    Notizen: In a plasma containing more than one ion species, the frequencies of the ion–ion hybrid resonances can be a sensitive measure of the densities of the minor species, but only if the frequency shifts caused by electron motions are small, as experimental observations suggest. By calculating the eigenfrequencies for ion–ion hybrid waves taking into account collisions, Landau damping, electron inertia, electromagnetic effects, and density gradients, it is found that the frequency shifts are not negligible unless k(parallel) is extremely small. Treatment of the sheath boundary conditions at the ends of a plasma column shows that the effective value of k(parallel) can indeed be sufficiently small, and that the uncorrected resonance frequency is surprisingly accurate over a wide parameter range. Data from an Ar–Xe discharge demonstrate the usefulness of this technique for measuring impurity concentrations.

    Notizen: Summary Glomerular filtration rate, kidney volume, and urinary albumin excretion rate were studied in otherwise healthy newly diagnosed Type 2 (non-insulin-dependent) diabetic patients, untreated at diagnosis, after short-term treatment and after 3 months treatment. In 10 patients (Group A) glomerular filtration rate (measured by the plasma clearance of 51-Cr-EDTA) decreased from the time of diagnosis 106.2±14.6 ml·min−1·1.73m2−1 (mean±SD) to 95.9±13.7 ml·min−1·1.73m2−1 after 3 months treatment (p=0.049). At the same time, mean plasma glucose was reduced from 13.3±3.2 mmol/l to 6.5±1.1 mmol/l. The fall in mean plasma glucose was correlated to the reduction in glomerular filtration rate, r=0.76, p=0.011. Kidney volume as measured by ultrasonic scanning was reduced from 264.0±33.7 ml/l.73m2 to 210.8±23.8 ml/l.73 m2 (p〈0.005). The relative decline in urinary albumin excretion rate was correlated to the fall in glomerular filtration rate, r=0.69, p=0.026. In 15 patients (Group B) 24-h urine collections were made during 9.5±3.2 days, urinary albumin excretion rate fell from the first to the last day in hospital from 14.0×/÷3.0 μg/min (geometric mean ×/÷ tolerance factor) to 7.0×/÷2.7 μg/min p=0.015. The relative decline was correlated to the change in mean plasma glucose, r=0.65, p=0.032. Thus, kidney function in Type 2 diabetic patients is influenced by metabolic control, although to a lesser extent than is seen in Type 1 (insulin-dependent) diabetic patients with comparable glycaemic control. Urinary albumin excretion is reduced by improvement in glycaemic control, to which it is significantly correlated. Long-term consequences of reduction in urinary albumin excretion on the development of diabetic nephropathy and on survival remains to be elucidated.

    Notizen: Summary A synthetic 24-mer oligodeoxynucleotide complementary to the region of proopiomelanocortin (POMC) mRNA that codes for the MSH core sequence (αMSH/ACTH[4-11]), ws synthesized and labelled in the 3′-end by use of terminal transferase. Probes tailed with either [3H]- or biotin-labelled nucleotides could be used for in situ hydridization studies. Biotinylated probes, hybridized to mouse and rat pituitary sections, were detected by avidinalkaline phosphatase or streptavidin-alkaline phosphatase procedures and development in 5-brome-4-chloro-3-indolyl phosphate (BCIP)-nitroblue tetrazolium (NBT). Proteinase K pretreatment of sections produced a drastic enhancement of the signal obtained, particularly in strongly fixed, paraffin-embedded material. The non-radioactive in situ hybridization technique compared favourably to radioactive in situ hybridization in terms of rapidity and precision of the localization. Controls involved deletion of the probe to prove that other components of the reaction sequence did not yield stain, digestion with RNase to prove that tissue RNA was necessary to bind the probe, prehybridazation (blocking) with unlabelled probe to prove that the biotinylated probe reacted with its anti-sense region and not its tail and Northern blotting to show that the probe reacted with only one species of pituitary RNA, having the size of mouse pituitary POMC mRNA. In addition, adrenalectomy, known to increase anterior lobe POMC, levels, resulted in both increased numbers and increased intensity of positive corticotroph-like cells. Synthetic oligodeoxynucleotides labelled with biotin appear to constitute attractive reagents for in situ hybridization studies when supported by appropriate control procedures.

    Notizen: Abstract Suriclone is a new anxiolytic drug belonging to the family of cyclopyrrolones. Although chemically entirely different from the benzodiazepines, it acts as a benzodiazepine agonist with very high affinity for the benzodiazepine receptors. In the present cross-over study, 33 out-patients with a diagnosis of neurotic anxiety were treated with suriclone (mean dose 2 mg/day) and diazepam (25 mg/day) in two 6-week periods. Both drugs had a significant anxiolytic effect, but diazepam appeared to have a better effect within the first 2 weeks of treatment, while no significant difference was seen after treatment for 6 weeks. Suriclone and diazepam had a different side effect profile: suriclone produced mainly dizziness, while diazepam caused sedation. This may reflect the fact that suriclone and benzodiazepines bind to distinct sites or different allosteric conformations of the benzodiazepine receptors.
