    Notes: Summary 3H-thymidine incorporation into mitochondrial DNA of the liver and the kidney cells of chick embryos and newborn mice in tissue culture was shown by means of electron microscope radioautography with accurate localization. In these cells, about 20% of all the mitochondria were labeled at their matrices between the cristae within 4 hours in contact with the radioisotope, which were removed by DN'ase. From the results, it is clear that the mitochondria of avian and mammalian cells in tissue culture synthesize DNA.

    Notes: Summary A method for electron microscopic demonstration of lipase has heen developed by means of lead precipitation technique, modifying the original technique for light microscopy by Gomori. Care was taken to reduce the concentration of lead nitrate solution for substitution in order to prevent both the non-specific lead deposition and the diffusion. Applying the method to the pancreatic acinar cells of mice, specific activity was observed in rough surfaced endoplasmic reticulum, Golgi apparatus, zymogen granules and secretory canaliculi.

    Notes: Summary In order to clarify the underlying mechanism of malignant transformation from cirrhosis to hepatoma the cell kinetics of hepatocytes were studied in these two conditions. The content and synthesis of DNA in hepatocyte nuclei were investigated, by means of Feulgen-microspectrophotometry and tritiated thymidine radioautography, in cirrhotic and noncancerous parts of hepatoma with concomitant cirrhosis. The distribution of ploidy patterns was widely spread, from hypodiploid to hyperpolyploid, in the noncancerous parts of a cirrhotic liver containing hepatoma. In normal liver, each paired nuclear DNA content of a binucleate cell recorded almost the same amount, whereas in the noncancerous as well as in hepatoma cells much difference of DNA content was observed between the paired nuclei of the binucleate cells. The ploidy pattern of hepatocytes in patients with liver cirrhosis, who had developed hepatoma during follow-up periods of several months to several years, appeared to resemble that in noncancerous parts of hepatoma cases. On the other hand, the incorporation of tritiated thymidine into hepatocytes was found to be markedly increased in noncancerous parts as well as in cirrhotic liver developing hepatoma during follow-up periods. These results suggest the possibility that the hepatocytes in noncancerous parts of hepatoma have deranged cell-kinetics which might be a driving factor for the development of malignancy.

    Notes: Summary A method has been developed for electron microscopic histochemical demonstration of phospholipase B, (lecithinase B, E C 3.1.1.5, lysolecithin acyl hydrolase), which hydrolyzes α- and β-positions of phospholipids in mouse liver, kidney and adrenal tissues. Tissues either fixed in cold 1% paraformaldehyde or unfixed were cut into 40 μm frozen sections and were incubated at 37° C in a medium at pH 6.6 or 4.5 containing 2 μM lysolecithin and 0.25 mM CaCl2 for 20 min. The fatty acids liberated by enzymatic hydrolysis were trapped as calcium precipitate and were converted to lead precipitate by treatment with lead nitrate. The reaction products were observed by electron microscopy to be localized on the end of the smooth endoplasmic reticulum at pH 6.6 and in lysosomes and lipid droplets at pH 4.5. It is concluded that the products showed the localization of phospholipase B activity.

    Notes: Summary Localization of instilled 14C-bupranolol was studied in the rabbit eye by means of light microscopic radioautography. After the administration of 50 μl of 14C-bupranolol into the eyes of two rabbits, both eyes were enucleated at intervals of 15 and 30 min respectively, and radioautographic procedures were carried out on cryostat sections by means of both drymounting and wet-mounting methods. Although with the former it was possible to demonstrate many radioautographic silver grains in the specimens, with the latter it was not possible to detect their localization. On the dry-mounting procedure, radioautographic grains were observed mainly in the epithelium of the ciliary body 15 and 30 min after the administration of 14C-bupranolol. The numbers of grains were counted on the 15 and 30 min specimens. They were decreased in the conjunctiva, and cornea 30 min after the administration. On the contrary, they were remarkably increased in the epithelium of the ciliary body. From these results it is concluded that the 14C-bupranolol first penetrates into the epithelium of the conjunctiva and cornea, then moves rapidly to the ciliary body, and that only very small amount of 14C-bupranolol is changed into the insoluble form. Also, these results suggest that bupranolol plays a role in the control of aqueous production in the ciliary processes.
