    Notes: Summary Using low-temperature postembedding techniques for immunoelectron microscopy, we succeeded in demonstrating the precise localization of bullous pemphigoid antigen (BP-Ag) in normal human skin. Small pieces (〈1 mm3) of normal adult skin were rapidly frozen in liquid propane at-190°C and subjected to freeze substitution with 100% methanol at-80°C. Specimens were embedded in Lowicryl K11M at-60°C which was polymerized under ultraviolet radiation at-60°C. Ultrathin sections were incubated with BP sera followed by rabbit anti-human IgG and colloidal-gold conjugated anti-rabbit IgG. Epidermal ultrastructure was generally well preserved: the basal cell plasma membrane and intra- and extracellular components of hemidesmosomes could be resolved. Gold particles were mainly distributed on and around the hemidesmosomes in both intra- and extracellular sites, with most of the labelling being inside the basal keratinocytes and within about 300 nm of the basal plasma membrane. No specific labelling was observed beneath melanocytes or when normal human serum was used as a control instead of BP serum. Our observations indicate that BP-Ag is localized in and around hemidesmosomes in normal human skin and that the antigen has both intracellular and extracellular domains with the major component occurring inside the cells.

    Notes: Summary The spatial and temporal pattern of oscillating temperatures on the cell surface of a plasmodial strand ofPhysarum polycephalum was measured with a sensitive thermal image camera. The longitudinal tension of the strand was studied simultaneously. In the absence of chemical stimulation, the phases of the temperature oscillation observed at various portions of the strand were entrained with almost coincidental phase. The temperature and tension oscillation were synchronized, although the phase difference between them was occasionally changed. With local chemical stimulation, the phase of the temperature oscillation advanced in the portion to which the plasmodium would be induced to migrate. The phases between temperature and tension oscillations then became constant. The mechanism by which the plasmodium processes local information of chemical stimulus to global information for the migration is discussed.

    Notes: The possibility of quantitative AES analysis using the whole EN(E) spectrum was examined for Au—Cu alloy samples. The EN(E) spectrum was measured under ϕ = 0° (normal) incidence of 10 keV electrons for the Au-44 at% Cu sample, and under ϕ = 0°, 30°, and 45° incidence of 10 keV primary electrons for the Au-20 at% Cu sample. Those spectra were compared with spectra which were obtained by linear combination of pure Au and Cu spectra measured under the same experimental conditions.Both measured and synthesized spectra coincided very well in the energy region lower than 1000 eV, except the region of slow secondary electrons. The results indicate that energy distribution of backscattered electrons of the Au—Cu alloy can be estimated well by linear combinations of pure sample spectra. They also indicate that, by comparison of the shape of the measured EN(E) spectrum with the synthesized one, the surface composition of the Au—Cu alloy sample can be estimated with reasonable accuracy (several % error).
