    Notes: Abstract: Addition of histamine (0.1 mM) to guinea-pig hippocampal slices causes a 20- to 30-fold increase in the accumulation of cyclic AMP compared with basal levels. This accumulation represents a balance between cyclic AMP production by adenylate cyclase and cyclic AMP breakdown mediated by phosphodiesterase (PDE). However, brain tissues are known to contain several different PDE isozymes. To determine which are involved in this response to histamine, the effect of isozyme-specific PDE inhibitors on cyclic AMP accumulation was examined in the hippocampus. MB 22948 (0.1 mM), an inhibitor of PDEs I and II, had no significant effect on the response to either 1 μM or 0.1 mM histamine. SKF 94120 (0.1 mM), a PDE III inhibitor, was also without effect in the presence of 1 μM histamine, although with 0.1 mM histamine, it caused a weak (1.25-fold compared with control), but statistically significant, enhancement of cyclic AMP accumulation. However, both rolipram (0.1 mM), a PDE IV inhibitor, and 3-isobutyl-l-methylxanthine (0.1 or 1 mM), an inhibitor of all forms of PDE, significantly increased cyclic AMP accumulation (2.8- to 6.5-fold compared with controls), and the relative size of this effect decreased with increasing histamine concentration. It is concluded that PDE IV is the main PDE isozyme involved in cyclic AMP turnover in guinea-pig hippocampal slices responding to histamine.

    Notes: Abstract The subcellular location of aspartate aminotransferase isozymes (EC 2.6.1.1) in the genusCapsella(Brassicaceae) was studied. The diploid speciesC. grandiflora andC. rubella have three AAT isozymes, including one located in the plastids. Each locus is duplicated in the tetraploidCapsella bursa-pastoris. Variation at the plastid-coding locus exceeded that at the other loci.C. bursa-pastoris had some unique alleles not detected in the diploid species. Segregation in open-pollinated families revealed thatCapsella grandiflora was outcrossing, whereasC. rubella was highly inbred, with most populations homozygous or uniform at all three loci. Inheritance in the tetraploid colonizerC. bursa-pastoris is disomic. This species was also predominantly selfing with outcrossing rates between 2% and 10%.

    Notes: Summary The origin of transfer (oriT) is the sequence within which conjugal transfer of plasmid DNA is initiated, and is absolutely required in cis for plasmid mobilization. We have cloned oriT from the 52 kb IncN plasmid R46 on a 600 bp fragment, and mapped the limits of the relevant sequence by deletion analysis and transposon mutagenesis. The nucleotide sequence of the oriT region contains 13 direct repeats of an 11 bp consensus sequence, 3 different pairs of 10 bp inverted repeats, and a segment that is extremely A-T rich. The direct repeats are within a region required for high frequency transfer and their sequence is such that their periodic alignment along the helix may induce curvature of the DNA. Analysis of Tn1725 insertions within the sequenced fragment of R46 revealed that, unlike most other transposons, transposition of Tn1725 can cause target sequence duplications of three different sizes.

    Notes: Summary The initial rate of radical copolymerization of vinyl acetate(D) and maleic anhydride(A) was measured dilatometrically in methyl ethyl ketone at 50°C. The rate was found to be maximum at the feed maleic anhydride mole fraction of 0.40,0.44 and 0.47 when the total monomer concentration was 0.75 M.1.50 M and 2.25 M, respectively. Participation of both free monomers and the complex was considered. Georgiev and Zubov's method gave α=kAD/kDA=0.24, βA=kAC/kAC=35,βD=kDC/kDA =2.2 and kAC/kDC=3.9. The equilibrium constant of donor-acceptor complexation between vinyl acetate and maleic anhydride was determined to be 0.030 in acetonitrile at 27°C.

    Notes: Summary A simple and reliable non-aqueous direct titration method for the determination of anhydride content in maleic anhydride-vinyl acetate copolymers is described. The procedure involves reaction of the anhydride with a large excess of aniline followed by direct titration of the resulting monoacid with ethanolic NaOH using potentiometric or thymol blue end point detection. The method overcomes many of the disadvantages of previously published methods, and its application to other copolymers is considered.

    Notes: Composition data for the copolymerization of styrene with maleic anhyride in ethyl methyl ketone at 50°C were obtained using a non-aqueous potentiometric direct titration method, and were analysed using a non-linear least squares optimization routine. Three variants of the penultimate model were fitted to the data: the four variable model (completely general), the three variable model (rmm = 0,0) and the more usual two variable model (rmm = rsm = 0,0). Of these, the two variable model was adequate to describe the data. Best estimates of the reactivity ratios for data previously reported in the literature were calculated, and in one case the best values were markedly different to those originally given.

The data were analysed using non-linear least squares minimization procedures to test the applicability of terminal, penultimate and complex models for both systems. In both systems, the penultimate model was an improvement over the terminal model, though the penultimate effect was more pronounced in the methyl methacrylate system. No evidence could be found for complex participation in either system.

    Notes: A nonaqueous potentiometric direct titration method was used to determine the composition diagrams for the copolymerization of maleic anhydride with styrene and with vinyl acetate in methyl ethyl ketone at 50°C. The data were analyzed using nonlinear least-squares minimization routines to fit composition equations for the terminal, penultimate, and complex models directly. The applicability of each model to both systems were evaluated statistically. The penultimate model was found to best describe both systems, although in the case of the maleic anhydride/vinyl acetate system this was only a small improvement over the terminal model. Although significant comonomer complexation occurs in both systems, the complex model did not provide statistically significant improvement in fit to the data compared with the terminal model.
