. En revanche, la méthode au xyléno. ne devient failble qu'à partir de valeurs de 100 mg NOn 3 − /kg de poids frais et, en raison des contraintes en travail et en temps, elle ne convient pas pour l'analyse du nitrate dans les tubercules de pommes de terre. Le tableau 2 résume et compare les méthodes testées.

Zusammenfassung Für die Nitratbestimmung in Kartoffelknollen wurden vier mit relative teringem Aufwand an Gerät und Reagenzien anwendbare Methoden geprüft und Einsatzmöglichkeiten beschrieben. Der Merckoquant®-Test eignet sich wegen der hohen methoden-bedingten Streuung vons=14,5–20,0% als halbquantitatives Verfahren für Routinekontrollen. Hohe Nitraygehalte von über 200 mg/kg Frischsubstanz (Frs.) werden überschätzt. Die ionensensitive Elktrode erlaubt bei einer relativen Standardabweichung der Messwerte vons=2,2% ab 50 mg, NO 3 − /kg Frs. an aufwärts eine voll quantitative und schnelle Analyse des Nitrats. Geringe Nitratgehalte (unter 50 mg/kg Frs.) werden allerdings überschätzt. Mit dem mikrobiologischen Verfahren können demgegenüber auch niedrige Nitratgehalte genau erfasst werden. Die 1,3,4-Xylenolmethode erweist sich dagegen wegen des hohen Arbeitsaufwandes und einer unteren Nachweisgrenze von 100 mg/kg Frs. für eine schnelle und exakte Nitratanalyse in Kartoffelknollen als relativ ungeeignet.
