    Notes: The variable-temperature proton nmr spectra of the oligoribonucleotides in the series CpApX and the series ApGpX, X = A, G, C, U, together with the parent dimers CpA and ApG have been measured. A complete analysis of all the nonexchangeable base proton resonances and ribose H-1′ proton resonances was made. The presence of trends in the shielding abilities of the various bases at both the nearest-neighbor and next-nearest-neighbor positions were identified. The observed shieldings could be used to predict the chemical shifts of protons in related systems. Based on the empirical results from ribodinucleoside monophosphates, the temperature-dependent behavior of the J1′2′ coupling constants of the triribonucleotides suggested that the compounds in the CpApX series stacked from the 5′-end to the 3′-end, while those in the ApGpX series stacked from the 3′-end to the 5′-end.

    Notes: A Monte Carlo analysis is presented which establishes a relationship between the rotational diffusion coefficients and the flexibility (persistence length, P) of short, wormlike chains. The results of this analysis are presented in terms of experimentally observable quantities; namely, the rotational relaxation times for the field-free decay of optical anisotropy. The pertinent theoretical quantity is R, defined as the ratio of the longest rotational relaxation time of a wormlike chain to the transverse rotational relaxation time of a rigid cylinder having the same axial length (L) and segmental volume. R, so defined, is essentially independent of the axial ratio of the cylinder for any value of L/P within the range of validity of the present analysis (axial ratio 〉 20; 0.1 〈 L/P 〈 5). It is pointed out that P can be determined with reasonable accuracy even in the absence of a precise knowledge of the local hydrodynamic radius of the chain.

    Notes: In the preceding article, a Monte Carlo analysis was presented which provides a quantitative numerical relationship between the rotational diffusion coefficients, as measured by the decay of optical anisotropy following an electric field pulse, and the flexibility (persistence length) of short, wormlike chains. In the present article, the results of the foregoing analysis are applied to the observed rates of decay of birefringence for a series of sequenced DNA fragments ranging in size from 104 to 910 base pairs. Under the conditions used in this study, the DNA fragments exist as native, duplex molecules. Furthermore, conditions are defined in which the observed relaxation times are not dependent on DNA concentration, field strength, or the duration of the pulse. It is pointed out that the ionic atmosphere associated with a wormlike polyion does not exert any significant (direct) influence on the rotational diffusion of the polyion and, therefore, that the rotational relaxation times are a true measure of the configurations of the DNA molecules in solution. Moreover, excluded-volume effects are shown not to be significant for the moderately short molecules employed in this study. The major conclusion of this study is that there is no strong ionic strength dependence of the persistence length for ionic strengths above 1 mM and that the persistence length, under conditions where electrostatic contributions are negligible, is approximately 500 Å. For ionic strengths significantly lower than 1 mM, electrostatic contributions to the stiffness of DNA become significant.

    Notes: Two new sesquiterpenoids, nakafuran-8 (1) and -9 (8), have been isolated from the marine sponge Dysidea fragilis and from its prey, the nudibranchs Hypselodoris godeffroyana and Chromodoris maridadilus. The structures were established by spectral analysis and chemical transformations. Both compounds possess antifeedant properties when assayed in the laboratory against common reef fishes, Chaetodon spp.

    Notes: A computerized fermentation system was developed for use in a research environment. Major goals in the design and construction of the system were flexibility and versatility. Direct digital control is employed for all low-level and high-level control loops. A microminicomputer hierarchical configuration is used to implement this control structure. The microcomputer is also utilized to simplify the interface with the fermentor, both at the hardware and minicomputer software levels. This computerized fermentation system provides accurate data acquisition, excellent control, and flexibility in the fermentation operation.

    Notes: The proton nmr spectra of the oligoribonucleotides in the series CpXpG, ApXpG, CpApXpUpG, and ApGpXpC (X = A, G, C, and U), together with the reference compounds CpG, ApG, CpApUpG, and ApGpC, have been measured. A complete analysis of all the nonexchangeable base protons and the ribose H-1′ protons was made. The insertion of a nucleotide X into a oligoribonucleotide led to shift changes at both nearest-neighbor and next-nearest-neighbor positions, which were rationalized in terms of the shielding abilities of the various bases. The derived shielding trends in the ApGpXpC series of compounds were successfully used to predict the chemical shifts of resonances in the related ApGpXpCpU series.

    Notes: A method is described for determining from dynamic viscoelastic data the gel time of thermosetting resins. Gelation of nine systems including epoxies, acetylene-terminated sulfone, and silicones was correlated with the crossover of the dynamic storage G′ and loss G″ moduli measured during isothermal curing. The effect of measurement frequency on gel time is discussed and related to the standard gel test. A lower temperature limit for the valid aplication of this technique was found to be at T 〉 Tgg + 30°C, where Tgg is the glass transition of the system at the gel point.

    Notes: The outer and inner surfaces of six commercial endotracheal tube cuffs were characterized using scanning electron microscopy (SEM) and energy dispersive x-ray analysis. Lateral tracheal wall pressure (LWP) was measured at various steps of cuff inflation using a mock-trachea system. The surface characteristics of the cuffs were found to vary widely. The surface of the Lanz cuff was relatively smooth but showed a regular array of circular depressions 0.8 μm in diameter. Uniform spheres also 0.8 μm in diameter were distributed through the thickness of the membrane. The American Hi-Lo cuff surface was also relatively smooth but was covered widely with flakes of material which x-ray showed to be a clay-like substance containing aluminum, silica, and potassium. The Harlake cuff was covered widely with roughly spherical, 5-μm-diam particles, probably starch granules. The membrane itself was smooth even at 3,000X. The Foregger cuff had a rough surface and was covered with the same clay-like particles seen on the American cuff. The inner surface with irregular, uneven areas were surrounded by fissures. The Rusch Armored tube cuff was completely covered by a continuous 2-μm-thick chlorinated coating. The surface was highly convoluted and irregular. The three cuff membranes showing relatively smooth surfaces, Lanz, American and Harlake, also produced relatively low LWP at various points of cuff inflation. While we have no data to indicate that cuff surface smoothness correlates with tracheal morbidity, it would seem prudent to select cuffs that produce low LWP's with smooth surfaces for clinical use.
