Notes: Abstract: The originally described sequence of human myelin basic protein peptide 45–89 has recently been shown to contain two errors which have now been resolved. In the present study fast atom bombardment mass spectrometry was utilized to analyze the primary sequence of the other portions, peptides 1–44 and 90–170 of human myelin basic protein. The results obtained confirm the accuracy of the primary sequence published for both of these terminal peptides.

Notes: Electromigration was studied in Al conductors underlaid with chemically vapor deposited W, and the results compared to the more conventional Al/TiW metallization. In Al/TiW conductors electromigration results in Al whisker growth, causing metal short rather than open-circuit failure. Contrary to expectations, the Al/W metallization was found to be more susceptible to the formation of whiskers during current stress, with Al/W lifetimes reduced by about one-half compared with Al/TiW. The temperature and current density failure dependencies were not significantly affected by the composition of the underlayer. The mechanical stress in the underlayer was not found to significantly affect the stress distribution in the overlying Al film.

Notes: Abstract A report of two patients in which a soft tissue mass, initially regarded as a malignant tumor, was shown to be the result of calcium pyrophosphate deposition disease. The first case, a woman aged 71 years, presented with a mass involving the right fifth finger. In the second case, also a women aged 71 years, the lesion involved the tissues adjacent to the right hip. Each lesion consisted of a mass of highly cellular tissue containing deposits of calcium pyrophosphate dihydrate crystals. the clinical, radiological, and pathological features of the two cases are compared with those of seven similar cases reported in the literature.

Notes: Abstract Hypersensitivity reactions occurring during barium studies of the gastrointestinal tract are rare. A case is presented with radiographically demonstrated angioedema in the stomach and small bowel accompanied by allergic rhinitis, which was apparently an allergic response to the barium sulfate suspension. The reaction was documented twice during separate challenges to the barium suspension performed several months apart.

Description / Table of Contents: Abstract A method for the determination of selenium in food and biological materials has been developed. The method involves sample decomposition in a closed system at 150 °C with nitric acid, perchloric acid treatment of the digest, reduction of selenium to the tetravalent state with hydrochloric acid followed by Se-2,3-diaminonaphthalene (DAN) complex formation and extraction in a single-step operation. Performance characteristics of the method (accuracy and repeatability) were extensively tested for eleven reference materials and ten practice samples covering both a wide variety of matrices and a wide range of selenium content. Almost all results for reference materials were within the 95% confidence range of the certified value or the best estimate thereof. The coefficient of variation for all samples ranged from 2.3 to 10.1%, average value 4.7%. Average recovery for spiking experiments — levels 0.060, 0.075 and 0.150 gm/g was 102%; range 84–114%.

Description / Table of Contents: Summary In the last decade 311 duplicate portions of the 24-h diet of Dutch citizens were collected and lyophilized. All samples were assayed for total arsenic and, in addition, the 20 samples which ranked highest in total arsenic were selected for arsenic speciation i.e., the separate determination of organic-bound arsenic and inorganic arsenic. For the determination of total arsenic, the test portion is wet-digested and the digest reduced with KI/SnCl2/Zn to give AsH3 which is measured by molecular absorption spectrometry. Inorganic arsenic is also measured as AsH3 after acid extraction of the test portion and subsequent NaBH4 reduction of the extract. The residue of the acid extraction and the NaBH4-reduced extract are combined, wet-digested and the digest reduced with NaBH4 to give AsH3 originating from organic-bound arsenic. Samples spiked at different levels with inorganic and organic-bound arsenic were analysed and gave recoveries for inorganic arsenic of 94% to 122% (n=6) and for organic-bound arsenic of 87% to 104% (n=6). On average, the inorganic fraction is 30% of the total arsenic intake; range 3 % to 50% (n=14). The inorganic arsenic fraction decreases with increasing level of total arsenic intake. The highest oral inorganic arsenic intake was 60 gg; the tolerable daily intake (TDI) for a 60-kg individual is 120 μg inorganic arsenic. Organic and inorganic arsenic measured were summed and compared with the results of the total arsenic determination; differences were less than 15%. (Since inorganic arsenic is a contaminant and not an additive, the abbreviation TDI is preferred here over ADI (acceptable daily intake) which is retained for additives.)

Notes: Summary To develop a screening procedure for the detection of restriction endonucleases in micromonosporae and catellatosporae based on efficiency of plating, eight different actinophages were isolated from soils enriched withMicromonospora species and one fromCatellatospora-enriched soil. The lytic actinophages all contained double-stranded DNA and the majority appeared, when examined by electron microscopy, to belong to Ackermann's type B1 since they had isometric heads and noncontractile tails. One actinophage was classified as type C1 because of its isometric head and very short noncontractile tail. The host ranges of the actinophages were determined on strains ofMicromonospora and selected species from other actinomycete genera of cell wall chemotype II. Type II restriction enzymes were isolated fromM. echinospora ssp.echinospora (ATCC 15837),M. purpurea (ATCC 15835) andM. zionensis (LL-100-125) and were designatedMecI,MpuI andMziI, respectively. Restriction enzymesMecI andMpuI are isoschizomers ofXhoI, whileMziI is an isoschizomer ofPvuII.

Notes: The preparation and the spectroscopic analysis of a dimeric adduct (V) obtained from 2-isopropenyl-5-butoxyoxazole (III) and α,α-azobis-isobutyronitrile (AIBN) (IV) are reported. It is noteworthy that (III) inhibits completely the homopolymerization of styrene. This is in strong opposition to the facile homopolymerization and copolymerization of 2-isopropenyl-4,5-dimethyloxazole with styrene. The smooth formation of (V) is also in sharp contrast to the lack of reactivity of (III) towards cationic initiators.

Notes: Summary A graphics display-oriented method for the computer-aided interpretation of the mass spectrum of a peptide in terms of its amino acid sequence is presented. The spectrum is obtained by collision induced decomposition of the protonated molecular ion (M+H)+ of the peptide generated by fast atom bombardment in the first double focussing mass analyzer of a tandem mass spectrometer and the product ion mass spectrum (CID spectrum) recorded by the second double focussing mass analyzer. The algorithm displays all series of peaks differing by the mass of -NH-CHR-CO- for the various amino acids. This display, which is very fast and can be augmented through interrogation by the user, greatly facilitates the determination of the amino acid sequence. The method is demonstrated on the spectra of a undecapeptide of (M+H)+ m/z = 1208.2 and a tetradecapeptide of (M+H)+ m/z = 1758.9.
